Water-storage capacity was measured inThuja occidentalis L.,Tsuga canadensis (L.) Carr., andAcer saccharum Marsh. during the dehydration of stem segments 1.5–2.5 cm in diameter. Stem water potential was measured with a temperature-corrected
stem hygrometer and cavitations were detected acoustically. Water loss was measured by weight change. Dehydration isotherms
consistently displayed three phases. The first phase, from water potential (Ψ) 0 to about −0.2 MPa, had a high capacitance
(C>0.4kg water lost· (1 of tissue)−1· MPa−1) and we have attributed this high C to capillary water as defined by Zimmermann (1983, Xylem structure and the ascent of
sap, Springer-Verlag). The second phase from Ψ=−0.5 to about −2.0 had the lowest C values (<0.02 kg·l−1·MPa−1) and was accompanied by a few cavitation events. This phase may have been a transition zone between capillary storage and
water released by cavitation events as well as water drawn from living cells of the bark. The third phase also had a high
C (about 0.07–0.22kg·l−1·MPa−1) and was associated with many cavitation events while Ψ declined below about −2.5 MPa; we presume the high capacitance was
the consequence of water released by cavitation events. We discuss the ecological adaptive advantage of these three phases
of water-storage in trees. In moist environments, water withdrawn from capillary storage may be an important fraction of transpiration,
but may be of little adaptive advantage. For most of the growth season trees draw mainly on elastic storage, but stem elastic
storage is less than leaf elastic storage and therefore unlikely to be important. In very dry environments, water relased
by cavitation events might be important to the short-term survival of trees. 相似文献

Lectin from crude extract of small black kidney bean (Phaseolus vulgaris) was successfully extracted using the reversed micellar extraction (RME). The effects of water content of organic phase (Wo), ionic strength, pH, Aerosol-OT (AOT) concentration and extraction time on the forward extraction and the pH and ionic strength in the backward extraction were studied to optimize the extraction efficiency and purification factor. Forward extraction of lectin was found to be maximum after 15 min of contact using 50 mM AOT in organic phase with Wo 27 and 10 mM citrate-phosphate buffer at pH 5.5 containing 100 mM NaCl in the aqueous phase. Lectin was backward extracted into a fresh aqueous phase using sodium-phosphate buffer (10 mM, pH 7.0) containing 500 mM KCl. The overall yield of the process was 53.28% for protein recovery and 8.2-fold for purification factor. The efficiency of the process was confirmed by gel electrophoresis analysis. 相似文献

Floral scent is an important part of volatile compounds emitted from plants, and is influenced by many environmental factors. In this study, the floral scent emitted from Lilium ‘siberia’, a common breed of lily, was collected by dynamic headspace at different levels of light intensity (0, 100, 300, 600, 1,000, and 1,500 μmol m?2 s?1) and temperature (10, 20, 30, and 40 °C). Using the automated thermal desorption-gas chromatography/mass spectrometry (ATD-GC/MS) technique, the components and release amounts were subsequently identified to investigate the influence of light and temperature on the emission of floral scent. The results revealed that the numbers and release amounts of floral scent components were significantly influenced by light intensity and temperature, showing the similar pattern: first increasing and then decreasing. After light intensity treatment, the maximum numbers and release amounts mainly appeared at 600 and 1,000 μmol m?2 s?1. For temperature treatment, 30 °C resulted in the highest numbers and release amounts of the floral scent components. At different levels of light intensity and temperature, terpenoid compounds showed the highest numbers and release amounts among the component categories. α-Ocimene and linalool were the two terpenoid compounds with the highest release amounts, and accounted for the highest proportion. The results obtained provide evidence that both light intensity and temperature trigger the emission of floral scent. The particular response mechanisms must be investigated in future research. 相似文献

以30—90妇体重莱芜猪和40—100kg体重鲁莱黑猪共84头去势公猪为试验对象(每组6头),采用相对定量RT-PCR方法,以β-actin作为内标,研究肌肉中编码Ⅲ型胶原的Col3al基因表达的发育性变化及其对肌肉中胶原蛋白含量和性质(溶解度)的影响。结果表明:研究的两个品种猪肌肉中Col3al基因表达的发育性变化基本一致,即随体重的增加,肌肉中Col3al mRNA表达呈逐渐增加趋势,但莱芜猪和鲁莱黑猪分别在70妇和80妇体重组表达量略有下降。总体上莱芜猪肌肉组织Col3al mRNA表达丰度显著高于鲁莱黑猪(P〈0.05)。相关分析表明,莱芜猪肌肉组织Col3al mRNA表达的发育性变化与总胶原和不溶性胶原含量呈极显著正相关(P〈0.01),与胶原溶解度呈极显著负相关(P〈0.01)。鲁莱黑猪肌肉组织Col3al mRNA表达的发育性变化与不溶性胶原和胶原溶解度分别呈显著正相关和负相关妒〈0.05)。研究结果提示:猪肌肉组织中Col3al基因表达具有明显的体重发育和品种特征,其mRNA表达对于肌内胶原的含量和性质有重要影响。 相似文献

Coating cellulose tris (3,5‐dimethylphenylcarbamate) (CDMPC) on silica gels with large pores have been demonstrated as an efficient way for the preparation of chiral stationary phase (CSP) for high‐performance liquid chromatography (HPLC). During the process, a number of parameters, including the type of coating solvent, amount of coating, and the method for subsequent solvent removing, have been proved to affect the performance of the resultant CSPs. Coating times and the concentration of coating solution, however, also makes a difference to CSPs' performance by changing the arrangement of cellulose derivatives while remaining the coating amount constant, have much less been studied before, and thereby, were systematically investigated in this work. Results showed that CSPs with more coating times exhibited higher chiral recognition and column efficiency, suggesting that resolution was determined by column efficiency herein. Afterwards, we also investigated the effect of coating amount on the performance of CSPs, and it was shown that the ability of enantio‐recognition did not increase all the time as the coating amount; and four of seven racemates achieved best resolution when the coating amount reached to 18.37%. At the end, the reproducibility of CDMPC‐coated CSPs were further confirmed by two methods, ie, reprepared the CSP‐0.15‐3 and reevaluated the effect of coating times. 相似文献

Most Rho family GTPases serve as key molecular switches in a wide spectrum of biological processes. An increasing number of studies have expanded their roles to the spermatogenesis. Several members of Rho family have been confirmed to be essential for mammalian spermatogenesis, but the precise roles of this family in male reproduction have not been well studied yet. Here we report a surprising function of an atypical and testis-specific Rho GTPase, RSA-14-44 in spermatogenesis. Featured by unique structural and expressional patterns, RSA-14-44 is distinguished from three canonical members of Rho cluster. Thus, we define RSA-14-44 as a new member of Rho GTPases family and rename it RhoS (Rho in spermatogenic cells). RhoS associates with PSMB5, a catalytic subunit of the proteasome, in a series of stage-specific spermatogenic cells. More importantly, RhoS does not directly modulate the cellular proteasome activity, but participates in regulating the stability of "unincorporated" PSMB5 precursors. Meanwhile, our data demonstrate that the activation of RhoS is prerequisite for negatively regulating the stability of PSMB5 precursors. Therefore, our finding uncovers a direct and functional connection between the Rho GTPase family and the pathway of proteasome biogenesis and provide new clues for deciphering the secrets of spermatogenesis. 相似文献

Studies investigating the association between interleukin-13 (IL-13) single nucleotide polymorphism (SNP) rs1800925 and allergic rhinitis risk have reported conflicting results. The aim of the present study was to conduct a meta-analysis assessing the possible association of IL-13 SNP rs1800925 with allergic rhinitis risk. The relevant studies were identified through a search of PubMed, Embase, ISI Web of Knowledge and Chinese National Knowledge Infrastructure until December 2011 and selected on the basis of the established inclusion criteria for publications. Five studies were included in the present meta-analysis (1411 cases and 3169 controls). The combined results based on all studies showed that IL-13 SNP rs1800925 was not associated with increased allergic rhinitis risk (T versus C: odds ratio (OR)=1.06, 95% confidence interval (CI)=0.94-1.20; C/T versus C/C: OR=1.12, 95% CI=0.97-1.29; T/T versus C/C: OR=1.00, 95% CI=0.69-1.44; C/T+T/T versus CC: OR=1.10, 95% CI=0.96-1.27; T/T versus C/C+C/T: OR=0.91, 95% CI=0.64-1.31). This meta-analysis supported that IL-13 SNP rs1800925 was not associated with allergic rhinitis. 相似文献

Switchgrass (Panicum virgatum L.; family Poaceae) is a warm-season C4 perennial grass. Tillering plays an important role in determining the morphology of aboveground parts and the final biomass yield of switchgrass. Auxin distribution in plants can affect a variety of important growth and developmental processes, including the regulation of shoot and root branching, plant resistance and biological yield. Auxin transport and gradients in plants are mediated by influx and efflux carriers. PvPIN1, a switchgrass PIN1-like gene that is involved in regulating polar transport, is a putative auxin efflux carrier. Neighbor-joining analysis using sequences deposited in NCBI databases showed that the PvPIN1gene belongs to the PIN1 family and is evolutionarily closer to the Oryza sativa japonica group. Tiller emergence and development was significantly promoted in plants subjected toPvPIN1 RNA interference (RNAi), which yielded a phenotype similar to that of wild-type plants treated with the auxin transport inhibitor TIBA (2,3,5-triiodobenzoic acid). A transgenic approach that inducedPvPIN1 gene overexpression or suppression altered tiller number and the shoot/root ratio. These data suggest that PvPIN1plays an important role in auxin-dependent adventitious root emergence and tillering. 相似文献
