Wild tubers of Dioscorea bulbifera (Db) and Dioscorea hirtiflora (Dh) mainly used as sources of famine food and in herbal preparations are often indiscriminately collected in Africa and Asia. Therefore, there is the need to complement wild sourcing of the tubers to promote their conservation. The present study reports in vitro tuberous induction (80%) for the first time from Dh cultured on MS?+?NAA (2.5 mg/L) with IC50 of 472.5?±?1.77 µg/mL using DPPH, whereas tuberous root (60%) from Db on MS?+?Kn (2.5 mg/L)?+?NAA (0.25 mg/L) had IC50 of 26.97?±?1.00 µg/mL. Genetic fidelity assessment of in vitro plants compared to the wild plants revealed similar amplicon size of amplified DNA using trnH–psbA and rbcL. Similarly, micromorphological diagnostic features like oil gland, crystals (raphides), trichome and stomata type were obtained from the epidermal peels of the wild and in vitro plants. The ethyl acetate (EtOAc) extract of the flesh of Dh (wild) had the highest catechin content (108.3?±?0.69 µg/g DW). Protocatechuic acid was highest in the methanol (MeOH) extract of the flesh of Dh (0.42?±?0.02 µg/g DW), while it was detected in trace amount in the in vitro tuberous roots of MeOH extracts of Dh treated with NAA. The in vitro protocol developed in this study could be employed to multiply Dioscorea bulbifera L. and Dioscorea hirtiflora Benth. to offer genetically stable clones for the optimization of bioactive compounds and germplasms conservation.  相似文献

Veronica orientalis and related species form one of the taxonomically most challenging subgroups within the genus Veronica. Hybridization and polyploidization on the one hand and convergent character evolution on the other have made delimitation of species difficult. Amplified fragment length polymorphisms and plastid DNA markers were used in conjunction with 54 morphological characters to study relationships among 35 accessions of V. orientalis Mill. as well as other closely related species of Veronica occurring in Turkey and adjacent areas of Georgia and Armenia. In addition, ploidy levels were estimated for 15 accessions using flow cytometry. Diploid to hexaploid individuals were detected in V. orientalis. Analysis of DNA markers demonstrated the non-monophyly of V. orientalis, especially with regard to V. fuhsii and V. multifida from Eastern Turkey, but nuclear and plastid DNA markers are largely incongruent. Neither demonstrates a clear biogeographic pattern. The morphological analysis reveals the distinction of V. orientalis subsp. carduchorum, which is weakly retrieved in some molecular analyses and some clustering according to geography. V. multifida is not monophyletic likely due to parallel evolution of pinnatifid leaves east and west of the Anatolian diagonal.  相似文献

Amber from the Triassic (Carnian) of Lunz, a locality which has produced a rich and famous fossil flora, was analysed using UV‐B‐fluorescence, attenuated total reflectance Fourier transform infrared spectroscopy (ATR‐FTIR) and pyrolysis gas chromatography mass spectroscopy (Py‐GC‐MS). The amber is classified as a class Ib resinite based on its chemical composition, which is characterized by bicyclic products derived from a regular labdatriene structure and the absence of succinic acid. The presence of diterpenoids and absence of triterpenoids is clear evidence of a gymnosperm origin for the Lunz amber, and the prevalence of sesquiterpenoids and diterpenoids point to a conifer family as a possible botanical source. In search of amber attached to identifiable floral remains we have screened new and historical collections of the macroflora from a number of localities of the Lunz area using UV‐B, and describe a striking yellow UV‐B‐fluorescence of cuticles of pteridosperms, ginkgophytes and conifers. This varies with the diagenetic state of the sediment and may be lost altogether. Detectable amounts of fluorescent compounds were observed in gymnosperm taxa, but not in any ferns and only very weakly in horsetails. This underlines that fluorescent compounds are derived from gymnosperm plants, mostly likely arising secondarily from cyclic hydrocarbons by desaturation during diagenesis as a parallel process in amber as well as in cuticles.  相似文献

We have utilized Amplified Fragment Length Polymorphisms (AFLP) in conjunction with chloroplast DNA (cpDNA) sequence data to study the genetic diversity in 53 accessions of Dioscorea dumetorum from six countries in West and Central Africa. Our results provide a comparison of the two marker systems with regards to their applicability to differentiate intraspecific genotypes and the grouping of the accessions based on localities of collection. A total of 1052 AFLP fragments (of which 94.1% were polymorphic) produced from twelve primer combinations indicate a relatively high level of polymorphism among the accessions. Three major genetic groups that do not strictly follow a geographic distribution pattern were identified using Neighbour-joining and the principal coordinate (PCo) analyses. Accessions from Togo showed higher numbers of private fragments and the highest percentage polymorphism (59.4%). The detection of highest genetic diversity in accessions from Nigeria and Togo and their relationship to other accessions suggest that these countries are the centre of origin and diversity of D. dumetorum. The moderately high genetic diversity (average of 61%) is suggesting great influence on the D. dumetorum germplasms through exchange and transfer of cultivars among local farmers in the sub-region. In contrast, DNA sequence data from the psbA-trnH and the rpoB-trnC chloroplast regions revealed no variation among accessions from the different localities and clearly differentiated by AFLP patterns. The results demonstrate the usefulness of the AFLP marker in generating high polymorphism in the D. dumetorum accessions from West and Central Africa and hence may be used for agronomic purposes.  相似文献

Thirty-six higher plants sampled from Olomoro, Irri, Uzere, and Oginni exploration sites in the Niger Delta region of Nigeria were subjected to GC/MS analysis to assess the occurrence, distribution and profiles of polycyclic aromatic hydrocarbons (PAHs) contained in them. The Σ28PAHs ranged from 335 to 3094 ng/g. The results of the nonparametric regression models showed that PAHs concentration in a plant cannot be used in isolation to deduce the total PAHs concentration in soils hosting the plant since PAHs concentration in a plant is influenced by the presence (or absence) of other plants in that location. A combination of Factor analysis (FA) and principal component analysis (PCA) were used to recognize PAHs concentration patterns among the plants in the studied locations and individual PAHs compounds. Woody annuals and perennial plants formed similar patterns in Oginni and Irri locations. Three main clusters were formed by all the compounds with naphthalene and 2-methylnaphthalene standing as outliers in all the four locations.
