Induction of male flowers in a pistillate line ofRicinus communis L. by silver and cobalt ions

Aqueous solutions of silver nitrate (10–100 g/plant) and cobalt chloride (125–500 g/plant), injected into the main stem of plants of the pistillate cv. 240 ofRicinus communis when the vegetative shoot apex was beginning to become reproductive, induced the formation of staminate (male) flowers with viable pollen in the normally strictly pistillate (female) terminal inflorescence, their number increasing with the dose of Ag⁺ and Co²⁺. No formation of bisexual flowers was noted. Female flowers pollinated with pollen from the induced male ones produced fruits and viable seeds.     

Avirulent mutants ofMacrophomina phaseolina andAspergillus fumigatus initiate infection inPhaseolus mungo in the presence of phaseolinone; levamisole gives protection

To evaluate the role of phaseolinone, a phytotoxin produced by Macrophomina phaseolina, in disease initiation, three nontoxigenic avirulent mutants of the fungus were generated by UV-mutagenesis. Two of them were able to initiate infection in germinating Phaseolus mungo seeds only in the presence of phaseolinone. The minimum dose of phaseoli-none required for infection in 30% seedlings was 2 5 mg/ml. A human pathogen, Aspergillus fumigatus was also able to infect germinating seeds of P. mungo in the presence of 5 mg/ml concentration of phaseolinone. Phaseolinone seemed to facilitate infection by A. fumigatus, which is not normally phytopathogenic, by reducing the immunity of germinating seedlings in a nonspecific way. Levamisole, a non-specific immunopotentiator gave protection against infection induced by A. fumigatus at an optimum dose of 50 mg/ml. Sodium malonate prevented the effects of levamisole.     

Regeneration through organogenesis in rattan

Organogenic cultures were induced from zygotic embryo and megagametophyte explants of the Central American cycad species, Dioon edule. Plant growth medium consisted of B5 major salts, Murashige and Skoog minor salts and organics, 400 mg l⁻¹ glutamine, 100 mg l⁻¹ arginine, 100 mg l⁻¹ asparagine, 60 g l⁻¹ sucrose, 8 g l⁻¹ Difco Bacto agar and was supplemented with kinetin (0 – 13.94 μM) and 2,4-dichlorophenoxyacetic acid (2,4-D) (0 – 9.05 μM) arranged as a 5×4 factorial in a randomized block design. Callus initiation occurred on a wide range of medium formulations from megagametophyte explants; however, shoot formation occurred only on medium supplemented with 2.26 μM 2,4-D. In comparison, callus initiation from explanted zygotic embryos occurred on fewer medium formulations, and adventitious shoot induction occurred from callus on formulations with 9.29–13.94 μM kinetin + 0.45–9.05 μM 2,4-D. Rooted shoots, derived from megagametophyte and zygotic embryo cultures, have been regenerated. This revised version was published online in June 2006 with corrections to the Cover Date.     

Induction of fertile male flowers in genetically female Cannabis sativa plants by silver nitrate and silver thiosulphate anionic complex

Summary Apical application of silver nitrate (AgNO₃; 50 and 100 g per plant) and silver thiosulphate anionic complex (Ag(S₂O₃) ₂ ^3– ; STS; 25, 50 and 100 g per plant) to female plants of Cannabis sativa induced the formation of reduced male, intersexual and fully altered male flowers on the newly formed primary lateral branches (PLBs); 10 g per plant of AgNO₃ was ineffective and 150 g treatment proved inhibitory. A maximum number of fully altered male flowers were formed in response to 100 g STS. The induced male flowers produced pollen grains that germinated on stigmas and effected seed set. Silver ion applied as STS was more effective than AgNO₃ in inducing flowers of altered sex. The induction of male flowers on female plants demonstrated in this work is useful for producing seeds that give rise to only female plants. This technique is also useful for maintaining gynoecious lines.     

Aptamer-Assisted Detection of the Altered Expression of Estrogen Receptor Alpha in Human Breast Cancer

An increase in the expression of estrogen receptors (ER) and the expanded population of ER-positive cells are two common phenotypes of breast cancer. Detection of the aberrantly expressed ERα in breast cancer is carried out using ERα-antibodies and radiolabelled ligands to make decisions about cancer treatment and targeted therapy. Capitalizing on the beneficial advantages of aptamer over the conventional antibody or radiolabelled ligand, we have identified a DNA aptamer that selectively binds and facilitates the detection of ERα in human breast cancer tissue sections. The aptamer is identified using the high throughput sequencing assisted SELEX screening. Biophysical characterization confirms the binding and formation of a thermodynamically stable complex between the identified DNA aptamer (ERaptD4) and ERα (Ka = 1.55±0.298×10⁸ M^-1; ΔH = 4.32×10⁴±801.1 cal/mol; ΔS = -108 cal/mol/deg). Interestingly, the specificity measurements suggest that the ERaptD4 internalizes into ERα-positive breast cancer cells in a target-selective manner and localizes specifically in the nuclear region. To harness these characteristics of ERaptD4 for detection of ERα expression in breast cancer samples, we performed the aptamer-assisted histochemical analysis of ERα in tissue samples from breast cancer patients. The results were validated by performing the immunohistochemistry on same samples with an ERα-antibody. We found that the two methods agree strongly in assay output (kappa value = 0.930, p-value <0.05 for strong ERα positive and the ERα negative samples; kappa value = 0.823, p-value <0.05 for the weak/moderate ER+ve samples, n = 20). Further, the aptamer stain the ERα-positive cells in breast tissues without cross-reacting to ERα-deficient fibroblasts, adipocytes, or the inflammatory cells. Our results demonstrate a significant consistency in the aptamer-assisted detection of ERα in strong ERα positive, moderate ERα positive and ERα negative breast cancer tissues. We anticipate that the ERaptD4 aptamer targeting ERα may potentially be used for an efficient grading of ERα expression in cancer tissues.     

Potential of doxorubicin as an antileishmanial agent.

Doxorubicin, a well characterized anticancer drug, was tested in vitro and in vivo for activity against Leishmania donovani. Activity in vitro was very high against both the promastigote and amastigote forms of this parasite with 50% effective dose (ED50) values on the order of 0.43 microM and 0.86 microM, respectively. An in vivo inhibition of spleen parasite burden up to 95% in an infected mouse model was achieved when a dosage of 625 micrograms doxorubicin/kg body weight/day was given in 4 consecutive doses, which is far less than the toxic dose. These results suggest that doxorubicin is highly active against visceral leishmaniasis and may be considered with second-line therapeutic agents such as amphotericin B and pentamidine.     

An ergonomic study on teenage girls working in the manual brick manufacturing units in the unorganized sectors in West Bengal, India

There are laws which protect and prevent the teenage girls from working. But still in the developing countries like India, Bangladesh, etc. teenage girls coming from low socio-economic background work, to add to the family income. There is dearth of data regarding the ergonomic studies of the teenage girls working in the manual brick manufacturing units of the unorganized sectors in India. Therefore, a field study was conducted in about four brick manufacturing units of West Bengal, India. Their job in these fields was to carry 8-10 unbaked and sun-dried bricks (49.11 +/- 2.13 Kg) from the brick field to the kiln-top and again carry the baked bricks back from the kiln to some other corner of the brick field, under the hot sun. Thus, this study aimed at evaluating their work load and their consequent work-related health effects. Results show that 86 % of them suffered from work-related pain in different body parts and there was an increase in their physiological and psychological parameters after work. It is concluded that the frequent use of awkward postures while at work (OWAS) and performing heavy to moderate types of tasks in the field are deteriorating their health conditions.     

Chromosome preparation from human bone marrow--technique modified for immediate clinical investigation.

A modified method for chromosome preparation from bone marrow aspirates of normal individuals and leukaemic patients is described for immediate clinical investigation. Hanks' balanced salt solution at different concentrations was used for in vitro incubation and hypotonic treatment. The method yields quite a high mitotic index with good metaphase spreads.
