Compression wood (CW) contains higher quantities of β-1-4-galactan than does normal wood (NW). However, the physiological
roles and ultrastructural distribution of β-1-4-galactan during CW formation are still not well understood. The present work
investigated deposition of β-1-4-galactan in differentiating tracheids of Cryptomeria japonica during CW formation using an immunological probe (LM5) combined with immunomicroscopy. Our immunolabeling studies clearly
showed that differences in the distribution of β-1-4-galactan between NW (and opposite wood, OW) and CW are initiated during
the formation of the S1 layer. At this stage, CW was strongly labeled in the S1 layer, whereas no label was observed in the S1 layer of NW and OW. Immunogold labeling showed that β-1-4-galactan in the S1 layer of CW tracheids significantly decreased during the formation of the S2 layer. Most β-1-4-galactan labeling was present in the outer S2 region in mature CW tracheids, and was absent in the inner S2 layer that contained helical cavities in the cell wall. In addition, delignified CW tracheids showed significantly more labeling
of β-1-4-galactan in the secondary cell wall, suggesting that lignin is likely to mask β-1-4-galactan epitopes. The study
clearly showed that β-1-4-galactan in CW was mainly deposited in the outer portion of the secondary cell wall, indicating
that its distribution may be spatially consistent with lignin distribution in CW tracheids of Cryptomeria japonica. 相似文献
Observation was made on early ontogeny of vascular cambium in the developing root ofGinkgo biloba L. After completion of root elongation, the vascular meristem gradually acquires cambial characteristics. Strips of the periclinal
division of cells in transverse section are observed on the inner side of phloem when the primary xylem and phloem in the
stele have been established. The strips are united into a continuous layer between phloem and xylem. In tangenital section,
the procambium shows a homogeneous structure, which is initially composed of short cells with transverse end walls and subsequently,
of long cells with tapering ends. Then, the procambium is organized into two systems of cells; axial strands of short cells
with transverse end walls resulting from the sporadic transverse divisions of long cells, and long cells with tapering ends.
Still later, the short cells are divided frequently in a trasverse plane exhibiting one or a few cells in width and several
decades of cells in height, while the long cells are elongated. The frequency of transverse divisions of the short cells decreases
in subsequent stages. Eventually, the short cells in axial strands are vertically separated from one another by the elongation
of neighboring long cells and by the decrease in the frequency of transverse divisions of short cells themselves. Cambial
initials occur in two forms; ray initials a few cells in height and one cell in width derived from the short cells, and fusiform

The effect of high pressure sodium vapor lamps and daylight-simulating fluorescent lights on the growth, hematology, and behavior of Sprague-Dawley rats has been investigated. Rats weaned under the sodium vapor lamps had slightly heavier adrenals than those exposed to the daylight lamps when the two lighting systems were equalized either for total irradiance or for scotopic illuminance. However, no differences were observed in the tail-flick response, hot plate response, or swimming endurance of rats housed under the two lighting conditions. No consistent differences were seen in the hemoglobin, red and white cell count, hematocrit, and mean cell volume between rats weaned under the high pressure sodium vapor lamps and daylight-simulating fluorescent lights. 相似文献
A cDNA encoding a new phytocystatin isotype named BCPI-1 was isolated from a cDNA library of Chinese cabbage flower buds. The BCPI-1 clone encodes 199 amino acids resulting in a protein much larger than other known phytocystatins. BCPI-1 has an unusually long C-terminus. A BCPI-1 fusion protein expressed in Escherichia coli strongly inhibits the enzymatic activity of papain, a cysteine proteinase. Genomic Southern blot analysis revealed that the BCPI gene is a member of a small multi-gene family in Chinese cabbage. The vegetative morphology and life history of Halopteris filicina (Grateloup) Kutzing, collected from Korea, were examined in laboratory culture. Field plants attaining 3–5 cm in height were epilithic, tufted, yellowish-brown, and produced numerous erect axes with alternately distichous branches from compact basal discs. They were cultured under a 12:12 h LD photoperiod at 10°-C, 15°C and 20°C to observe the influence of temperature on reproduction. At 10°C plants grew only vegetatively, whereas at 15°C and 20°C they produced unilocular sporangia. Unispores released from sporangia developed into monoecious, anisogamous gametophytes that formed plurilocular female and male gametangia on the same lateral branches. The zygotes, by fusion of female macrogametes and male microgametes, developed into sporophytes bearing unilocular sporangia, whereas the unfused female gametes germinated parthenogenetically. This species was confirmed to have an isomorphic life history, basically similar to the other species of Sphacelariales. 相似文献
A meta-cleavage pathway for the aerobic degradation of aromatic hydrocarbons is catalyzed by extradiol dioxygenases via a two-step mechanism: catechol substrate binding and dioxygen incorporation. The binding of substrate triggers the release of water, thereby opening a coordination site for molecular oxygen. The crystal structures of AkbC, a type I extradiol dioxygenase, and the enzyme substrate (3-methylcatechol) complex revealed the substrate binding process of extradiol dioxygenase. AkbC is composed of an N-domain and an active C-domain, which contains iron coordinated by a 2-His-1-carboxylate facial triad motif. The C-domain includes a β-hairpin structure and a C-terminal tail. In substrate-bound AkbC, 3-methylcatechol interacts with the iron via a single hydroxyl group, which represents an intermediate stage in the substrate binding process. Structure-based mutagenesis revealed that the C-terminal tail and β-hairpin form part of the substrate binding pocket that is responsible for substrate specificity by blocking substrate entry. Once a substrate enters the active site, these structural elements also play a role in the correct positioning of the substrate. Based on the results presented here, a putative substrate binding mechanism is proposed. 相似文献
The human health risk of soils contaminated with As, Pb, Cu, and Zn was evaluated based on pseudo-total concentrations of metal(loid)s, the physiologically based extraction test (PBET), and diffusive gradients in thin films (DGT). Non-carcinogenic (NCR) and carcinogenic (CR) risks exceeded the U.S. Environmental Protection Agency criteria under both the residential and non-residential scenarios. Human bioavailable concentrations (PBET) were much lower than pseudo-total concentrations. The Hazardous Index of NCR (HI (NCR)) for the PBET in the studied soils was 67% and 94% less than that for pseudo-total concentration, respectively, under the non-residential and residential scenarios. Similarly, CR for the PBET was also 65% and 93% less for the two soils. The concentration of metal(loid)s accumulated in the DGT resin was highly correlated with the PBET-extractable concentration (R2 > 0.649). Therefore, for both the CR and HI (NCR), the DGT-calculated risk was linearly related to the PBET-calculated risk for the studied soils under both scenarios. The results suggest that DGT uptake and PBET-extracted concentrations are good surrogates for risk estimation and that both J1 and J2 soils require remediation before their use for residential or non-residential purposes. 相似文献
