Excised grains of wheat (Triticum aestivum) varieties HD 2285 (relatively tolerant) and HD 2329 (susceptible type) were incubated for 1 hr at 15 degrees, 25 degrees, 35 degrees and 45 degrees C. In an another treatment, excised grains were incubated for 1 hr at increasing temperature (15 degrees, 25 degrees, 35 degrees and 45 degrees C) continuously, thus exposing the grains to gradual rise in temperature. The above treated grains were then analysed for the activity of soluble starch synthase (SSS) and granule bound starch synthase (GBSS) assayed at 20 degrees C. SSS activity decreased as the pre-exposure temperature was higher, though the tolerant variety showed lesser decrease. Decrease in SSS activity was lesser when excised grains were exposed to gradual rise in temperature from 15 degrees to 45 degrees C as compared to direct exposure to 45 degrees C. Pre-exposure of excised grains to different temperatures however, had no significant effect on GBSS activity.  相似文献

Wheat (Triticum aestivum L. cv. HD 2329 and DL 1266-5) and sunflower (Helianthus annuus L. cv. MSFH 17 and MRSF 1754) plants were grown in field under atmospheric (360±10 cm3 m−3, AC) and elevated (650±50 cm3 m−3, EC) CO2 concentrations in open top chambers for entire period of growth and development till maturity. Net photosynthetic rate (P N) of wheat cvs. when compared at the same internal CO2 concentration (C i), by generating P N/C i curves, showed lower P N in EC plants than in AC ones. EC-grown wheat cultivars also showed a lesser response to irradiance than AC plants. In sunflower cultivars, P N/C i curves and irradiance response curves were not significantly different in AC and EC plants. CO2 and irradiance responses of photosynthesis, therefore, further revealed a down-regulation of P N in wheat but not so in sunflower under long-term CO2 enrichment. Wheat cvs. accumulated in leaves mostly sugars, whereas sunflower accumulated mainly starch. This further strengthened the view that accumulation of excess assimilates in the leaves under EC as starch is not inhibitory to P N.  相似文献

Wheat (T. aestivum) cvs. Kalyansona and Kundan grown under atmospheric (CA) and elevated CO2 concentrations (650±50 cm3 m-3 - CE) in open top chambers were examined for net photosynthetic rate (PN), stomatal limitation (l s) of P N, ribulose-1,5-bisphosphate carboxylase (RuBPC) activity, and saccharide content of the leaves. The P N values of both CA- and CE-grown plants compared at the same CO2 concentration showed a down regulation under CE at the post-anthesis stage. The negative acclimation of P N appeared to be due to both stomatal and mesophyll components, and the RuBPC activity got also adjusted. There was a decrease in activation state of RuBPC under CE. In connection with this, an increased accumulation of saccharides in wheat leaf under CE was observed. Kalyansona, owing to its larger sink potential in terms of the number of grains, showed a greater enhancement under CE in both post-ear emergence dry matter production and grain yield. Under CE, this cultivar also showed a lower down regulation of P N than Kundan. This revised version was published online in June 2006 with corrections to the Cover Date.  相似文献

Wheat (Triticum aestivum L.) cvs. HD 2285 (relatively tolerant) and WH 542 (susceptible) were exposed to ambient and elevated temperature (3–4 °C higher) in open top chambers during post anthesis period. The grain yield components were determined at the time of maturity. In order to elucidate the basis of differential tolerance of these cultivars, the excised developing grains (20 d after anthesis) of ambient grown plants were incubated at 15, 25, 35 and 45 °C for 2 h and then analysed for the activities of soluble starch synthase (SSS), granule bound starch synthase (GBSS), kinetic parameters of SSS and content of heat shock protein (HSP 100). The elevated temperature during grain development significantly decreased grain growth in WH 542 whereas no such decrease was observed in HD 2285. High temperature tolerance of HD 2285 was found to be associated with higher catalytic efficiency (Vmax/Km) of SSS at elevated temperature and higher content of HSP 100.  相似文献

Two wheat varieties, T. durum (HD 4502) and T. aestivum (Kalyansona) were examined for photosynthesis rate and contents of sugars and rubisco protein in the flag leaf, at forenoon and afternoon at anthesis stage. A decrease in photosynthesis rate was observed in the afternoon compared to forenoon in both the varieties and was associated with an increase in non-reducing sugars and a decrease in rubisco content in the leaves.  相似文献

Wheat (Triticum aestivum L. cv. HD 2285) was grown in control (C) and heated (H) open top chambers (OTCs) for entire period of growth and development till maturity. The mean maximum temperature of the entire period was 3 °C higher in H-compared to C-OTCs. Net photosynthetic rate (P N) measured at different temperature (20–40 °C) of C-and H-grown plants showed greater sensitivity to high temperature in H-plants. P N measured at respective growth temperature was lower in H-compared to C-plants. The CO2 and irradiance response curves of photosynthesis also showed lesser response in H-compared to C-plants. The initial slope of P N versus internal CO2 concentration (P N/C i) curve was lower in H-than C-plants indicating ribulose-1,5-bisphosphate carboxylase/oxygenase (RuBPCO) limitation. In irradiance response curve, the plateau was lower in H-compared to C-plants which is interpreted as RuBPCO limitation. RuBPCO content in the leaves of C-and H-plants, however, was not significantly different. Ribulose-1,5-bisphosphate carboxylase (RuBPC) initial activity was lower in H-plants, whereas activity of fully activated enzyme was not affected, indicating a decrease in activation state of the enzyme. This was further substantiated by the observed decrease in RuBPCO activase activity in H-compared to C-plants. RuBPCO activase was thus sensitive even to moderate heat stress. The decrease in P N under moderate heat stress was mainly due to a decrease in activation state of RuBPCO catalysed by RuBPCO activase.  相似文献

Wheat (T. durum cvs. HD 4502 and B 449, T. aestivum cvs. Kalyansona and Kundan) and sunflower (Helianthus annuus L. cv. Morden) were grown under atmospheric (360±10 cm3 m–3, AC) and elevated CO2 (650±50 cm3 m–3, EC) concentration in open top chambers for entire period of growth and development till maturity. Leaf net photosynthetic rate (P N) of EC-grown plants of wheat measured at EC was significantly decreased in comparison with AC-plants of wheat measured at EC. Sunflower, however, showed no significant depression in P N in EC-plants. There was a decrease in ribulose-1,5-bisphosphate carboxylase (RuBPC) activity, its activation state and amount in EC-plants of wheat, whereas no significant decrease was observed in sunflower. The above different acclimation to EC in wheat and sunflower was related with saccharide constituents accumulated in the leaves. Under EC, sunflower accumulated in the leaves more starch, whereas wheat accumulated more sugars.  相似文献

With rising level of CO2 in the atmosphere plants are expected to be exposed to higher concentration of CO2. Since, CO2 is a substrate limiting photosynthesis particularly in C3 plants in the present atmosphere, the impact of elevated CO2 would depend mainly on how photosynthesis acclimates or adjusts to the long term elevated level of CO2. Photosynthetic acclimation is a change in photosynthetic efficiency of leaves due to long term exposure to elevated CO2. This change in photosynthetic efficiency could be a biochemical adjustment that may improve the overall performance of a plant in a high CO2 environment or it could be due to metabolic compulsions as a result of physiological dysfunction. Acclimation has generally become synonymous with the word response, if long term exposure to elevated CO2 decreases the photosynthesis rate (Pn) at a given CO2 level, it is called negative acclimation, if it stimulates Pn at a given CO2 level, it is called positive acclimation. Photosynthetic acclimation is clearly revealed by comparing Pn of ambient and elevated CO2 grown plants at same level of CO2. Species level differences in acclimation to elevated CO2 have been reported. The physiological basis of differential photosynthetic acclimation to elevated CO2 is discussed in relation to the regulation of photosynthesis and photosynthetic carbon partitioning at cellular level.  相似文献
