The nephridium of the dwarf male of Bonellia viridis was investigated by means of transmission electron microscopy. The nephridium proved to be of a distinct protonephridial type and not a metanephridium as maintained in the older literature. The nephridium is composed of a ciliated duct that projects into the coelom. Five crown cells at the end of the duct function as terminal filtration cells. Each crown cell has a bundle of about 20 cilia, surrounded by a labyrinthic weir of cell processes that are presumably involved in filtration. The ciliary bundles enter the nephridial duct through perforations of the adjacent tubule cells. This finding of a protonephridium in a minute, coelomate animal that lacks a circulatory system corroborates a recently formulated functional theory on the distribution of nephridial types.  相似文献

The ade6-M26 mutation of Schizosaccharomyces pombe increases conversion frequency in comparison with the nearby mutation ade6-M375. In order to investigate the effect of ade6-M26 on crossover frequency, heteroallelic ade6 duplications were constructed by integration of plasmids carrying the marker gene ura4. One ade6 gene carries either of the mutations M26 or M375 while the other ade6 copy carries the L469 mutation in both duplications. The duplication with ade6-M26 yields Ade(+) recombinants at significantly higher frequencies in meiosis, but not in mitosis. Tetrad analysis and physical characterization of spore clones from recombination tetrads demonstrate that conversions, unequal crossovers and intrachromatid exchanges occur at higher frequencies but with unaltered proportions among them. The conversion events show a pronounced bias when M26 is involved: they take place preferentially at the M26 allele. Thus the ade6-M26 mutation not only enhances conversion frequency as demonstrated before, but also crossover frequency. It displays the properties expected for a preferred site of initiation of general meiotic recombination. The duplications also yielded new information on ectopic recombination in S. pombe: ectopic crossovers occur in the duplications at much higher frequency than among naturally dispersed homologous sequences.  相似文献

Abstract The phylum Placozoa contains the most simply organized multicellular animals: they have a thick lower epithelium and a thin upper epithelium with some single cells sandwiched between these layers. Placozoans are generally considered to have no nerve cells or specialized sensory cells. However, the results of this investigation demonstrate the existence of a specific set of cells which react with antibodies against the neuropeptide family RFamide. The RFamide-positive cells of Trichoplax adhaerens are positioned at some distance along the margin of the animal.  相似文献

The point mutation M26 in the ade6 gene of Schizosaccharomyces pombe increases recombination frequency by an order of magnitude in comparison with other mutations in the same gene. The hypothesis is tested that this hot spot of recombination requires a specific nucleotide sequence at the M26 site. The DNA sequence is altered systematically by in vitro mutagenesis, and the resulting sequences are introduced into the ade6 gene in vivo by gene replacement. It results that any change of the heptanucleotide ATGACGT leads to loss of high frequency of recombination. Thus this oligonucleotide sequence is necessary for high frequency of recombination, but it seems not to be sufficient.  相似文献

Intergenic conversion is a mechanism for the concerted evolution of repeated DNA sequences. A new approach for the isolation of intergenic convertants of serine tRNA genes in the yeast Schizosaccharomyces pombe is described. Contrary to a previous scheme, the intergenic conversion events studied in this case need not result in functional tRNA genes. The procedure utilizes crosses of strains that are homozygous for an active UGA suppressor tRNA gene, and the resulting progeny spores are screened for loss of suppressor activity. In this way, intergenic convertants of a tRNA gene are identified that inherit varying stretches of DNA sequence from either of two other tRNA genes. The information transferred between genes includes anticodon and intron sequences. Two of the three tRNA genes involved in these information transfers are located on different chromosomes. The results indicate that intergenic conversion is a conservative process. No infidelity is observed in the nucleotide sequence transfers. This provides further evidence for the hypothesis that intergenic conversion and allelic conversion are the result of the same molecular mechanism. The screening procedure for intergenic revertants also yields spontaneous mutations that inactivate the suppressor tRNA gene. Point mutations and insertions of A occur at various sites at low frequency. In contrast, A insertions at one specific site occur with high frequency in each of the three tRNA genes. This new type of mutation hot spot is found also in vegetative cells.  相似文献

Trace element signatures of otolith edges and cores from 335 austral hake (Merluccius autralis) were analysed using LA-ICPMS from samples collected in Chilean and Falkland Islands'' waters, in order to provide potential insights into stock discrimination and migrations. Fish were caught in two locations in Chile and four locations in the south-west of the Falkland Islands Shelf. Univariate and multivariate analyses of trace element signatures in the edges of otoliths, representing adult fish, were not able to distinguish between samples collected in Chile and the Falkland Islands. Cluster analyses based on Ward’s similarity/distance metric suggested that it was possible to identify two groups from core signatures. Further analyses of this perceived clustering of the core concentrations revealed that this was largely due to the wide spread of Sr/Ca ratios in the otoliths’ cores. Gaussian finite mixtures using MCMC methods confirmed that Sr/Ca ratios form two separate distributions with significantly different mean values while concentrations of other elements showed no evidence of the presence of two or more distributions. The results suggest that there is only one spawning stock of austral hake with spawning situated in and around the Chilean fjords (43°30’S– 47°S) and the variation in Sr/Ca ratios likely suggests complex salinity structuring in this area.  相似文献

A newly compiled data set of nearly complete sequences of the large subunit of the nuclear ribosome (LSU or 28S) sampled from 31 diverse medusozoans greatly clarifies the phylogenetic history of Cnidaria. These data have substantial power to discern among many of the competing hypotheses of relationship derived from prior work. Moreover, LSU data provide strong support at key nodes that were equivocal based on other molecular markers. Combining LSU sequences with those of the small subunit of the nuclear ribosome (SSU or 18S), we present a detailed working hypothesis of medusozoan relationships and discuss character evolution within this diverse clade. Stauromedusae, comprising the benthic, so-called stalked jellyfish, appears to be the sister group of all other medusozoans, implying that the free-swimming medusa stage, the motor nerve net, and statocysts of ecto-endodermal origin are features derived within Medusozoa. Cubozoans, which have had uncertain phylogenetic affinities since the elucidation of their life cycles, form a clade-named Acraspeda-with the scyphozoan groups Coronatae, Rhizostomeae, and Semaeostomeae. The polyps of both cubozoans and hydrozoans appear to be secondarily simplified. Hydrozoa is comprised by two well-supported clades, Trachylina and Hydroidolina. The position of Limnomedusae within Trachylina indicates that the ancestral hydrozoan had a biphasic life cycle and that the medusa was formed via an entocodon. Recently hypothesized homologies between the entocodon and bilaterian mesoderm are therefore suspect. Laingiomedusae, which has often been viewed as a close ally of the trachyline group Narcomedusae, is instead shown to be unambiguously a member of Hydroidolina. The important model organisms of the Hydra species complex are part of a clade, Aplanulata, with other hydrozoans possessing direct development not involving a ciliated planula stage. Finally, applying phylogenetic mixture models to our data proved to be of little additional value over a more traditional phylogenetic approach involving explicit hypothesis testing and bootstrap analyses under multiple optimality criteria.
