Activation of T cells through the T cell antigen receptor (TCR) results in the rapid tyrosine phosphorylation of a number of cellular proteins, one of the earliest being a 100 kDa protein. We have sought to identify this 100 kDa substrate by partially purifying the protein by antiphosphotyrosine (APT) affinity purification, in order to obtain amino acid sequence data and, using this information, to isolate the cDNA clone encoding the molecule. We report here that the amino acid sequence data showed pp100 to be the murine equivalent of porcine valosin containing protein (VCP), a finding confirmed from the cloning and sequencing of the murine pp100 cDNA. Sequence analysis has shown VCP to be a member of a family of ATP binding, homo-oligomeric proteins, and the mammalian homolog of Saccharomyces cerevisiae cdc48p, a protein essential to the completion of mitosis in yeast. We also provide proof that both endogenous and expressed murine VCP are tyrosine phosphorylated in response to T cell activation. Thus we have identified a novel component of the TCR mediated tyrosine kinase activation pathway that may provide a link between TCR ligation and cell cycle control.  相似文献

Bacteroides nodosus is the primary causative agent of ovine foot rot. Virulent isolates of this bacterium contain fimbriae which appear to play a major role in both infectivity and protective immunity. This paper presents the cloning and expression in Escherichia coli of the gene encoding the structural subunit of the fimbriae of B. nodosus. Total DNA was isolated from B. nodosus VCS 1001 (serogroup A), digested with HindIII, and inserted into the positive-selection vector pTR262. Recombinant E. coli clones were screened directly with anti-fimbrial antiserum by using a colony immunoassay. Several positive colonies were identified, each of which contained the same 5.5-kilobase HindIII insert. The prototype has been designated pBA101. Some clones also contained additional flanking sequences from the B. nodosus genome. Western transfer analyses verified that the positive clones were producing the B. nodosus fimbrial structural subunit, molecular weight ca. 17,500. The level of expression of the antigen in E. coli was comparable to that in B. nodosus itself and was unaffected by the insertion site or orientation of the cloned fragment, indicating that synthesis was being directed from an internal promoter. Restriction mapping and deletion analyses localized the fimbrial subunit gene to the vicinity of a PvuII site near the central region of the original HindIII insert. The expressed antigen was located in the membrane-cell wall fraction and may be exposed on the surface of the recombinant E. coli cells.  相似文献

Summary and conclusions Darwin's theory of evolution brought to an end the static view of nature. It was no longer possible to think of species as immortal, with secure places in nature. Fluctuation of population could no longer be thought of as occurring within definite limits which had been set at the time of creation. Nor was it any longer possible to generalize from the differential reproductive potentials, or from a few cases of mutualism between species, that everything in nature was fitted to produce general ends, and reciprocal uses. 134 The appeal to design could no longer be substituted for answers to questions concerning animal demography. Instead, the dynamics of a population had to be viewed as the outcome of species' struggle against animate and inanimate factors in the environment. Both the members of a species and the environmental factors tend to vary randomly, and therefore neither evolution nor population dynamics could be fully understood alone. For this reason Darwin's linking of the two subjects was inevitable and not merely an historical accident. Since Darwin had shown that no automatic equilibrium existed, he demonstrated the importance of closer study of the causes of population dynamics and extinction. He also indicated that an understanding of population depends upon the development of a broad knowledge in ecology.Viewed from another direction, Darwin's work ended the early modern era of population studies by clarifying three interrelated problems which were important for understanding population: extinction, distribution, and the nature of species. The components of his answer had been discussed in the eighteenth century, but there had not existed enough evidence for the completion of the revolution in thought which had then begun. At the beginning of the nineteenth century, Playfair found the evidence for extinction conclusive, and, in spite of Lamarck, Curvier convinced the scientific world that there could no longer be any doubt about it. This was a step the importance of which, with his limited knowledge of biogeography and population, Cuvier could not have fully realized. Lamarck attempted, with his evolutionary theory, to circumvent the necessity for admitting extinction, but he overestimated the adaptability of organisms and in doing so he underestimated the importance of competition and the whole field of ecology. On the other hand, he was not willing to let questions such as the origin of species remain taboo to science. The origin of species was a biogeographical as well as a paleontological question. Humboldt correlated environment with the distribution of species and conveyed the impression that plant communities are subject to change. De Candolle, following the lead of Linnaeus and Humboldt, emphasized the ecological aspects of biogeography, not only the importance of habitat and range, clearly showing the ecological effects of competition. The entomologists Kirby and Spence took a faltering step toward understanding the relationship between population and ecological role, but they fell short of any significant new conclusions. Neither they nor Swainson could fully comprehend the new perspective of De Candolle.Lyell was able to bring together the evidence from these three lines of investigation and weave them into an important synthesis that almost accomplished that Darwin later did. Although opposing Lamarck's theory of evolution, Lyell had a dynamic view of ecology. He realized that population dynamics offered an important key to the understanding of biogeography. Since he knew that species become extinct, he investigated closely the factors which could either preserve or extinguish species. While explaining these factors, he described the interrelationships of species in greater detail than had ever been done before. Forbes continued to develop Lyell's ecological concepts, and his first-hand field experience enabled him to describe biotic communities more concretely than Lyell had.Having the advantages of Lyell's understanding and his own experience from a global voyage, Darwin could take the final step from the static to the dynamic concept of life. He had seen populations fluctuating and also fossil species in South America, and on the Galapagos Islands he had encountered a biogeographical problem that could not be credibly solved without the idea of evolution. However, the bare idea of evolution did not fully answer his questions. He sought physiological causes of extinction before he read Malthus and realized that De Candolle and Lyell had correctly emphasized the importance of competition. Darwin found that, in order to understand evolution, he needed to improve his understanding of ecology. He wanted to know when populations were most easily decimated, how extensive were competition and cooperation, what effects parasites have upon populations, and what changes occur in biotic communities when a species is either added or subtracted. He contributed to some extent to answering these questions. Though there remained much for others to do, there was now a new and more secure theoretical framework within which later studies could be interpreted. As Ernst Mayr has observed, Darwin's consistent thinking in terms of population has had an impact on biological theory and practice which is second only to his sponsorship of natural selection as the mechanism of evolution. 135  相似文献

Using FDC-P1 cells stably transfected with a murine erythropoietin receptor cDNA as a model, we recently have shown that erythropoietin (EPO), IL-3 and GM-CSF each induce the rapid phosphorylation of a common cytosolic target, i.e., a M(r) 100,000 phosphoprotein "pp100". Presently, we demonstrate that cytokine-induced phosphorylation of pp100 is primarily at tyrosine residues. This is shown by Western blotting with the anti-phosphotyrosine antibody PY20, and by the resistance of [32P]-pp100 to hydroxide-mediated hydrolysis of phosphates. These data, together with the recent observation by Linnekin et al. that pp100/p97 apparently associates directly with EPO receptors, suggest that pp100 may comprise an immediate common component in the signal transduction pathways of EPO, IL-3, GM-CSF and possibly other type I/II cytokine receptors. Additional analyses suggest that pp100 is distinct from a previously described M(r) 100,000 cytosolic target which is tyrosine phosphorylated in hematopoietic cells upon activation of T-cell receptors.  相似文献

The seasonal distribution of benthic species in the water column above and below the thermocline in a small eutrophic lake is described. During summer stratification populations of Spirostomum spp, Loxodes spp., Plagiopyla and Deltopylum become established in the plankton on or below the oxycline/thermocline. At shallow sites no migration occurred and populations of the migratory species in the benthos were sparse, with the exception of Plagiopyla which occurred in high densities in the sediment. Two distinct planktonic populations are established during stratification: an epilimnetic community of obligate planktonic ciliates and a hypolimnetic community of benthic migrants.  相似文献

Background aimsWe recently showed that co-transplantation of mesenchymal stromal cells (MSCs) improves islet function and revascularization in vivo. Pre-transplant islet culture is associated with the loss of islet cells. MSCs may enhance islet cell survival or function by direct cell contact mechanisms and soluble mediators. We investigated the capacity of MSCs to improve islet cell survival or β-cell function in vitro using direct and indirect contact islet-MSC configurations. We also investigated whether pre-culturing islets with MSCs improves islet transplantation outcome.MethodsThe effect of pre-culturing islets with MSCs on islet function in vitro was investigated by measuring glucose-stimulated insulin secretion. The endothelial cell density of fresh islets and islets cultured with or without MSCs was determined by immunohistochemistry. The efficacy of transplanted islets was tested in vivo using a syngeneic streptozotocin-diabetic minimal islet mass model. Graft function was investigated by monitoring blood glucose concentrations.ResultsIndirect islet-MSC co-culture configurations did not improve islet function in vitro. Pre-culturing islets using a direct contact MSC monolayer configuration improved glucose-stimulated insulin secretion in vitro, which correlated with superior islet graft function in vivo. MSC pre-culture had no effect on islet endothelial cell number in vitro or in vivo.ConclusionsPre-culturing islets with MSCs using a direct contact configuration maintains functional β-cell mass in vitro and the capacity of cultured islets to reverse hyperglycemia in diabetic mice.  相似文献

There is accumulating evidence of condition-dependent mate choice in many species, that is, individual preferences varying in strength according to the condition of the chooser. In humans, for example, people with more attractive faces/bodies, and who are higher in sociosexuality, exhibit stronger preferences for attractive traits in opposite-sex faces/bodies. However, previous studies have tended to use only relatively simple, isolated measures of rater attractiveness. Here we use 3D body scanning technology to examine associations between strength of rater preferences for attractive traits in opposite-sex bodies, and raters’ body shape, self-perceived attractiveness, and sociosexuality. For 118 raters and 80 stimuli models, we used a 3D scanner to extract body measurements associated with attractiveness (male waist-chest ratio [WCR], female waist-hip ratio [WHR], and volume-height index [VHI] in both sexes) and also measured rater self-perceived attractiveness and sociosexuality. As expected, WHR and VHI were important predictors of female body attractiveness, while WCR and VHI were important predictors of male body attractiveness. Results indicated that male rater sociosexuality scores were positively associated with strength of preference for attractive (low) VHI and attractive (low) WHR in female bodies. Moreover, male rater self-perceived attractiveness was positively associated with strength of preference for low VHI in female bodies. The only evidence of condition-dependent preferences in females was a positive association between attractive VHI in female raters and preferences for attractive (low) WCR in male bodies. No other significant associations were observed in either sex between aspects of rater body shape and strength of preferences for attractive opposite-sex body traits. These results suggest that among male raters, rater self-perceived attractiveness and sociosexuality are important predictors of preference strength for attractive opposite-sex body shapes, and that rater body traits –with the exception of VHI in female raters– may not be good predictors of these preferences in either sex.  相似文献
