The recently described antimicrobial peptide melectin (MEP, GFLSILKKVLPKVMAHMK-NH2) exhibits high antimicrobial activity against Gram-positive and Gram-negative bacteria. Here we describe the synthesis and biological activities of 23 new analogues of MEP. We studied the influence of dimerization and tetramerization (MAP-constructs of MEP) on the antimicrobial and hemolytic activities, as well as the role of Met in positions 14 and 17 of the peptide chain. Oxidation of the Met to Met(O) and Met(O2) decreases antimicrobial activity of all tested bacteria if the peptide is in the monomeric form, however, only to Staphylococcus aureus if in the form of dimer or tetramer. Dimerization and tetramerization increase the undesirable hemolytic activity of the peptides. Interestingly, substitution of Leu for Val in position 6 leads to the decrease of hemolytic activity. Introduction of the isosteric amino acid Nle into positions 14 or 17 or both leads to slight increase of hemolytic activity under preservation of high antimicrobial activities. Unfortunately, dimerization again leads to an increase of hemolytic activity.  相似文献

The aim of this study was to examine whether variables commonly used in aerobic exercise testing are influenced by menstrual cycle phases and use of oral contraceptive (OC) in female rowers. Twenty-four eumenorrheic female rowers distinguished on the basis of both menstrual status and athleticism participated in this study and were divided into competitive cyclic athletes (n = 8), recreationally trained cyclic athletes (n = 7), and recreationally trained athletes taking OC pills (ROC; n = 9). Rowers performed 2 incremental tests to voluntary exhaustion on a rowing ergometer during 2 different phases of the menstrual cycle: the follicular phase (FP) and the luteal phase (LP). The study variables were power output (Pa), heart rate (HR), oxygen consumption (VO2), carbon dioxide production (VCO2), minute ventilation (VE), the mean respiratory exchange ratio, and ventilatory equivalents of O2 (VE/VO2)) and CO2 (VE/VCO2), which were measured at maximal and at the aerobic-anaerobic transition intensities. In addition, maximal blood lactate (La) values after the test were obtained. When comparing Pa, &OV0312;o2, HR, and La values, no significant differences (p > 0.05) between FP and LP at maximal load and at threshold intensity were found in all 3 groups of the rowers studied. However, we observed higher values (p < 0.05) for VE/VCO2 at both intensities in LP compared with FP in the ROC group. In conclusion, sport-specific endurance performance was not influenced by the phase of the normal menstrual cycle and the synthetic menstrual cycle of the OC users in the rowers studied. Therefore, normally menstruating female rowers and female rowers taking OC pills should not be concerned about the timing of their menstrual cycle with regard to optimized sport-specific endurance performance.  相似文献

The ongoing COVID‐19 pandemic represents an unprecedented global health crisis. Here, we report the identification of a synthetic nanobody (sybody) pair, Sb#15 and Sb#68, that can bind simultaneously to the SARS‐CoV‐2 spike RBD and efficiently neutralize pseudotyped and live viruses by interfering with ACE2 interaction. Cryo‐EM confirms that Sb#15 and Sb#68 engage two spatially discrete epitopes, influencing rational design of bispecific and tri‐bispecific fusion constructs that exhibit up to 100‐ and 1,000‐fold increase in neutralization potency, respectively. Cryo‐EM of the sybody‐spike complex additionally reveals a novel up‐out RBD conformation. While resistant viruses emerge rapidly in the presence of single binders, no escape variants are observed in the presence of the bispecific sybody. The multivalent bispecific constructs further increase the neutralization potency against globally circulating SARS‐CoV‐2 variants of concern. Our study illustrates the power of multivalency and biparatopic nanobody fusions for the potential development of therapeutic strategies that mitigate the emergence of new SARS‐CoV‐2 escape mutants.  相似文献

We measured nitrous oxide (N2O), dinitrogen (N2), methane (CH4), and carbon dioxide (CO2) fluxes in horizontal and vertical flow constructed wetlands (CW) and in a riparian alder stand in southern Estonia using the closed chamber method in the period from October 2001 to November 2003. The replicates’ average values of N2O, N2, CH4 and CO2 fluxes from the riparian gray alder stand varied from −0.4 to 58 μg N2O-N m−2 h−1, 0.02–17.4 mg N2-N m−2 h−1, 0.1–265 μg CH4-C m−2 h−1 and 55–61 mg CO2-C m−2 h−1, respectively. In horizontal subsurface flow (HSSF) beds of CWs, the average N2 emission varied from 0.17 to 130 and from 0.33 to 119 mg N2-N m−2 h−1 in the vertical subsurface flow (VSSF) beds. The average N2O-N emission from the microsites above the inflow pipes of the HSSF CWs was 6.4–31 μg N2O-N m−2 h−1, whereas the outflow microsites emitted 2.4–8 μg N2O-N m−2 h−1. In VSSF beds, the same value was 35.6–44.7 μg N2O-N m−2 h−1. The average CH4 emission from the inflow and outflow microsites in the HSSF CWs differed significantly, ranging from 640 to 9715 and from 30 to 770 μg CH4-C m−2 h−1, respectively. The average CO2 emission was somewhat higher in VSSF beds (140–291 mg CO2-C m−2 h−1) and at the inflow microsites of HSSF beds (61–140 mg CO2-C m−2 h−1). The global warming potential (GWP) from N2O and CH4 was comparatively high in both types of CWs (4.8 ± 9.8 and 6.8 ± 16.2 t CO2 eq ha−1 a−1 in the HSSF CW 6.5 ± 13.0 and 5.3 ± 24.7 t CO2 eq ha−1 a−1 in the hybrid CW, respectively). The GWP of the riparian alder forest from both N2O and CH4 was relatively low (0.4 ± 1.0 and 0.1 ± 0.30 t CO2 eq ha−1 a−1, respectively), whereas the CO2-C flux was remarkable (3.5 ± 3.7 t ha−1 a−1). The global influence of CWs is not significant. Even if all global domestic wastewater were treated by wetlands, their share of the trace gas emission budget would be less than 1%.  相似文献

The aim of this study is to estimate emissions of greenhouse gases CO2, CH4 and N2O, and the effects of drainage and peat extraction on these processes, in Estonian transitional fens and ombrotrophic bogs. Closed-chamber-based sampling lasted from January to December 2009 in nine peatlands in Estonia, covering areas with different land-use practices: natural (four study sites), drained (six sites), abandoned peat mining (five sites) and active peat mining areas (five sites). Median values of soil CO2 efflux were 1,509, 1,921, 2,845 and 1,741 kg CO2-C ha?1 year?1 from natural, drained, abandoned and active mining areas, respectively. Emission of CH4-C (median values) was 85.2, 23.7, 0.07 and 0.12 kg ha?1 year?1, and N2O-N ?0.05, ?0.01, 0.18 and 0.19 kg ha?1 year?1, respectively. There were significantly higher emissions of CO2 and N2O from abandoned and active peat mining areas, whereas CH4 emissions were significantly higher in natural and drained areas. Significant Spearman rank correlation was found between soil temperature and CO2 flux at all sites, and CH4 flux with high water level at natural and drained areas. Significant increase in CH4 flux was detected for groundwater levels above 30 cm.  相似文献

Specificity (= the degree of ecological specialisation) is one of the fundamental concepts of the science of ecology. Ambiguities on how to define and measure specificity have however complicated respective research efforts. Here we propose that, in insects, a behavioural trait – adult oviposition latency in captivity without a favourable host plant – correlates with a species’ specificity in larval host use. In the absence of a suitable host, monophagous insects are expected to wait for a long time before commencing oviposition, with the long waiting time corresponding to careful host location behaviour in nature. Polyphagous insects, in contrast, should be selected for an increased oviposition rate at the expense of the quality of oviposition substrate encountered and will on average have a short latency time. Using experimentally derived data on oviposition latency, we performed a phylogenetically informed analysis based on Bayesian inference to show that this variable correlates with host specificity (larval diet breadth) in a sample of north European species of geometrid moths. A closely related index – the probability to lay any eggs on an unfavourable substrate – shows an analogous pattern. To provide an example of how these indices can be applied, we compare our sample of geometrid moths from northern Europe with a sample from equatorial Africa. A comparative analysis based on an original phylogenetic reconstruction found no differences between the two study sites in parameters of oviposition behaviour. We conclude that behavioural tests can provide information about ecological interactions when the latter can not be directly recorded. Our example study also hints at the possibility that host specificity of herbivores is not necessarily higher in a tropical region compared to a temperate one.  相似文献

1. Comparative studies on insect life histories are facilitated by the increasing availability of reliable phylogenies but are hampered by the scarcity of comparable data. Fortunately, morphological proxies of some life‐history traits can be measured on preserved specimens. 2. This study compared values of size‐related life‐history traits among a tropical (Ugandan) and a temperate (Estonian) assemblage of geometrid moths. 3. A comparative analysis based on an originally derived phylogeny revealed that tropical moths were, on average, larger than temperate ones. Tropical moths also had somewhat lower relative abdomen masses than temperate ones. This indicates that the tropical rather than the temperate moths tend to use an income (rather than capital) breeding strategy. Nevertheless, no difference was found in a related index of pro‐ovigeny. When body size was accounted for, tropical moths were found to lay smaller eggs than temperate ones. 4. The differences between the two compared areas are consistent with selection on higher mobility of the moths imposed by the more diverse tropical vegetation. Relatively larger eggs of temperate moths may constitute an adaptation to overcome the presumably stronger quantitative defences of their host plants. 5. Overall, however, we conclude that the differences in ecologically relevant size‐related traits are relatively low among moth assemblages of a tropical and a temperate forest region, indicating that these environments may not impose radically different selective pressures on insect life histories.  相似文献

The major facilitator superfamily transporter Rv1410 and the lipoprotein LprG (Rv1411) are encoded by a conserved two‐gene operon and contribute to virulence in Mycobacterium tuberculosis. Rv1410 was originally postulated to function as a drug efflux pump, but recent studies suggested that Rv1410 and LprG work in concert to insert triacylglycerides and lipoarabinomannans into the outer membrane. Here, we conducted microscopic analyses of Mycobacterium smegmatis lacking the operon and observed a cell separation defect, while surface rigidity measured by atomic force microscopy was found to be increased. Whereas Rv1410 expressed in Lactococcus lactis did not confer drug resistance, deletion of the operon in Mycobacterium abscessus and M. smegmatis resulted in increased susceptibility toward vancomycin, novobiocin and rifampicin. A homology model of Rv1410 revealed a periplasmic loop as well as a highly conserved aspartate, which were found to be essential for the operon’s function. Interestingly, influx of the fluorescent dyes BCECF‐AM and calcein‐AM in de‐energized M. smegmatis cells was faster in the deletion mutant. Our results unambiguously show that elevated drug susceptibility in the deletion mutant is caused by increased drug influx through a defective mycobacterial cell envelope and not by drug efflux mediated by Rv1410.  相似文献

The interactions of valinomycin, macrocyclic depsipeptide antibiotic ionophore, with ammonium cation NH4+ have been investigated. Using quantum mechanical density functional theory (DFT) calculations, the most probable structure of the valinomycin-NH4+ complex species was predicted. In this complex, the ammonium cation is bound partly by three strong hydrogen bonds to three ester carbonyl oxygen atoms of valinomycin and partly by somewhat weaker hydrogen bonds to the remaining three ester carbonyl groups of the valinomycin ligand. The strength of the valinomycin-NH4+ complex was evaluated experimentally by capillary affinity electrophoresis. From the dependence of valinomycin effective electrophoretic mobility on the ammonium ion concentration in the background electrolyte, the apparent binding (association, stability) constant (Kb) of the valinomycin-NH4+ complex in methanol was evaluated as log Kb = 1.52 +/- 0.22.  相似文献
