Restriction assay of mtDNA has been made in 6 salmon species form the genus Oncorhynchus and one species from the related genus, i.e. Salvelinus malma. The size of the mitochondrial genome was found to be identical and equal to 16.7 kilobases. The digestion patterns of mtDNA cleaved by 5 restriction endonucleases (Eco RI, Bgl I, Bgl II, Hind III, and Pst I) were used for analysis of the levels of interspecific variation and for estimation the matrix of mtDNA sequence differentiation. It was found that the level of nucleotide sequence divergence (p) in the genus Oncorhynchus varies within 1.7-6.7%. Minimum p value was observed in a pair O. keta--O. gorbuscha, maximum one--between O. masu and other species. With respect to similarity in their mtDNA, three groups may be distinguished: 1) O. gorbuscha--O. keta; 2) O. nerka--O. kisutch, O. tschawytscha; 3) O. masu. Mean value of intergeneric level of sequence divergence between Oncorhynchus and Salvelinus was found to be equal to 8%. On the basis of mtDNA analysis, the dendrogram of similarity of the species was plotted which is consistent in principle with current viewpoints on phylogenetic relations among the Pacific salmon.  相似文献

Some new data on the expansion, host-animals, biology and morphology of different developmental stages of the trematode Exorchis oviformis in the southern Prymorye are given. It was revealed, that the first intermediate host of that species is Stenothyra (S.) recondita. Different species of freshwater fishes play a role of a second intermediate host. The definitive host of E. oviformis is a bullhead (Parasilurus asotus).  相似文献

In 41 individuals of South Asian Dolly Varden char Salvelinus curilus, nucleotide sequences of tRNA-Pro gene fragment (27 bp) and mtDNA control region (483-484 bp) were analyzed. The fish were collected in 20 localities covering virtually the whole range of the species: Kuril Islands, Sakhalin Island. and Primorye. In addition, six individuals of three other char species (S. albus, S. malma, and S. leucomaenis), which are closely related to S. curilus and inhabit the Russian Far East, were examined. In all, we detected 12 different variants of mtDNA haplotypes that formed three distinct groups differing in 14--20 nucleotide positions. The first group consisted of six haplotypes found in S. curilus in Kuril Islands, Sakhalin, and Primorye (mtDNA phylogroup OKHOTSKIA). The second group comprised four haplotypes representing the mtDNA phylogroup BERING, which had been described earlier (Brunner et al, 2001); they were found in S. curilus in Kuril Islands and Sakhalin, as well as in S. albus and S. malma in Kamchatka and northern Kurils. The third group included two haplotypes detected in S. leucomaenis. The existence of two mtDNA lineages (OKHOTSKIA and BERING) in S. curilus from Kurils and Sakhalin was explained by hybridization and DNA transfer from S. malma to S. curilus. The absence of the BERING haplotypes in S. curilus from Primorye water reservoirs is related to the physical isolation of the Sea of Okhotsk and Sea of Japan basins in past epochs. On the basis of comparing phylogenetic trees, constructed from the data on allozyme and mtDNA variation, we suggest that in this case, an indirect transfer of mtDNA in Alpinoid chars--> S. malma-->S. curilus chain could occur.  相似文献

Comparative analysis of personal sequence data for the mtDNA control region (926 to 928 bp) from eight-barbel loaches inhabiting eight localities in the Amur River basin (4) and the Sea of Japan (4) and the GeneBank/NCBI data for the Lefua individuals from the other regions of the world showed that eight-barbel loaches from Primorskii krai water basins were marked by a specific group of mtDNA haplotypes. This finding is considered as supporting the species status of L. pleskei. Genetic distances within L. pleskei are small (on average 0.355) and close to those within L. nikkonis (on average 0.48%). The distances between this species pair are the least (on average 2.15%) among all other pairs compared. In MP, ML, and Bayesian trees, L. pleskei and L. nikkonis haplotypes formed a common clade with high statistically significant support. In all tree variants, L. costata mtDNA haplotypes were located out of the group of interest. A clade consisting of highly diverged lineages of Lefua sp. and L. echigonia haplotypes occupied even more independent position. The mtDNA haplotypes of L. pleskei and L. costata from the Amur River basin were evolutionary young and derived from the haplotypes found in these species from the Sea of Japan (L. pleskei) or the Yellow Sea (L. costata) basins. It is thereby suggested that both species rather recently migrated into the Amur River system. According to the molecular clock data, basal diversification of the eight-barbel loach lineages took place at the end of middle Miocene (about 11 to 12 Myr ago), while divergence of L. pleskei and L. costata ancestral forms probably occurred approximately, 5 Myr ago. Since all main lineages of eight-barbel loaches were found in the Sea of Japan basin (continental coastline and the islands), the divergence order and dispersal patterns of the Lafua species might have been largely determined by the geological development pattern of this water body and the adjacent territories.  相似文献

On the basis of comparison of the cytochrome b gene nucleotide sequences from genetic databases, the possible phylogenetic relationships of mitochondrial DNA (mtDNA) among all major lineages of Salmoninae (Brachymystax, Parahucho, Salvelinus, Salmo, Parasalmo, and Oncorhynchus) were examined. Three different phylogenetic methods (UPGMA, NJ, and ML) yielded phylogenetic trees of essentially the same topology: (((Brachymystax, Parahucho), Salvelinus, Salmo), (Parasalmo, Oncorhynchus)). The results obtained using the maximum parsimony method were less clear. Apparently, the divergence of the main salmonid lineages occurred during a relatively short time period; hence, the number of synapomorphs marking the order of their divergence was extremely low. This may account for the relative failure to use the maximum parsimony method of phylogenetic reconstruction. The problem of concordance of mtDNA and species phylogenetic schemes is discussed. Their discrepancy in salmonids may be caused by interspecific introgressive hybridization.  相似文献

Autopsies of Abbottina rivularis from southern Primorye (drainage-basin of Artyomovka River, Razdolnaya River and Khanka Lake) and southern Sakhalin Island (Maloye Chibisanskoye Lake) revealed high indices of diplostomum-infected lenses of these fishes. The metacercariae have been identified as Diplostomum parviventosum Dubois, 1932, D. huromense (La Rue, 1927), D. helveticum (Dubois, 1929), D. mergi Dubois, 1932, Diplostomum sp. The most lenses of parasitized eyes possessed dorsally situated sperical protrusions of the lens capsule ("cyst"). Earlier, this phenomenon was found by Larson (1965) in naturally infected bullheads (Ictalurus). The metacercarial infection level and its dynamics, age-composition of metacercariae in both lenses and "cysts", and "cysts" production are discussed.  相似文献

Parasitological analysis of sockeye salmon fry from Lake Azabach'e has shown reliable differences in the infection between aboriginal and migratory in the lake fry with the nematode Cystidicola farionis from the swim bladder, that is connected with differences in the nature of their feeding and ecology. It was found out that the infection of migratory fry occurs only in the lake during feeding and wintering periods, one-summer-old fry 35 to 57 mm in length being primarily infected. Quantitative criteria (the abundance of immature nematodes of C. farionis, the type of feeding) were ascertained, which in combination with scale characters differentiate 95.9 per cent of fry into aboriginal and migratory.  相似文献
