This paper investigates the kinetic parameters of Na+–K+-ATPase in glial, neuronal, and synaptosomal enriched fractions isolated from rabbit cerebral cortex. Under normal conditions, kinetic parameters-Vmax and K 0.5 K+ -of Na+–K+-ATPase are the same in the three fractions, suggesting that this enzyme behaves as the same molecular entity. Following a cryogenic lesion, the alterations of these parameters appear to be different in the different fractions. These data suggest that the same enzyme exhibits various responses when exposed to the same pathological event. The dissimilar lipid composition of the Na+–K+-ATPase environment, and/or different adaptative responses to abnormal ion concentrations in glial, neuronal, and synaptosomal fractions could account for these different responses.  相似文献

In order to detect serum antibodies against clinically important Old and New World hantaviruses simultaneously, multiparametric indirect immunofluorescence assays (IFAs) based on biochip mosaics were developed. Each of the mosaic substrates consisted of cells infected with one of the virus types Hantaan (HTNV), Puumala (PUUV), Seoul (SEOV), Saaremaa (SAAV), Dobrava (DOBV), Sin Nombre (SNV) or Andes (ANDV). For assay evaluation, serum IgG and IgM antibodies were analyzed using 184 laboratory-confirmed hantavirus-positive sera collected at six diagnostic centers from patients actively or previously infected with the following hantavirus serotypes: PUUV (Finland, n = 97); SEOV (China, n = 5); DOBV (Romania, n = 7); SNV (Canada, n = 23); ANDV (Argentina and Chile, n = 52). The control panel comprised 89 sera from healthy blood donors. According to the reference tests, all 184 patient samples were seropositive for hantavirus-specific IgG (n = 177; 96%) and/or IgM (n = 131; 72%), while all control samples were tested negative. In the multiparametric IFA applied in this study, 183 (99%) of the patient sera were IgG and 131 (71%) IgM positive (accordance with the reference tests: IgG, 96%; IgM, 93%). Overall IFA sensitivity for combined IgG and IgM analysis amounted to 100% for all serotypes, except for SNV (96%). Of the 89 control sera, 2 (2%) showed IgG reactivity against the HTNV substrate, but not against any other hantavirus. Due to the high cross-reactivity of hantaviral nucleocapsid proteins, endpoint titrations were conducted, allowing serotype determination in >90% of PUUV- and ANDV-infected patients. Thus, multiparametric IFA enables highly sensitive and specific serological diagnosis of hantavirus infections and can be used to differentiate PUUV and ANDV infection from infections with Murinae-borne hantaviruses (e.g. DOBV and SEOV).  相似文献

Bryozoan and foraminiferal assemblages from a core drilled in the Kusdili (Kadiköy, Istanbul) Upper Pleistocene-Holocene sequence were studied. Assemblages are absent from the very basal and top parts but they are well represented all along the middle part of the sequence. All the species found presently live in the Mediterranean. In the area sedimentation seemingly started about 13?000 years ago in very shallow near-shore environments with the deposition of pebbly sediments rapidly evolving to mud. Near the base, the presence of erosional surface allows to recognize three successive sedimentary cycles. Bryozoan and foraminiferal assemblages found in the middle part of the sequence testify to a shallow water, estuarine or lagoonal paleo-environment, in which effects of low salinity waters, more obvious toward the top, could be related to the reestablishment of a connection between the Marmara Sea and the Black Sea through the Bosphorus, about 7000 years BP.  相似文献

Thyroid status is crucial in energy homeostasis, but despite extensive studies the actual mechanism by which it regulates mitochondrial respiration and ATP synthesis is still unclear. We studied oxidative phosphorylation in both intact liver cells and isolated mitochondria from in vivo models of severe not life threatening hyper- and hypothyroidism. Thyroid status correlated with cellular and mitochondrial oxygen consumption rates as well as with maximal mitochondrial ATP production. Addition of a protonophoric uncoupler, 2,4-dinitrophenol, to hepatocytes did not mimic the cellular energetic change linked to hyperthyroidism. Mitochondrial content of cytochrome oxidase, ATP synthase, phosphate and adenine nucleotide carriers were increased in hyperthyroidism and decreased in hypothyroidism as compared to controls. As a result of these complex changes, the maximal rate of ATP synthesis increased in hyperthyroidism despite a decrease in ATP/O ratio, while in hypothyroidism ATP/O ratio increased but did not compensate for the flux limitation of oxidative phosphorylation. We conclude that energy homeostasis depends on a compromise between rate and efficiency, which is mainly regulated by thyroid hormones.  相似文献

The difference in parameters of visually guided and memory-guided saccades was shown. Increase in the memory-guided saccade latency as compared to that of the visually guided saccades may indicate the deceleration of saccadic programming on the basis of information extraction from the memory. The comparison of parameters and topography of evoked components N1 and P1 of the evoked potential on the signal to make a memory- or visually guided saccade suggests that the early stage of the saccade programming associated with the space information processing is performed predominantly with top-down attention mechanism before the memory-guided saccade and bottom-up mechanism before the visually guided saccade. The findings show that the increase in the latency of the memory-guided saccades is connected with decision making at the central stage of the saccade programming. We proposed that wave N2, which develops in the middle of the latent period of the memory-guided saccades, is correlated with this process. Topography and spatial dynamics of components N1, P1 and N2 testify that the memory-guided saccade programming is controlled by the frontal mediothalamic system of selective attention and left-hemispheric brain mechanisms of motor attention.  相似文献

Dynamic study of 3D localization of the equivalent current dipole (ECD) sources of visual evoked potentials (EP) in the human brain was performed in 18 healthy subjects using a two-dipole model. Dipole tracing was performed for relatively early EP components (N1, P1, and N2) with 1-ms step. The analysis confirmed localization of these ECDs mainly in the right occipital cortex and revealed their successive shift over this area in the anterior-medial direction and then backwards in all subjects during generation of the EP components. Typically, some successive arch-like trajectories of the shift were revealed (75.8%); their duration was relatively standard (about 25 ms) and did not depend on the stimulus shape and EP phase. Between the 1st and the 2nd trajectories (110-120 ms after the stimulus onset) a jump in ECD coordinates in the medial direction was found in 85% of cases. Possible significance of the findings for the insight into dynamic topography of the visual feature processing in the human brain is discussed.  相似文献

We investigated the effects of the general anesthetic Etomidate on oxidative phosphorylation in isolated rat liver mitochondria. The study of each electron transfer site shows that there is an inhibition: mainly at complex I but also, to a lesser extent, at complex III. Moreover, with succinate as substrate, the increase in non-phosphorylating respiration is accompanied by a decrease in ΔΨ. However, this effect is not due to classical uncoupling of oxidative phosphorylation, since ADP addition at high Etomidate concentrations restores the transmembrane difference of electrical potential. Also, in the same range of Etomidate concentration, the ATP/O ratio is not significantly affected. In conclusion, the main effect of Etomidate is to decrease the oxidative phosphorylation rate without changing yield. The H+ leak which appears under non-phosphorylating conditions becomes negligible in physiological conditions.  相似文献

Dermacentor reticulatus ticks are recognized as the most important vectors of Babesia canis, the aetiological agent of canine babesiosis occurring throughout Europe. Vector competence of D. reticulatus for B. canis is well described and experimentally determined; however, by using molecular analysis it was proven so by one recent study in Russia. Herein, the additional molecular evidence of B. canis infection in D. reticulatus ticks collected in Slovakia is provided. Using PCR followed by sequencing of distinctive amplicons we determined the presence of Babesia canis canis in one of 100 tested adult ticks. Two zoonotic pathogens, Francisella tularensis and Coxiella burnetii, were previously isolated from D. reticulatus ticks in Slovakia. In our samples, we detected only the presence of F. tularensis.  相似文献

BACKGROUND: Helicobacter pylori infection is associated with gastric cancer. Study with the Big Blue mouse model has reported a mutagenic effect associated with the H. pylori infection, as a result in part of oxidative DNA damage. The present work investigates the consequences of a deficiency in the OGG1 DNA glycosylase, responsible for the excision of 8-oxo guanine, on the inflammatory and genotoxic host response to the infection. MATERIALS AND METHODS: Big Blue Ogg1-/- C57BL/6 mice were orally inoculated with H. pylori strain SS1 or vehicle only, and sacrificed after 1, 3, or 6 months. The serologic response, histologic lesions, mutant frequency, and spectra of mutations were assessed in the stomach and compared to what observed in the wild-type (Wt) context. RESULTS: Inflammatory lesions induced in the gastric mucosa of H. pylori-infected mice, corresponding to a moderate gastritis, were less severe in Ogg1-/- than in Wt Big Blue mice. Analysis of antimicrobial humoral immunity exhibited a lower IgG2a serum level (Th1 response) after 6 months of infection in Ogg1-/- than in the Wt mice. In these conditions, the H. pylori-SS1 infection in the Ogg1-/- mice did not induce a mutagenic effect at the gastric epithelial cells level, either after 3 or 6 months. CONCLUSIONS: The inactivation of the OGG1 DNA glycosylase in mouse leads to less severe inflammatory lesions and abolished the mutagenic effect at the gastric epithelial cells level, induced by the H. pylori infection. These data suggest for the OGG1deficiency a protective role against inflammation and genotoxicity associated to the H. pylori infection.  相似文献
