alpha-Zingiberene synthase (ZIS), a sesquiterpene synthase gene that was isolated from lemon basil (Ocimum basilicum L.), encodes an enzyme that catalyzes the formation of alpha-zingiberene, and other sesquiterpenes, from farnesyl diphosphate. Transgenic tomato fruits overexpressing ZIS under the control of the fruit ripening-specific tomato polygalacturonase promoter (PG) accumulated high levels of alpha-zingiberene (224-1000 ng g(-1) fresh weight) and other sesquiterpenes, such as alpha-bergamotene, 7-epi-sesquithujene, beta-bisabolene and beta-curcumene, whereas no sesquiterpenes were detected in non-transformed control fruits. The ZIS-transgenic fruits also produced monoterpenes, such as alpha-thujene, alpha-pinene, beta-phellandrene and gamma-terpinene (1-22 ng g(-1) fresh weight), which were either not detected or were found only in minute concentrations in control fruits. Recombinant ZIS overexpressed in Escherichia coli catalyzed the formation of these monoterpenes from geranyl diphosphate. As the ZIS protein apparently lacks a transit peptide, and is localized in the cytosol, the production of monoterpenes in the transgenic tomatoes suggests that a pool of geranyl diphosphate is available in the cytosol. The phenotype of the ZIS-transgenic tomatoes was the same as that for wild-type tomatoes, with regard to plant vigor and shape, but transgenic plants exhibited a small decrease in lycopene content. This study thus showed that the synthesis of both mono- and sesquiterpenes can be enhanced by the ectopic expression of a single transgene in tomato fruit, and it further demonstrated the interconnection between the pools of terpenoid precursors in the plastids and the cytosol. 相似文献

Specific binding ofNicotiana nuclear protein(s) to subterminal regions of theAc transposable element was detected using gel mobility shift assays. A sequence motif (GGTAAA) repeated in both terminal regions ofAc, was identified as the protein binding site. Mutation of two nucleotides in this motif was sufficient to abolish binding. Based on a series of competition assays, it is deduced that there is cooperative binding between two repeats, each similar to the GGTAAA motif. The binding protein is probably similar to a previously characterized maize protein which binds to a GGTAAA-containing motif located in the ends ofMutator. Moreover, we show that DNA fromDs1 competes for protein binding toAc termini, and we show, by sequence analysis, that GGTAAA binding sites are present in the terminal region ofTgm1, Tpn1, En/Spm, Tam3 andDs1-like elements. The ectendomycorrhizal fungus Terfezia boudieri is known to secrete auxin. While some of the effects of fungal auxin on the plant root system have been described, a comprehensive understanding is still lacking. A dual culture system to study pre mycorrhizal signal exchange revealed previously unrecognized root–fungus interaction mediated by the fungal auxin. The secreted fungal auxin induced negative taproot gravitropism, attenuated taproot growth rate, and inhibited initial host development. Auxin also induced expression of Arabidopsis carriers AUX1 and PIN1, both of which are involved in the gravitropic response. Exogenous application of auxin led to a root phenotype, which fully mimicked that induced by ectomycorrhizal fungi. Co-cultivation of Arabidopsis auxin receptor mutants tir1-1, tir1-1 afb2-3, tir1-1 afb1-3 afb2-3, and tir1-1 afb2-3 afb3-4 with Terfezia confirmed that auxin induces the observed root phenotype. The finding that auxin both induces taproot deviation from the gravity axis and coordinates growth rate is new. We propose a model in which the fungal auxin induces horizontal root development, as well as the coordination of growth rates between partners, along with the known auxin effect on lateral root induction that increases the availability of accessible sites for colonization at the soil plane of fungal spore abundance. Thus, the newly observed responses described here of the root to Terfezia contribute to a successful encounter between symbionts. 相似文献

Segments of distal and proximal parts of the cotyledons, hypocotyls and epicotyls of yellow pitaya [Selenicereus megalanthus (Cactaceae)], were cultured on Murashige and Skoog (MS) medium supplemented with thidiazuron (TDZ). The proximal part of the cotyledons was found to be the most responsive tissue, maximal bud regeneration being achieved in the presence of 200 M thidiazuron. However, TDZ does not change the pattern of regeneration competence, which remains restricted to the proximal part of the cotyledons. The best medium for shoot elongation was found to be MS lacking plant growth regulators. Rooting was induced on MS medium supplemented with 5.3 M naphthaleneacetic acid. Rooted plantlets were transferred to a soil medium and acclimatized successfully in a greenhouse. 相似文献

BACKGROUND AND AIMS: Flowers of the vine cacti of the genera Hylocereus and Selenicereus grown in Israel must be hand pollinated due to self-incompatibility and lack of efficient pollinators. In controlled pollination experiments, it was found that the time elapsed between pollination and ripening depends on the source of the pollen. Therefore a study was made of some effects of the pollen source on fruit development. METHODS: Flowers of Hylocereus polyrhizus were pollinated on the same day with different pollen sources and the stigmas were covered. Fruits were collected 4 d after reaching full colour. RESULTS: Pollinating flowers of Hylocereus polyrhizus with Selenicereus grandiflorus and S. megalanthus pollen delayed ripening by 1 and 3 weeks, respectively, as compared with ripening of fruits pollinated with Hylocereus undatus pollen. Other fruit characteristics affected by the pollen source were fruit size, pulp dry weight and number of seeds per fruit, all being significantly reduced, while peel dry weight was significantly increased by S. megalanthus pollen. Total soluble sugar content was reduced in H. polyrhizus fruits pollinated with S. grandiflorus pollen. No other major traits were affected. CONCLUSIONS: The results are evidence for the existence of metaxenia, i.e. an effect of pollen on maternal tissues, in cacti fruits. This pollen effect on the fruit-ripening time may be used for extending the marketing period of H. polyrhizus fruits. 相似文献

Khat (Catha edulis Forsk.) is a flowering perennial shrub cultivated for its neurostimulant properties resulting mainly from the occurrence of (S)-cathinone in young leaves. The biosynthesis of (S)-cathinone and the related phenylpropylamino alkaloids (1S,2S)-cathine and (1R,2S)-norephedrine is not well characterized in plants. We prepared a cDNA library from young khat leaves and sequenced 4,896 random clones, generating an expressed sequence tag (EST) library of 3,293 unigenes. Putative functions were assigned to > 98% of the ESTs, providing a key resource for gene discovery. Candidates potentially involved at various stages of phenylpropylamino alkaloid biosynthesis from L-phenylalanine to (1S,2S)-cathine were identified. 相似文献
