Computer analysis revealed seven potential variable-number tandem-repeat (VNTR) loci in the Vibrio cholerae genome. Specific primers were designed to amplify locus VcA located on chromosome 2 and containing a TGCTGT repeat. The locus was found in all tested strains from aV. cholerae strain collection, the repeat number varying from 3 to 23. In total, 14 VcA alleles were observed. The VcA locus was proposed as a marker for the molecular typing of V. cholerae strains. 相似文献

The study of the activity, protein charge and isoenzyme LDG spectrum after a single injection of formaldehyde solution has demonstrated a rise in the protein negative charge, which in the absence of ischemia was observed for 20 min. A more than 5-min increase in LDG protein charge observed in ischemia and independent of changes in LDG isoenzyme spectrum was significantly less expressed in ischemic brain after a preliminary injection of formaldehyde solution. With cerebral blood flow preserved, an injection of formaldehyde resulted in the alteration of LDG spectrum, namely a rise in LDG3+4+5 content. Formaldehyde injection prior to ischemia prevented the development of a similar effect, which was significantly more expressed in ischemic brain without a preliminary formaldehyde injection. 相似文献

Methods of wide-angle X-ray scattering (WAXS), high resolution solid-state 13C NMR, and Fourier transform IR-spectroscopy are applied to study supermolecular structures and functional compositions of lignocellulose samples of wood and grass origins and powdered celluloses (PC) obtained from them under identical hydrolysis conditions. It was shown by WAXS that the structure of cellulose I is preserved in samples of powdered celluloses, however, an increased degree of crystallinity and cross-section sizes of crystallites are observed in PC samples. Specific features of changes in the supermolecular structure of cellulose occurred after the hydrolysis, i.e., an increase in the content of cellulose Iβ in PC compared to the initial samples, are established by 13C NMR method. It was shown by means of 13C NMR and Fourier transform IR-spectroscopy that the functional chemical composition of lignocelluloses is weakly affected by the hydrolysis. The presence of residual lignin functional groups in the samples is confirmed. 相似文献

The influence of the human tumor suppressor PTEN on sensitivity of tumor cells to cytostatic drugs was studied. Rat ras-transformed (N-ras Asp12 ) fibroblasts were stably transfected with a full-size PTEN gene. Transfected clone was characterized by an enhanced expression of PTEN and a more normal phenotype in comparison with the parental cells. The effect of transient transfection with PTEN on the sensitivity of several malignant cell lines to the cytostatic drugs colchicine and adriablastine was studied. These drugs differ from each other in action mechanisms and intracellular targets. The tumor cell lines tested in this study included parental cell lines and stable sublines possessing drug resistance due to overexpression of P-glycoprotein. In all cell lines, introduction of exogenous PTEN caused a decrease in proliferation rates. This indicated that transgene was active. The chemosensitivity of some drug-resistant sublines was changed after PTEN transfection, but the drug sensitivity of parental cell lines remained unaffected. The effect of PTEN overexpression on chemosensitivity of malignant cells to cytostatic drugs was found to depend both on their mechanisms of action and on the origin of transfected cells. Our data suggest that PTEN is involved into the molecular mechanisms of drug resistance in cells studied. 相似文献

In the yeast Dipodascus magnusii, which is auxotrophic for thiamine and biotin, during cultivation on glucose with excessive thiamine concentration, pyruvate metabolism was shown to result in the synthesis of fermentation products, namely, ethanol and, to a lesser extent, lactate. Substantial synthesis of ethyl acetate was also observed under these conditions. Introduction of nicotinic acid (NA) into the medium resulted in time separation of ethanol and lactate production. It was shown that cultivation of the yeast under biotin deficiency resulted in nearly complete suppression of aerobic production of ethanol and cessation of ethyl acetate synthesis, whereas lactate synthesis was activated as early as in the first hours of cultivation. Upon introduction of NA under these conditions, lactate concentration sharply increased. These results show that the combination of thiamine and biotin with other vitamins can stimulate utilization of the pyruvate pool in yeasts towards formation of considerable amounts of lactate, which is typical only of cells of higher eukaryotes and bacteria. 相似文献

The impact of monoclonal antibodies (mAb) biotinylation on the output and the repertoire of selected peptides in the biopanning procedure were tested. A comparative analysis of the peptides selected from phage library using the biotinylated and non-biotinylated mAb 2F5 was performed. It was shown that the output of peptides homologous to the native epitope was 1.7-fold higher for biotinylated antibodies, whereas the binding capacity of the selected phages with mAb 2F5 in ELISA was higher in the case of using non-biotinylated antibodies. It should be noted that the phages exposing peptides, which have 4-5 amino acid sequence similarity with the native epitope, demonstrate the highest binding affinity. The phages that expose peptides with 3 amino acid sequence similarity demonstrate different binding affinity: from the smallest to the largest. Based on the obtained data, it is safe to suggest that the rational biopanning may proceed in accordance with the task. 相似文献
