Using radioimmunoassay, the concentration of carbonic anhydrase III in the livers of adult male rats was found to be approx. 30-times greater than that observed in mature females. Castration of male rats led to a marked reduction in liver carbonic anhydrase III concentrations which could be partially restored to control levels by testosterone replacement. Administration of testosterone to ovariectomised female rats induced about a 5-fold increase in liver carbonic anhydrase III concentration. Immunoprecipitation analysis of the products of liver mRNA translation in vitro with antiserum specific for carbonic anhydrase III showed that hormonal control of the levels of carbonic anhydrase III in liver is mediated by changes in the amount of translatable carbonic anhydrase III mRNA. Marked changes in liver carbonic anhydrase III concentrations were also observed in developing and ageing male rats.  相似文献

A specific and sensitive radioimmunoassay for the rat carbonic anhydrase III isoenzyme was developed. High concentrations of carbonic anhydrase III were detected in soleus muscle and male liver. Female liver and other skeletal muscles contained significantly lower concentrations, and only trace amounts were found in heart, prostate, kidney, brain, plasma, urine and, possibly, erythrocytes.  相似文献

The southern Persian/Arabian Gulf experiences extreme seasonal temperature variation (> 20 °C) making it among the most hostile reef environments on Earth. Previous anecdotal evidence has suggested that seasonal temperature changes may influence regional reef fish assemblages, but to date research has been limited. To examine the influence of temperature on reef fish abundance and composition, we performed visual surveys in summer and in winter over three years at three reefs in the southern Gulf (Dhabiya, Saadiyat and Ras Ghanada). Overall abundance of fishes was 40% higher in summer than in winter, and multivariate analyses showed strong and significant differences in overall seasonal community structure, consistent at all sites and across all years. Seasonal differences were largely driven by nine of the 30 observed species, which together accounted for 70% of the divergence in community structure between summer and winter. Of these nine species, Lutjanus ehrenbergii, Lutjanus fulviflamma, Plectorhinchus sordidus and Abudefduf vaigiensis were significantly more abundant in summer, Parupeneus margaritatus and Acanthopagrus bifasciatus, were significantly more common on reefs in winter. We discuss these changes in terms of seasonal physiological and ecological constraints, and explore the implications of these changes on the functional ecology of reef fishes in this thermally variable and extreme environment.

Background

The effects of microchannel diameter in hydroxyapatite (HAp) substrates on osteoblast behavior were investigated in this study. Microchannels of 100, 250 and 500 μm diameter were created on hydroxyapatite disks. The changes in osteoblast precursor growth, differentiation, extra cellular matrix (ECM) secretion and cell attachment/orientation were investigated as a function of microchannel diameter.

Results

Curvature did not impact cellular differentiation, however organized cellular orientation was achieved within the 100 and 250 μm microchannels (mc) after 6 days compared to the 12 days it took for the 500mc group, while the flat substrate remained disorganized. Moreover, the 100, 250 and 500mc groups expressed a specific shift in orientation of 17.45°, 9.05°, and 22.86° respectively in 24 days. The secreted/mineralized ECM showed the 100 and 250mc groups to have higher modulus (E) and hardness (h) (E?=?42.6GPa; h?=?1.6GPa) than human bone (E?=?13.4-25.7GPa; h?=?0.47-0.74GPa), which was significantly greater than the 500mc and control groups (p?<?0.05). It was determined that substrate curvature affects the cell orientation, the time required for initial response, and the shift in orientation with time.

Conclusions

These findings demonstrate the ability of osteoblasts to organize and mineralize differentially in microchannels similar to those found in the osteons of compact bone. These investigations could lead to the development of osteon-like scaffolds to support the regeneration of organized bone.

Analysis of telomere length in Dolly, a sheep derived by nuclear transfer   总被引:3,自引:0,他引:3  
We have used a (TTAGGG) oligonucleotide probe to demonstrate that ovine telomeres are composed of (TTAGGG) repeat arrays and to compare the terminal restriction fragment lengths of sheep derived by natural mating and nuclear transfer. Here we show that ovine somatic telomeres decrease in length with age, and that Dolly, derived by the transfer of 6-year-old adult somatic nucleus, exhibits diminished terminal restriction fragment lengths. The decrease is consistent with the age of the donor tissue and telomere erosion during in vitro culture. Nuclear transfer does not restore telomere lengths. Dolly otherwise appears physiologically and phenotypically normal for her breed and age. We further report on apparent telomere lengthening in sheep, occurring during the first year in naturally derived lambs.  相似文献
