Cesium ions block potassium channels in biological membranes in a voltage dependent manner. For example, external cesium blocks inward current with little or no effect on outward current. Consequently, it produces a characteristic N-shaped current-voltage relationship. We have modeled this result by single file diffusion of ions in a narrow channel spanning the membrane with a special blocking site in the channel for cesium ions. The model enables us to make detailed comparisons of the effects of cesium on potassium channels in different types of biological membranes.  相似文献

A new proprietary de novo peptide design technique generated ten 15-residue peptides targeting and containing the leading nontransmembrane hydrophobic autocorrelation wavelengths, “modes”, of the human m1 muscarinic cholinergic receptor, m1AChR. These modes were also shared by the m4AChR subtype (but not the m2, m3, or m5 subtypes) and the three-finger snake toxins that pseudoirreversibly bind m1AChR. The linear decomposition of the hydrophobically transformed m1AChR amino acid sequence yielded ordered eigenvectors of orthogonal hydrophobic variational patterns. The weighted sum of two eigenvectors formed the peptide design template. Amino acids were iteratively assigned to template positions randomly, within hydrophobic groups. One peptide demonstrated significant functional indirect agonist activity, and five produced significant positive allosteric modulation of atropine-reversible, direct-agonist-induced cellular activation in stably m1AChR-transfected Chinese hamster ovary cells, reflected in integrated extracellular acidification responses. The peptide positive allosteric ligands produced left-shifts and peptide concentration-response augmentation in integrated extracellular acidification response asymptotic sigmoidal functions and concentration-response behavior in Hill number indices of positive cooperativity. Peptide mode specificity was suggested by negative crossover experiments with human m2ACh and D2 dopamine receptors. Morlet wavelet transformation of the leading eigenvector-derived, m1AChR eigenfunctions locates seven hydrophobic transmembrane segments and suggests possible extracellular loop locations for the peptide-receptor mode-matched, modulatory hydrophobic aggregation sites.  相似文献

A procedure for in vitro propagation of dioecious papaya clones is described. A high rate of success in culture estbalishment was obtained when axillary buds were taken from lateral shoots of hedged rooted cuttings grown in a greenhouse. Seasonal endophytic contamination was suppressed by shaking propagules for 24 h in 300 mgl-1 rifampicin or by incorporating it at 50 mgl-1 into the medium. Murashige & Skoog (MS) basal medium supplemented with 0.5 mgl-1 6-benzyladenine and 0.1 mgl-1 naphthaleneacetic acid was used for establishment and proliferation. The addition of 160 mgl-1 adenine sulfate improved multiplication and shoot growth. An elongation stage on MS medium supplemented with 1.0 mgl-1 kinetin and 0.05 mgl-1 naphthaleneacetic acid was necessary before rooting. Rooting was obtained at a high rate on half-strength macroelements of MS medium supplemented with 1.0 mgl-1 indole-3-butyric acid. Commercial plots of papaya plants obtained through this procedure already exist.  相似文献

Two Red Sea faviid species, Favia favus and Platygyra lamellina spawn eggs and sperm once a year, during the summer. External fertilization occurs 0.5 h after spawning, and mobile gastrulae appear 20 h later. Four stages in the early ontogenesis of these corals are described. The slow development (2–3 months) to the polyp stage in broadcasting species is attributed to the lack of zooxanthellae in their planulae and their appearance in the primary polyp only at a later stage. Survivorship of one-month-old primary polyps is ca 0.21% and 0.25% in F. favus and P. lamellina respectively, from the populations of 2–9-day-old planulae. Despite these low rates of survival, both species form dense populations in the Gulf of Eilat.  相似文献

Hydropathy plots are often used in place of missing physical data to model transmembrane proteins that are difficult to crystallize. The sequential maxima of their graphs approximate the number and locations of transmembrane segments, but potentially useful additional information about sequential hydrophobic variation is lost in this smoothing procedure. To explore a broader range of hydrophobic variations without loss of the transmembrane segment-relevant sequential maxima, we utilize a sequence of linear decompositions and transformations of the n-length hydrophobic free energy sequences, Hi, i = 1...n, of proteins. Constructions of hydrophobic free energy eigenfunctions, psil, from M-lagged, M x M autocovariance matrices, CM, were followed by their all-poles, maximum entropy power spectral, Somega(psil), and Mexican Hat wavelet, Wa,b(psil), transformations. These procedures yielded graphs indicative of inverse frequencies, omega-1, and sequence locations of hydrophobic modes suggestive of secondary and supersecondary protein structures. The graphs of these computations discriminated between Greek Key, Jelly Role, and Up and Down categories of antiparallel beta-barrel proteins. With these methods, examples of porins, connexins, hexose transporters, nuclear membrane proteins, and potassium but not sodium channels appear to belong to the Up and Down antiparallel beta-barrel variety.  相似文献

We have studied the effects of external cesium and rubidium on potassium conductance of voltage clamped squid axons over a broad range of concentrations of these ions relative to the external potassium concentration. Our primary novel finding concerning cesium is that relatively large concentrations of this ion are able to block a small, but statistically significant fraction of outward potassium current for potentials less than approximately 50 mV positive to reversal potential. This effect is relieved at more positive potentials. We have also found that external rubidium blocks outward current with a qualitatively similar voltage dependence. This effect is more readily apparent than the cesium blockade, occurring even for concentrations less than that of external potassium. Rubidium also has a blocking effect on inward current, which is relieved for potentials more than 20-40 mV negative to reversal, thereby allowing both potassium and rubidium ions to cross the membrane. We have described these results with a single-file diffusion model of ion permeation through potassium channels. The model analysis suggests that both rubidium and cesium ions exert their blocking effects at the innermost site of a two-site channel, and that rubidium competes with potassium ions for entry into the channel more effectively than does cesium under comparable conditions.  相似文献

The conjugates of the main determinant of penicillin, benzylpenicilloyl (BPO), with various protein carriers can stimulate mononuclear cells from human peripheral blood lymphocytes (PBL) to increase PGE2 secretion. Thus, BPO bound to either human γ-globulin (BPO-HGG), bovine γ-globulin (BPO-BGG), and keyhole limpet menocyanin (BPO-KLH) increased the PGE2 level 8–20 times above the level produced by stimulation with the carriers alone. Since in previous studies it was shown that preincubation with BPO-HGG can suppress the thymidine uptake after subsequent stimulation with BPO-KLH, we investigated if the induction of this unresponsiveness could be caused by PGE2. The results of this study show that: (a) although the amount of PGE2 secreted by the stimulation of BPO-HGG was 20-fold greater than the basic level, this amount is not sufficient to suppress significantly the thymidine uptake; (b) preincubation of PBL with higher than physiological concentration of externally added PGE2 caused significant suppression of thymidine uptake after stimulation with BPO-KLH, but the addition of physiological concentration of PGE2 did not show the same effect; (c) preincubation with BPO-HGG had no effect on PGE2 level after subsequent challenge with BPO-KLH; and (d) inhibitor of PGE fails to influence the suppression of BPO-KLH proliferation induced by BPO-KLH.  相似文献

The activation of potassium ion conductance in squid axons by voltage-clamp depolarization is delayed when the depolarizing step is preceded by a conditioning hyperpolarization of the axonal membrane. Moreover, the control conductance kinetics superpose with the delayed kinetics when they are translated along the time axis by an amount equal to the delay. We have found that the degree of superposition with internally perfused axons depends upon voltage-clamp protocol. The kinetics superpose almost exactly for modest test depolarizations, whereas they clearly fail to superpose completely for more positive levels of membrane depolarization. We have modeled these results by incorporating a time dependence into the rate constant of activation of potassium channel gates in the Hodgkin and Huxley model of potassium ionic conductance.  相似文献

Events of mass coral bleaching and mortality have increased in recent decades worldwide, making coral recruitment more important than ever in sustaining coral-reef ecosystems and ensuring their resilience. During the last four decades, the coral reefs of Eilat have undergone severe deterioration due to both anthropogenic and natural causes. Recruitment failure has been frequently suggested as one of the main mechanisms underlying this deterioration. Here we assess the demographic replenishment and resilience potential of the local reefs, i.e., the potential for new sexually derived corals to recruit and exceed the community’s mortality rate. We present a detailed analysis of coral community demography, obtained by means of high-resolution photographic monitoring of permanently marked plots. Coral spats as small as 1 mm were documented and the detailed dynamics of coral recruitment and mortality were recorded, in addition to other common ecological measurements. The cumulative quantity of recruited individuals was twofold to fivefold higher than total mortality. The most significant predictor variable for coral recruitment among all ecological parameters measured was the available substrate for settlement, and the survival of recruited corals was correlated with reef structural complexity. Two consecutive annual reproductive seasons (June–September of each year) with high recruitment rates were monitored. Combined with the high survival of recruits and the increase in coral live cover and abundance, the findings from this study indicate an encouraging potential for recovery of these reefs.
