Melanin complex of the fungus Inonotus obliquus 总被引:6,自引:0,他引:6
The fungus Inonotus obliquus (Pers.) Pil. synthesised high-molecular-weight phenolic pigments that were assigned to melanins according to their physicochemical properties. It was showed that copper ions (0.008%), pyrocatechol (1.0 mM), and tyrosine (20.0 mM) stimulated the melanogenesis. The production of melanin correlated with the synthesis of o- and p-diphenoloxidases. The fungal melanin had strong antioxidant and genoprotective effects. 相似文献

Pigments synthesized by micromycetes Paecilomyces variotii and Aspergillus carbonarius are true melanins. Copper ions and bicyclic phenolic compounds stimulated melaninogenesis, whereas benzotriazole inhibited this process. Precursors of melanin pigments were obtained and identified. p-Hydroxybenzoic acid was shown to be the main product of melanin degradation. It was demonstrated that the total carbohydrate content in the submerged mycelium of xylotrophic basidiomycetes falls into the range of 34.66–57.57%. Free carbohydrates of the cell cytosol amounted to 14.52–28.64%; structural carbohydrates, to 20.14–32.70%. Qualitative composition of carbohydrates was studied, which allowed for attribution of structural polysaccharides to hetero- and homoglycans with - and -glycoside bonds. 相似文献

Aflatoxins are polyketide-derived secondary metabolites produced by Aspergillus spp. The toxic effects of aflatoxins have adverse
consequences for human health and agricultural economics. The aflR gene, a regulatory gene for aflatoxin biosynthesis, encodes a
protein containing a zinc-finger DNA-binding motif. AFLR-Protein three-dimensional model was generated using Robetta server.
The modeled AFLR-Protein was further optimization and validation using Rampage. In the simulations, we monitored the
backbone atoms and the C-α-helix of the modeled protein. The low RMSD and the simulation time indicate that, as expected, the
3D structural model of AFLR-protein represents a stable folding conformation. This study paves the way for generating computer
molecular models for proteins whose crystal structures are not available and which would aid in detailed molecular mechanism of

New inhibitors of melanin formation by micromycetes Aspergillus carbonarius, Alternaria alternata, and Paecilomyces variotii and basidiomycetes Inonotus obliquus and Phellinus robustus were found. Precursors of melanin pigments were isolated and identified. p-Hydroxybenzoic acid was identified among the products of alkaline degradation of melanin formed by micromycetes, whereas in the case of macromycetes this was protocatechuic acid. Therefore, melanins of the former were found to belong to the dihydronaphthalene group, whereas those of the latter belong to catechols. 相似文献

Visualization of molecular structures aids in the understanding of structural and functional roles of biological macromolecules.
Macromolecular transport between the cell nucleus and cytoplasm is facilitated by the nuclear pore complex (NPC). The ring
structure of the NPC is large and contains several distinct proteins (nucleoporins) which function as a selective gate for the passage
of certain molecules into and out of the nucleus. In this note we demonstrate the utility of a python code that allows direct
mapping of the physiochemical properties of the constituent nucleoporins on the scaffold of the yeast NPC׳s cytoplasmic view. We
expect this tool to be useful for researchers to visualize the NPC based on their physiochemical properties and how it alters when
specific mutations are introduced in one or more of the nucleoporins. The code developed using Python is available freely from the

Fungal melanin pigments were shown to display a high antioxidant activity. An increase in the number of methyl substituents
in benzidine molecules of melanins obtained from micromycetes and macromycetes was accompanied by a decrease in the efficiency
of inhibition of peroxidase-mediated oxidation. Melanins were found to have considerable gene-protecting properties. Pigments
isolated from macromycetes and applied at a much lower concentration than those obtained from micromycetes prevented damage
to bacteriophage-λ DNA induced by products of peroxidase-mediated degradation of aminobiphenyls. 相似文献
