Chlorophyll (Chl) a was measured every 10 m from 0 to 150 min the Transition Domain (TD), located between 37 and 45°N,and from 160°E to 160°W, in May and June (Leg 1) andin June and July (Leg 2), 1993–96. Total Chl a standingstocks integrated from 0 to 150 m were mostly within the rangeof 20 and 50 mg m–2. High standing stocks (>50 mg m–2)were generally observed westof 180°, with the exceptionof the sporadic high values at the easternmost station. Thetotal Chl a standing stock tended to be higher in the westernTD (160°E–172°30'E) than in the central (175°E–175°W)and eastern (170°W–160°W) TD on Leg 1, but thesame result was not observed on Leg 2. It was likely that largephytoplankton (2–10 and >10 µm fractions) contributedto the high total Chl a standing stock. We suggest that thehigh total Chl a standing stock on Leg 1, in late spring andearly summer, reflects the contribution of the spring bloomin the subarctic region of the northwestern Pacific Ocean. Thedistribution of total Chl a standing stock on Leg 2 was scarcelyaffected by the spring phytoplankton bloom, suggesting thattotal Chl a standing stock is basically nearly uniform in theTD in spring and summer. Moreover, year-to-year variation inthe total Chl a standing stock was observed in the western TDon Leg 1, suggesting that phytoplankton productivity and/orthe timing of the main period of the bloom exhibits interannualvariations.  相似文献

Two monoclonal antibodies (MAbs) reacting with spirosins from Enterobacteriaceae were obtained in a course of screening MAbs to spirosin from Yersinia enterocolitica SYT-11-72 (YE72). The antibodies were designated MAbs-S44 and S50. They were IgG2b and IgG2a, respectively, both with k light chains. On Western blotting after limited proteolysis of YE72 spirosin with Staphylococcus aureus V8 protease, they reacted markedly with peptide fragments of 27 and 35 kDa, suggesting the presence of an antigenic determinant on the fragments. When supernatant cell lysate from Escherichia coli K12 was chromatographed on DEAE-cellulose and Sepharose CL-6B columns successively, a 96-kDa protein with alcohol dehydrogenase (ADH) activity was always associated with reactivity to MAb-S50. These findings combined with N-terminal amino acid sequences clearly indicate the identity of spirosin to ADH in E. coli.  相似文献

Isolation of bacteria from wet skin lesions was attempted using Naruto Research Institute Otsuka Atrichia (NOA) mice, which develop such lesions spontaneously at a high rate. As a result, Staphylococcus aureus was demonstrated to have colonized the wet skin lesions at high density. In addition, the isolated S. aureus was found to be similar to the strain of S. aureus thought to colonize the eczematous lesions seen in humans with atopic dermatitis. Furthermore, a survey of the S. aureus colonization status of NOA mice with no wet skin lesions confirmed colonization at higher density than in HR-1 mice as control, indicating that the skin of the NOA mouse has the novel characteristic of increased susceptibility to S. aureus colonization. Thus, by using changes in S. aureus counts as an index, the NOA mouse can be expected to serve as a useful animal model for evaluating the effects of topical antiseptics. The antiseptic effects of an ointment and a lotion containing chlorhexidine gluconate were confirmed using this animal model.  相似文献

The effects of housing on the onset time and prevalence of wet skin lesions were investigated in NOA mice, which spontaneously develop these lesions at a high rate. Wet skin lesions developed earliest in mice that were housed individually. For mice that were housed in groups, the lesions developed earlier in mice with non-littermate group housing than in mice with littermate group housing. The prevalence of lesions was in the following order: individual housing > non-littermate group housing > littermate group housing. These results suggest that socio-psychological factors are involved in the etiology of wet skin lesions in the NOA mouse. Under individual housing conditions, two other novel characters of the NOA mouse were also observed, specifically, development of dry skin and wet skin lesions at the tail root. These characteristics developed early and with high prevalence and were easily observed on external examination. Therefore, these novel characteristics observed in NOA mice are potential markers of the psychological state of the animals.  相似文献

Protein disulphide isomerase (PDI) has been isolated as a binding protein of bisphenol A (BPA) in the rat brain. In this study, we determined binding sites of BPA to PDI and characterized the binding site. First, we identified the BPA-binding domain with ab, b'a'c, a, b, b' and a'c fragment peptides of PDI by surface plasmon resonance spectroscopy. BPA interacted with ab, b'a 'c, a and b', suggesting that a and b' domains are important in their interaction. Second, ab, b'a'c, a,b,b',a', abb'a', abb', b'a', Δb' and a'c fragment peptides were used for their isomerase activity with RNase as a substrate. BPA could inhibit the activity of peptide fragments including b', suggesting that b' domain contributes to inhibition of catalytic activity of PDI by BPA. Next, we investigated the BPA-binding capacity of PDI by amino acid substitution. PDI lost the BPA-binding activity by the mutation of H258 and mutation of Q245 and N300 also decreased its activity. Furthermore, acidic condition increased the BPA-binding activity of PDI. These results suggest that the charge of these amino acid especially, H258, is important for the BPA to bind to PDI.  相似文献

Size-fractionated chlorophyll a (Chla)-specific productivity (μgC μgChla −1 h−1) was measured at 11 stations off the northern coast of the South Shetland Islands during summer. The Chla-specific productivity of the 2- to 10 or 10- to 330-μm fraction was highest at 100% and 23% light depths. The Chla-specific productivity of the 2- to 10-μm fraction was generally highest, and that of the <2 or 10- to 330-μm fraction was sometimes highest at 12% and 1% light depths. Temperature was less than 3°C within the euphotic zone at all stations. The hypothesis of Shiomoto et al., according to which Chla-specific productivity of picophytoplankton (<2 μm) is not significantly higher than that of larger phytoplankton (>2 μm) in water colder than 10°C, was supported on condition that light is not limited for larger phytoplankton. In a previous study, we reported the development of grossly observable dry skin in all of the Naruto Research Institute Otsuka Atrichia (NOA) mice that were housed individually. In the present study, dermal physiological function tests were conducted and the usefulness of this dry skin model for evaluating the efficacy of topical moisturizers was assessed. As a result, we have confirmed a marked reduction in the water content of the stratum corneum in these animals. Therefore, the development of dry skin in the NOA mouse strain under individual housing conditions may be expected to serve as a useful animal model for evaluating topical moisturizers. Specifically, the water content of the stratum corneum was restored in proportion to the oil content of the ointment base used to treat the animals, and the moisturizing effects of urea were confirmed in animals treated with urea-containing ointment. In addition, when the animals that had been housed individually were returned to group housing conditions, the water content of the stratum corneum was restored, with a corresponding improvement in dry skin. This finding suggests that socio-psychological factors are involved in the etiology of dry skin in individually housed NOA mice.  相似文献

Biogenic silica (BSi), lithogenic silica (LSi), particiulate organic carbon (POC) and nitrogen (PON), and chlorophyll a (Chl a) concentration levels were measured in the surface waters (<100 m) off the northern coast of the South Shetland Islands in summer 1991. High concentration levels of BSi and LSi were recorded in the oceanic area and the coastal area, respectively. However, marked regional differences were not observed for POC, PON and Chl a concentrations. The mean BSi/POC atomic ratio (±SD) in the oceanic area (0.27±0.17) was 6 times that in the coastal area (0.045±0.020), except for the bloom situation (0.19±0.029). In contrast, the mean POC/PON atomic ratio was not significantly different in the coastal area (5.9±1.4) and the oceanic area (5.2±1.7). Nitzschia spp. were the dominant diatoms in the oceanic area but not in the coastal area. High BSi/POC ratios have been reported for blooms dominated by Nitzschia spp. even in the coastal regions of the Antarctic Ocean. The area difference in the BSi/POC ratios was probably related to the difference in species composition of phytoplankton and not to regional contrast. This species contributes significantly to high BSi/POC ratios in the Antarctic Ocean.  相似文献
