Wireworms, the larvae of click beetles (Elateridae), are difficult to manage due to their habitats and behaviour. Wireworms pose a major threat to the wheat crop in the north‐western USA. Seed treatment with neonicotinoids, biological control management and some cultural controls are recommended to manage these pests. Trap cropping is an emerging way to manage wireworms. In strawberry and potato crops, trap cropping has been found effective at attracting wireworms away from the principal crop. An earlier study in the Golden Triangle area of Montana found that pea and lentil could be effective trap crops for managing wireworms in spring wheat. In the present study, experiments were conducted at two locations. The effectiveness of peas and lentils as trap crops with wheat at different seeding densities was assessed [pea at 0, 4, 8, 16 seeds/sq.ft. or 0, 43, 86, 172 seeds/sq.m.; lentil at 0, 6, 12, 18 OR 0, 65, 130, 194 seeds/sq.m.; both with wheat at 0, 11, 22, or 28 seeds/sq.ft. or 0, 120, 230, 300 seeds/sq.m.] in a randomized design where all three crops were intercropped. Both trap crops were found to be effective in protecting wheat at standard seeding rates of 8 seeds/sq.ft. for pea and 12 seeds/sq.ft. for lentils. At these seeding rates, higher numbers of wireworms were found to be attracted to the trap crop, resulting in higher yield (7%–10%) of the associated spring wheat plant stands at 22 seeds/sq.ft. To develop an effective trap crop strategy, the pea–wheat and lentil–wheat spatial patterns that are possible need to be assessed in further field trials. Proper design and evaluation of the cost–benefit ratio of pea and lentil as trap crops are likely to produce good results for wheat crops in Montana.  相似文献

PIEZO channels are force sensors essential for physiological processes, including baroreception and proprioception. The Caenorhabditis elegans genome encodes an orthologue gene of the Piezo family, pezo-1, which is expressed in several tissues, including the pharynx. This myogenic pump is an essential component of the C. elegans alimentary canal, whose contraction and relaxation are modulated by mechanical stimulation elicited by food content. Whether pezo-1 encodes a mechanosensitive ion channel and contributes to pharyngeal function remains unknown. Here, we leverage genome editing, genetics, microfluidics, and electropharyngeogram recording to establish that pezo-1 is expressed in the pharynx, including in a proprioceptive-like neuron, and regulates pharyngeal function. Knockout (KO) and gain-of-function (GOF) mutants reveal that pezo-1 is involved in fine-tuning pharyngeal pumping frequency, as well as sensing osmolarity and food mechanical properties. Using pressure-clamp experiments in primary C. elegans embryo cultures, we determine that pezo-1 KO cells do not display mechanosensitive currents, whereas cells expressing wild-type or GOF PEZO-1 exhibit mechanosensitivity. Moreover, infecting the Spodoptera frugiperda cell line with a baculovirus containing the G-isoform of pezo-1 (among the longest isoforms) demonstrates that pezo-1 encodes a mechanosensitive channel. Our findings reveal that pezo-1 is a mechanosensitive ion channel that regulates food sensation in worms.  相似文献

Soil nematode community response to treatments of three, four-year crop rotations (spring wheat-pea-spring wheat-flax, spring wheat-green manure-spring wheat-flax, and spring wheat-alfalfa-alfalfa-flax) under conventional and organic management, and native tall grass restoration (restored prairie) were assessed in June 2003, and July and August 2004. The research site was the Glenlea Long-term Rotation and Crop Management Study, in the Red River Valley, Manitoba, established in 1992. The nematode community varied more with sample occasion than management and rotation. The restored prairie favored high colonizer-persister (c-p) value omnivores and carnivores, and fungivores but less bacterivores. The restored prairie soil food web was highly structured, mature and low-to-moderately enriched as indicated by structure (SI), maturity (MI) and enrichment (EI) index values, respectively. Higher abundance of fungivores and channel index (CI) values suggested fungal-dominated decomposition. Nematode diversity was low even after more than a decade of restoration. A longer time may be required to attain higher diversity for this restored fragmented prairie site distant from native prairies. No consistent differences were found between organic and conventional management for nematode trophic abundance, with the exception of enrichment opportunists of the c-p 1 group which were favored by conventional management. Although EI was lower and SI was higher for organic than conventional their absolute values suggested decomposition channels to be primarily bacterial, and fewer trophic links with both management scenarios. A high abundance of fungivores in the rotation including the green manure indicates greater fungal decomposition.  相似文献

The estimation and maintenance of connectivity among local populations is an important conservation goal for many species at risk. We used Bayesian statistics and coalescent theory to estimate short- and long-term directional gene flow among subpopulations for two reptiles that occur in Canada as peripheral populations that are geographically disjunct from the core of their respective species’ ranges: the black ratsnake and the Blanding’s turtle. Estimates of directional gene flow were used to examine population connectivity and potential genetic source-sink dynamics. For both species, our estimates of directional short- and long-term gene flow were consistently lower than estimates inferred previously from F ST measures. Short- and long-term gene flow estimates were discordant in both species, suggesting that population dynamics have varied temporally in both species. These estimates of directional gene flow were used to identify specific subpopulations in both species that may be of high conservation value because they are net exporters of individuals to other subpopulations. Overall, our results show that the use of more sophisticated methods to evaluate population genetic data can provide valuable information for the conservation of species at risk, including bidirectional estimates of subpopulation connectivity that rely on fewer assumptions than more traditional analyses. Such information can be used by conservation practitioners to better understand the geographic scope required to maintain a functional metapopulation, determine which habitat corridors within a working landscape may be most important to maintain connectivity among subpopulations, and to prioritize subpopulations with respect to their potential to act as genetic sources within the metapopulation.  相似文献

We used mitochondrial DNA and microsatellite loci to examine the phylogeographic patterns of the most broadly distributed lizard in eastern North America, the five-lined skink (Eumeces fasciatus). We infer that longitudinal phylogeographic patterns in E. fasciatus are consistent with fragmentation due to refugial and post-glacial dynamics, but that deep divergences within the species imply historical fragmentation that predates the Pleistocene. The effect of multiple refugia is implied from our nested clade analyses, including a northern refugium in Wisconsin. Analysis of population structure using nuclear microsatellite data within the species suggests the importance of glacial dynamics in shaping more recent genetic structuring within one widely distributed lineage that ranges from the Mississippi River to the Atlantic Ocean in longitude and from southern Ontario to the Gulf of Mexico in latitude. Results shed light on the historical processes that have influenced current population structure of a temperate lizard, support the striking similarity of longitudinal phylogeographic structure across many herpetofaunal species in eastern North America, and illustrate the utility of employing multiple markers in phylogeographic studies.  相似文献

High-performance liquid chromatography (HPLC) with UV absorption detection was employed to measure the amounts of 8-hydroxy-2'-deoxyguanosine (8-OH-dG) produced from the nucleoside 2'-deoxyguanosine (dG) under varying reaction conditions using iron and H(2)O(2). The results indicate that 8-OH-dG produced from the reaction of iron and H(2)O(2) with dG can undergo reaction with free (i.e., unchelated) Fe(III) and that adding the chelating agent ethylenediaminetetraacetic acid (EDTA) after the reaction prevents this from occurring. It also appears that the free radical species generated by iron-EDTA chelates in pH 7.4 N-(2-hydroxyethyl)piperazine-N'-(2-ethanesulfonic acid) (Hepes) buffer is either not formed or unstable in unbuffered aqueous solution. Finally, 8-OH-dG levels are significantly larger when Fe(II) is allowed to bind to the nucleoside dG prior to addition of H(2)O(2). However, production of 8-OH-dG from unbound Fe(II) is also relevant. The results of this work show that differing reaction conditions in vivo, especially at the cellular level, will affect significantly the measured yields of 8-OH-dG. These results also have implications for studies involving DNA and the ability to distinguish between 8-OH-dG produced from free iron and iron bound to both phosphate groups and the DNA base guanine.  相似文献

The stem nematode, a parasite of the herbaceous perennial weed, Cirsium arvense (L.) Scop. and identified as Ditylenchus dipsaci (Kühn) Filipjev, was reported in the Canadian prairies in 1979. Recently, D. weischeri Chizhov parasitizing Cirsium arvense was described in Russia, and it has been shown that this species is not an agricultural pest. In this study, we examined Ditylenchus species found in field pea (Pisum sativum L.) grain harvest samples in 2009 and 2010 and from C. arvense shoots in pea fields in the Saskatchewan, Alberta, and Manitoba provinces. Samples from 538 fields (mainly yellow pea) were provided by 151 growers throughout the main pea-growing area of the Canadian prairies. Of the samples collected, 2% were positive for Ditylenchus. The population density of the nematode ranged between 4 and 1,500 nematodes kg-1 pea harvest sample and related to presence of C. arvense seeds. Positive samples occurred in 2009 but not in 2010 and were from throughout the pea-growing area of the Canadian prairies and not related to cropping history. C. arvense collected from yellow pea fields in Saskatchewan and Manitoba, but not Alberta, were infested with Ditylenchus. Morphological and molecular (ITS-PCR-RFLP) traits indicated that this species belongs to D. weischeri. The results indicated the stem nematode found in yellow pea grain is D. weischeri which resided with C. arvense seeds and debris to pea samples. Unlike D. dipsaci, D. weischeri is not a nematode pest of economic importance; therefore, its presence in the pea harvest samples was not a concern.  相似文献

The foundational concept of habitat lies at the very root of the entire science of ecology, but inaccurate use of the term compromises scientific rigor and communication among scientists and nonscientists. In 1997, Hall, Krausman & Morrison showed that ‘habitat’ was used correctly in only 55% of articles. We ask whether use of the term has been more accurate since their plea for standardization and whether use varies across the broader range of journals and taxa in the contemporary literature (1998–2012). We searched contemporary literature for ‘habitat’ and habitat‐related terms, ranking usage as either correct or incorrect, following a simplified version of Hall et al.'s definitions. We used generalized linear models to compare use of the term in contemporary literature with the papers reviewed by Hall et al. and to test the effects of taxa, journal impact in the contemporary articles and effects due to authors that cited Hall et al. Use of the term ‘habitat’ has not improved; it was still only used correctly about 55% of the time in the contemporary data. Proportionately more correct uses occurred in articles that focused on animals compared to ones that included plants, and papers that cited Hall et al. did use the term correctly more often. However, journal impact had no effect. Some habitat terms are more likely to be misused than others, notably ‘habitat type’, usually used to refer to vegetation type, and ‘suitable habitat’ or ‘unsuitable habitat’, which are either redundant or nonsensical by definition. Inaccurate and inconsistent use of the term can lead to (1) misinterpretation of scientific findings; (2) inefficient use of conservation resources; (3) ineffective identification and prioritization of protected areas; (4) limited comparability among studies; and (5) miscommunication of science‐based findings. Correct usage would improve communication with scientists and nonscientists, thereby benefiting conservation efforts, and ecology as a science.  相似文献

Aim  To examine range-wide patterns of genetic diversity in association with range limits in a broadly distributed temperate lizard, and to identify the contributions of a series of environmental, geographical and historical variables to the observed patterns. Reduced genetic diversity may limit local adaptation in peripheral populations, thereby limiting their ability to adapt to marginal environmental conditions, possibly explaining the existence of temporally stable range limits.
Location  Various sampling locales throughout eastern and central USA and southern Ontario, Canada.
Methods  Genetic diversity of nuclear DNA microsatellites was estimated for each of 38 populations from across the range of eastern North America's most broadly distributed lizard, the five-lined skink, Plestiodon fasciatus (Linnaeus, 1758).
Results  Local climatic conditions and an interaction between distance from range border and glaciation history best predicted a population's present-day genetic diversity. Overall, peripheral populations had reduced genetic diversity relative to that of central populations, but this difference was attributable to the reduced genetic diversity in peripheral populations to the north and west that are not bordered by any obvious physical boundaries.
Main conclusions  Some, but not all, peripheral populations had reduced genetic diversity relative to that of more central populations, which probably arose through an interaction of ecological and historical factors. Peripheral populations that were bordered by an obvious boundary (e.g. an ocean) had higher diversity than peripheral populations that were not bordered by an obvious physical barrier to range expansion, suggesting that reduced intrapopulation genetic diversity is associated with range limits in the five-lined skink.  相似文献
