In spinal cord injury, the injury could trigger some inhibitory signal cascades to promote chondroitin sulfate proteoglycans (CSPGs), the structures of scar tissues, formation. CSPGs could limit axonal regeneration mainly through the glycosaminoglycan (GAG) chain in the lesion site were suggested. We hypothesized that the digestion of CSPGs by chondroitinase ABC (ChABC) might decrease the inhibitory effects of limiting axonal re-growth after spinal cord injury. We compared the digesting products of CSPGs such as 2B6 by ChABC with the untreated control group and found no immunostaining of 2B6 in control group. The smaller size scars of ChABC-treatment were observed via CS-56, a type of CSPGs, 8 weeks after transection by immunohistochemistry. The inhibitory effects of CSPGs withdraw GAGs following ChABC-treatment would reduce, and immunopositive GAP-43 newly outgrown fibers were identified. In the animal trials, ChABC-treatment could improve motor function through BBB locomotor's test and reduce limiting ability of scar tissues to promote axonal regeneration via changing the structure of CSPGs by immunohistochemistry with GAP-43.  相似文献

Oncogenic transformation of normal cells often involves epigenetic alterations, including histone modification and DNA methylation. We conducted whole-genome bisulfite sequencing to determine the DNA methylomes of normal breast, fibroadenoma, invasive ductal carcinomas and MCF7. The emergence, disappearance, expansion and contraction of kilobase-sized hypomethylated regions (HMRs) and the hypomethylation of the megabase-sized partially methylated domains (PMDs) are the major forms of methylation changes observed in breast tumor samples. Hierarchical clustering of HMR revealed tumor-specific hypermethylated clusters and differential methylated enhancers specific to normal or breast cancer cell lines. Joint analysis of gene expression and DNA methylation data of normal breast and breast cancer cells identified differentially methylated and expressed genes associated with breast and/or ovarian cancers in cancer-specific HMR clusters. Furthermore, aberrant patterns of X-chromosome inactivation (XCI) was found in breast cancer cell lines as well as breast tumor samples in the TCGA BRCA (breast invasive carcinoma) dataset. They were characterized with differentially hypermethylated XIST promoter, reduced expression of XIST, and over-expression of hypomethylated X-linked genes. High expressions of these genes were significantly associated with lower survival rates in breast cancer patients. Comprehensive analysis of the normal and breast tumor methylomes suggests selective targeting of DNA methylation changes during breast cancer progression. The weak causal relationship between DNA methylation and gene expression observed in this study is evident of more complex role of DNA methylation in the regulation of gene expression in human epigenetics that deserves further investigation.  相似文献

Species of Broussonetia have been essential in the development of papermaking technology. In Japan and Korea, a hybrid between B. monoica and B. papyrifera (= B. × kazinoki) known as kōzo and daknamu is still the major source of raw materials for making traditional paper washi and hanji, respectively. Despite their cultural and practical significance, however, the origin and taxonomy of kōzo and daknamu remain controversial. Additionally, the long-held generic concept of Broussonetia s.l., which included Sect. Allaeanthus and Sect. Broussonetia, was challenged as phylogenetic analyses showed Malaisia is sister to the latter section. To re-examine the taxonomic proposition that recognizes Allaeanthus, Broussonetia, and Malaisia (i.e., Broussonetia alliance), plastome and nuclear ribosomal DNA (nrDNA) sequences of six species of the alliance were assembled. Characterized by the canonical quadripartite structure, genome alignments and contents of the six plastomes (160,121–162,594 bp) are highly conserved, except for the pseudogenization and/or loss of the rpl22 gene. Relationships of the Broussonetia alliance are identical between plastome and nrDNA trees, supporting the maintenance of Malaisia and the resurrection of Allaeanthus. The phylogenomic relationships also indicate that the monoecy in B. monoica is a derived state, possibly resulting from hybridization between the dioecious B. kaempferi (♀) and B. papyrifera (♂). Based on the hypervariable ndhF-rpl32 intergenic spacer selected by sliding window analysis, phylogeographic analysis indicates that B. monoica is the sole maternal parent of B. × kazinoki and that daknamu carries multiple haplotypes, while only one haplotype was detected in kōzo. Because hybridizations between B. monoica and B. papyrifera are unidirectional and have occurred rarely in nature, our data suggest that daknamu might have originated via deliberate hybrid breeding selected for making hanji in Korea. On the contrary, kōzo appears to have a single origin and the possibility of a Korean origin cannot be ruled out.

Danshuei River in the northwestern tip of Taiwan dischargesinto the boundary coastal waters between the East China Seaand the Taiwan Strait. We conducted a 5-year study between October1998 and September 2003 to assess the influence of the northeast(NE) and the southwest (SW) monsoons on copepod assemblagesin the area. A total of 110 copepod species were identified.Temora turbinata, Paracalanus aculeatus, Acrocalanus gibber,Parvocalanus crassirostris and Oithona rigida comprised 80%of the copepod assemblages that consisted of coastal speciesfrom the East China Sea and species associated with the KuroshioBranch Current. The effect of the NE monsoon was observed duringa short period of each winter when species such as Calanus sinicuswere transported into the study area by the China Coastal Current.In summer, species such as A. gibber, Acrocalanus gracilis andCanthocalanus pauper may be transported into the study areafrom Southern Taiwan by the combined effect of the SW monsoonand the Kuroshio Current. Influence of the NE monsoon on thecopepod assemblages in terms of introduction of species fromthe Yellow Sea and the East China Sea during winter was observedevery year but was unlikely to represent an important carbonsource into the region due to its short-term nature. Local speciessuch as T. turbinata, P. crassirostris and O. rigida form amajor component of the copepod assemblage. The high copepoddiversity in the area was caused by the year-round presenceof many local species and the aggregation of different speciesfrom subtropical, tropical, and temperate water masses. Monsoon-drivenwater currents and the Kuroshio Branch Current appeared to maskthe effect of river discharge in the region.  相似文献

Kawasaki disease (KD) typically occurs in children aged under 5 years and can cause coronary artery lesions (CALs). Early diagnosis and treatment with intravenous immunoglobulin can reduce the occurrence of CALs; therefore, identifying a good biomarker for diagnosing KD is essential. Here, using next-generation sequencing in patients with recurrent KD, those with viral infection, and healthy controls, we identified dysregulated circulating microRNAs as diagnostic biomarkers for KD. Pathway enrichment analysis illustrated the putative role of these miRNAs in KD progression. Their expression levels were validated using real-time polymerase chain reaction (qPCR). Fifteen dysregulated circulating miRNAs (fold changes >2 and <0.5) were differentially expressed in the recurrent KD group compared with the viral infection and control groups. These miRNAs were significantly involved in the transforming growth factor-β, epithelial–mesenchymal transition, and cell apoptosis signaling pathways. Notably, their expression levels were frequently restored after intravenous immunoglobulin treatment. Among the candidates, miR-24-3p expression level was significantly higher in patients with recurrent KD compared with healthy controls or viral infection controls (p < 0.001). Receiver operating characteristic analysis revealed that high miR-24-3p expression levels may be a potential biomarker for KD diagnosis. In conclusion, we identified miR-24-3p significantly higher in KD patients, which may be a potential diagnostic biomarker for KD.  相似文献

A field survey was conducted to investigate the presence of Borrelia burgdorferi sensu lato (s.l.) in six counties of Taiwan. Spirochetes were successfully isolated from one rodent ear sample out of 485 rodent ears and 53 live, fed tick (Ixodes granulatus) samples. The spirochetes were confirmed to be B. burgdorferi s.l. by real-time PCR. In addition, 23 of 113 tick samples were tested positive for Borrelia DNA according to real-time PCR. The Borrelia isolate from the rodent and the 23 Borrelia DNA samples from the ticks were identified as B. valaisiana-related genospecies by phylogenetic analysis based on flagellin gene sequences. These findings suggest that the Borrelia valaisiana-related strains are maintained in a zoonotic cycle between tick vectors and reservoir hosts in Taiwan.  相似文献

Hydrology and trophic relationships are frequently reported for inducing changes in mesozooplankton communities. This study investigated the distribution and abundance of mesozooplankton in the boundary waters between the East China Sea and the Taiwan Strait. Samples were collected using a NOR-PAC zooplankton net towed horizontally at a depth of 2 m, at eight stations along a transect, in March (spring) and October (autumn) 2005. The abundance of mesozooplankton was significantly higher during autumn than spring. Densities of many groups (e.g., Noctiluca scintillans, pteropods, copepods, mysids, euphausiids, and other larva) increased in October. During both seasons, copepods represented more than 50% of the total zooplankton abundance. Noctiluca scintillans, appendicularians, and Calanus sinicus were dominant in spring, indicating their association with the cold waters of the East China Coastal Current. Appendicularians and N. scintillans were mainly associated with the coastal waters of Taiwan; whereas C. sinicus was concentrated in the offshore waters along the coast of Mainland China. Chaetognaths, Temora turbinata, Acrocalanus spp., and radiolarians were dominant in autumn, showing their association with the warm waters of the Kuroshio Branch Current. Oncaea venusta was relatively abundant during both seasons. Our study shows that, in addition to the influence of seasonal changes in the water masses, the distribution and composition of mesozooplankton are highly influenced by trophic interactions between zooplankton taxa, in the boundary waters of the Taiwan Strait and the East China Sea.  相似文献
