Thermowaves spreading through the cerebral cortex were observed and investigated during acute experiments on white rats using a new technique — that of thermoencephaloscopy (TES) through the intact skull. These waves were induced by regular visual stimulation (at intervals of 1.5–3 min) or by initiating one of the flashes and spread through the cortex along trajectories of five basic types; amplitude of thermowaves: 0.005–0.1°C; length: 10–15 mm; duration 1.2–11.4 sec; velocity: 1–33 mm/sec; extent of pathway: 2–56 mm. They appeared with a high degree of probability (of 0.92) during the interval between 15 sec before and 26 sec after the flash. Numbers of moving waves declined by the point of stimulus application and rose by 7–8 sec after the flash. Waves arose in 50% of cases in the contralateral visual cortex (areas 17 and 18a), spreading to the midline and crossing to the ipsilateral hemisphere (areas 17, 18a, and 7). Local waves moving along a circular trajectory were also discovered in the contralateral visual cortex. Several types of wave, differing in trajectory, also arose in the ipsilateral visual cortex. The effect of parameters of local photic stimulation of different points of the receptive field on the characteristics of dynamic reorganization of receptive fields of cortical and geniculate visual neurons within microintervals of time observed previously was studied in computer-controlled experiments on unanesthetized, curarized cats. Dependence on the degree of widening of the receptive field and the temporal characteristics of this process on the background illumination level, intensity, contrast, area, duration, energy, and orientation of a local rectangular or circular photic stimulus flashing in random order at 100 points of the tested part of the visual field was studied. It was concluded that the background illumination level and the intensity, size, duration, and orientation of the stimulus have a specific effect on dynamic reorganization of the receptive field. The effects of all the parameters studied on the dynamics of the receptive field were shown to be nonlinear functions with optimal values that differed for different cells. Orientation selectivity of 24 neurons in area 17 of the visual cortex at different intensities of test bars of light, flashing against a constant light background in the center of the receptive field, was investigated in acute experiments on immobilized cats. Five neurons were invariant in orientation tuning to stimulus intensity (contrast): Although the magnitude of the response and acuteness of orientation selectivity were modified, preferential orientation was unchanged. More than half of the cells studied (13) were classed as noninvariant, for their preferential orientation was significantly shifted by 22–90° with a change in contrast. Small shifts of the peak of orientation selectivity, not statistically significant, were observed for the other neurons. Invariant neurons, unlike noninvariant, were characterized by preferential horizontal and vertical orientation, a lower frequency of spontaneous and evoked discharges, and the more frequent presence of receptive fields of simple type. The mechanisms of the change of orientation selectivity during contrast variation and also the different use of the two types of cells in orientation detection operations are discussed.  相似文献

A high molecular weight glycoconjugate has been isolated from neurite-producing neuronal tumor cells in culture and has been designated as I(0) based on its elution characteristics in gel filtration chromatography. This molecule cannot be found in a variety of nonneuronal cells. I(0) is found in the substratum-attached material or cell fraction of neurite-producing neuroblastoma cells, depending upon culture conditions. It is found in the substratum-bound fraction of B104 rat neuroblastoma cells during serum starvation and in the EGTA-detached cell fraction of B104 cells grown in chemically defined N2 medium. It occurs only in the cell fraction of the human neuroblastoma line Platt. Examination of behavioral variants of the B104 rat line further strengthens the association of I(0) with neurite production; the constitutive neurite-producing E(R)B9 variant contains I(0) while the non-neurite-producing E(R)A11 variant does not. I(0) is large, eluting in the void volume of sepharose-CL2B columns. Radioiodination of intact cells with lactoperoxidase shows I(0) to be a cell surface component. Metabolic radiolabeling studies show that it contains a high proportion of polysaccharide to protein, does not contain mannose, and is unsulfated. Alkaline borohydride reduction release two size classes of large polysaccharide chain. The alkaline reduction results, along with the mannose incorporation studies, show the presence of O-glycosidic linkages and few, if any, N-linkages. Resistance to nitrous acid deamination, insensitivity to glycosaminoglycan lyases, and the absence of sulfation, indicate that I(0) does not contain the glycosaminoglycans hyaluronic acid, chondroitin-, dermatan-, or heparin- sulfates. Affinity column chromatography reveals high binding affinity of I(0) to polyornithine and no binding to gelatin (collagen) or the glycosaminoglycans hyaluronate and heparin. These studies describe a unique high molecular weight glycoconjugate on the surface of neurite-producing neuroblastoma cell lines from two species.  相似文献

Dynamics of orientational tuning in 59 primary visual cortex neurons were investigated before and after sombrevine-induced anesthesia during acute experiments on immobilized cats using temporal slice techniques. A dynamic shift in preferred orientation of a flashing light strip, during which peak amplitude of spike discharges was noted (at an angle of between 22 and 157°) occurred as response developed in two-thirds of the cells. We had previously named this effect "scanning the orientational range" [9]. Scanning declined significantly in 45% of the sample, culminating in complete disappearance of this effect in some cells following sombrevine action. Scanning intensified in 30%, while dynamics of tuning remained unchanged in 25% of units. Sombrevine administration induced change in the preferred stimulus orientation of 60% of the neurons (referred to as "unstable" cells) and remained constant in "stable" cells (= 40%). Dynamic changes in preferred stimulus orientation were 2.5 times as high as those of stable cells in the waking state. The scanning effect declined significantly in 60% of "unstable" neurons under the action of anesthesia and remained unchanged in not more than 6%. At the same time, orientational tuning did not alter in the "stable" cell group in 46% of units, either declining (25%) or increasing (29%) in the remaining scanning ranges.Institute of Higher Nervous Activity and Neurophysiology, Academy of Sciences of the USSR, Moscow. We have developed a new approach to the measurement of phylogenetic signal in character state matrices called relative apparent synapomorphy analysis (RASA). RASA provides a deterministic, statistical measure of natural cladistic hierarchy (phylogenetic signal) in character state matrices. The method works by determining whether a measure of the rate of increase of cladistic similarity among pairs of taxa as a function of phenetic similarity is greater than a null equiprobable rate of increase. Our investigation of the utility and limitations of RASA using simulated and bacteriophage T7 data sets indicates that the method has numerous advantages over existing measures of signal. A first advantage is computational efficiency. A second advantage is that RASA employs known methods of statistical inference, providing measurable sensitivity and power. The performance of RASA is examined under various conditions of branching evolution as the number of characters, character states per character, and mutations per branch length are varied. RASA appears to provide an unbiased and reliable measure of phylogenetic signal, and the general approach promises to be useful in the development of new techniques that should increase the rigor and reliability of phylogenetic estimates.   相似文献

We have analyzed nucleic acid and amino acid sequence alignments of a variety of voltage-sensitive ion channels, using several methods for phylogenetic tree reconstruction. Ancient duplications within this family gave rise to three distantly related groups, one consisting of the Na+ and Ca++ channels, another the K+ channels, and a third including the cyclic nucleotide-binding channels. A series of gene duplications produced at least seven mammalian homologues of the Drosophila Shaker K+ channel; clones of only three of these genes are available from all three mammalian species examined (mouse, rat, and human), pointing to specific genes that have yet to be recovered in one or another of these species. The Shaw-related K+ channels and the Na+ channel family have also undergone considerable expansion in mammals, relative to flies. These expansions presumably reflect the needs of the high degree of physiological and neuronal complexity of mammals. Analysis of the separate domains of the four-domain channels (Ca++ and Na+) supports their having evolved by two sequential gene duplications and implies the historical existence of a functional two-domain channel.   相似文献

Reconstructions of the human-African great ape phylogeny by using mitochondrial DNA (mtDNA) have been subject to considerable debate. One confounding factor may be the lack of data on intraspecific variation. To test this hypothesis, we examined the effect of intraspecific mtDNA diversity on the phylogenetic reconstruction of another Plio- Pleistocene radiation of higher primates, the fascicularis group of macaque (Macaca) monkey species. Fifteen endonucleases were used to identify 10 haplotypes of 40-47 restriction sites in M. mulatta, which were compared with similar data for the other members of this species group. Interpopulational, intraspecific mtDNA diversity was large (0.5%- 4.5%), and estimates of divergence time and branching order incorporating this variation were substantially different from those based on single representatives of each species. We conclude that intraspecific mtDNA diversity is substantial in at least some primate species. Consequently, without prior information on the extent of genetic diversity within a particular species, intraspecific variation must be assessed and accounted for when reconstructing primate phylogenies. Further, we question the reliability of hominoid mtDNA phylogenies, based as they are on one or a few representatives of each species, in an already depauperate superfamily of primates.   相似文献

Selectivity and invariability of tuning were studied in 51 neurons of the primary visual cortex (area 17); cruciform and angular figures (CF and AF, respectively) of different configurations and orientations were presented in their receptive fields. Twenty-three neurons, or 45% of the studied cells, demonstrated selective sensitivity to these figures. Their responses considerably (2.38±0.36 times, on average) increased, as compared with those evoked by presentation of a single bar of preferred orientation. In the examined group, 2 cells demonstrated sensitivity both to the CF and AF. A wide range of detector properties related to the CF and AF analysis was found in the analyzed neuronal population. Detectors of configuration of these figures are described. Selective sensitivity to the angle between branches of these figures was observed in 17 neurons, and responses of 2 neurons among them showed invariability to orientation of these figures. Four cells were selective for orientation and were insensitive to configuration, and 4 other cells showed no specific sensitivity to either of these properties, but were sensitive to the appearance of a CF itself in their receptive field (these cells were regarded as invariant detectors of crossing nodes). Data inconsistent with the hierarchic principle of detection of the above properties are presented.
