A study was made of the effect of heparin on the capacity of the human blood lymphocytes to blast-transformation in response to the stimulation with antilymphocytic gamma-globulin and phytohemagglutinin. Heparin proved to inhibit more intensively the transformation of lymphocytes stimulated by the antilymphocytic gamma globulin. The depressive effect arose when heparin was injected one hour before the mitogen, simultaneously with it and one hour after it.  相似文献

A study of bacterial surface oligosaccharides were investigated among different strains of Neisseria gonorrhoeae to correlate structural features essential for binding to the MAb 2C7. This epitope is widely expressed and conserved in gonococcal isolates, characteristics essential to an effective candidate vaccine antigen. Sample lipooligosaccharides (LOS), was prepared by a modification of the hot phenol-water method from which de-O-acetylated LOS and oligosaccharide (OS) components were analyzed by ES-MS-CID-MS and ES-MSnin a triple quadrupole and an ion trap mass spectrometer, respectively. Previously documented natural heterogeneity was apparent from both LOS and OS preparations which was admixed with fragments induced by hydrazine and mild acid treatment. Natural heterogeneity was limited to phosphorylation and antenni extensions to the alpha-chain. Mild acid hydrolysis to release OS also hydrolyzed the beta(1-->6) glycosidic linkage of lipid A. OS structures were determined by collisional and resonance excitation combined with MS and multistep MSn which provided sequence information from both neutral loss, and nonreducing terminal fragments. A comparison of OS structures, with earlier knowledge of MAb binding, enzyme treatment, and partial acid hydrolysis indicates a generic overlapping domain for 2C7 binding. Reoccurring structural features include a Hepalpha(1-->3)Hepbeta(1-->5)KDO trisaccharide core branched on the nonreducing terminus (Hep-2) with an alpha(1-->2) linked GlcNAc (gamma-chain), and an alpha-linked lactose (beta-chain) residue. From the central heptose (Hep-1), a beta(1-->4) linked lactose (alpha-chain), moiety is required although extensions to this residue appear unnecessary.   相似文献

Four male mice (1-month-old) of BALB strain have been studied. Three types of cells participate in formation of cellular lining of the terminal branchioles: ciliated, Clara cells and villose cells which are included in the neuroepithelial bodies composition. Clara cells are numerous and they are similar in their ultrastructural organization, they contain a great number of polymorphous secretory granules, single mitochondria and a poorly developed endoplasmic reticulum. A comparatively large number of pores of Khone are revealed: there are 4-8 pores per one alveole, and one pore is situated on 197.4 mcm2 of the alveolar surface. Alveolocytes of the second type of make the main actively secreting cell population of the alveolar passages and alveoles. Simultaneously, at the state of secretion there are 76% of the cells. The secretion of alveolar surfactant is realized by means of merocrine exocytosis. Relative volumetric fraction of mitochondria in alveolocytes of the II type comprises 12% and that of "cytophospholiposomes" (osmiophilic laminar bodies)--11%.  相似文献

Experiments were carried out on linear mice immunized with sheep erythrocytes; it was found that the primary immune respose developed against the background of significant changes in the state of the sympathico-adrenal system, whose activity was determined by the dynamics of catecholamines in the blood and in the tissues of a number of organs, including the thymus, the spleen and the lymph nodes. By comparing the value of specific and neurohumoral indices it was revealed that the neurohumoral shifts preceded the maximal development of the immune response. On the example of studying the catecholamine dynamics the opinion on a close association between the state of the regulatory mechanisms and the effector formations responsible for the formation of specific immunological reactions was confirmed. It is suggested that a full-value immunological response developed on condition of activation of the sympathico-adrenal system.  相似文献

Nitric oxide (NO) released from the endothelium or from NO-donors is a powerful vasodilator. Its effect is mediated partly by vascular smooth muscle high conductance calcium-activated potassium (Kca) channels. Contradictory data exist as to whether NO activated the KCa channel directly or indirectly via protein kinase G (PKG). Thus the hypothesis that NO-donors can activate the KCa directly was investigated using the patch-clamp technique and freshly isolated smooth muscle cells from the rat tail artery. In inside-out experiments, the activity of KCa-channels was increased 1.61 +/- 0.20-fold (n = 10) by 10 microM SNP and 1.45 +/- 0.17-fold (n = 8) by 10 microM SNAP. However, the activity of KCa channels was also increased 1.46 +/- 0.20-fold (n = 8) by addition of the experimental bath solution. Thus these results suggest that NO released from NO-donors cannot activate KCa channel of the rat tail artery smooth muscle cells directly.  相似文献
