The comparative effectiveness of the inhibitory influence of tetanic (100 Hz) stimulation of the hypothalamus, amygdala, and limbic cortex on electromyograph (EMG) response in m. digastricus produced by electrical stimulation of tooth pulp afferents was investigated in cats anesthetized by a mixture of chloralose and Nembutal. It was found that the most pronounced inhibition of nociceptive EMG response was produced by stimulating the medial and lateral structures of the hypothalamus, a less intense response by stimulating central and medial amygdaloid nuclei, and the least reaction by stimulation of different areas of the limbic cortex. It was shown that mechanisms underlying inhibition of the jaw-opening reflex produced by hypothalamic tetanic stimulation is unconnected with the concomitant increase in blood pressure. The inhibitory action of hypothalamic tetanic stimulation following blood pressure stabilization persists; this would suggest a primary but not baroafferent mechanism underlying inhibition of activity in trigeminal nucleus nociceptive neurons. Pronounced and protracted depression of jaw-opening reflex takes place following long-term pressor response produced by injecting noradrenaline intravenously. The same pronounced and protracted pressor response occurs under the effects of angiotensin but without any noticeable change in the amplitude of nociceptive EMG response. Parasympathetic discharges evoked by application of single and paired stimuli to central, corticomedial, and basolateral nuclei of the amygdala were recorded from the pelvic nerve (PN) in anesthetized and immobilized cats. The discharges were found to include three main negative waves (I, II, and III waves, appearing with the latencies of 16.0–17.8, 74–87, and 171–184 msec, respectively). The first two waves were seen most regularly. Sometimes the I wave was preceded by a super-early negative component of a low amplitude appearing with a latency of 8.5–10.7 msec. The discharges evoked by stimulation of various nuclei of the amygdala did not significantly differ from each other in their thresholds, time courses, and amplitudes, which suggests a relatively diffuse distribution of parasympathetic efferent structures in the amygdala. It was shown, using paired stimuli, that a response to the second stimulus needed a long-lasting period (a few tens of milliseconds) to be restored. The origin of the components of the amygdalofugal parasympathetic discharges recorded from the PN, as well as the corresponding pathways they spread through, are discussed. The effect of synthetic LVV-hemorphin-7 and hemorphin-7 on hypothalamo-pituitary-adrenocortical axis activity in response to endotoxin-induced stress was studied. The intraperitoneal (ip) endotoxin (lipopolysaccaride, LPS) (0.5 mg/kg) administration in combination with hemorphin (1 mg/kg) induce significant decrease in plasma corticosterone and modest decrease in plasma levels of tumor necrosis factor-alpha (TNFα) in compare with elevated levels of both corticosterone and TNFα in plasma of rats received LPS administration alone. Increased activity of calcineurin in both plasma and brain of rats received ip administration of LPS, was recovered under LPS + hemorphin treatment. In two independent proteome analysis, using 2-dimensional fluorescence difference gel electrophoresis and the isotope coded protein label technology, peptidyl-prolyl cis-trans-isomerase A (cyclophilin A) was identified as regulated by hemorphins protein in mouse brain. A therapeutic potential of hemorphins and mechanisms of their homeostatic action in response to endotoxin-induced stress are discussed.  相似文献

Antimicrobial peptides are promising alternative to traditional antibiotics and antitumor drugs for the battle against new antibiotic resistant bacteria strains and cancer maladies. The study of their structural and dynamics properties at physiological conditions can help to understand their stability, delivery mechanisms, and activity in the human body. In this article, we have used molecular dynamics simulations to study the effects of solvent environment, temperature, ions concentration, and peptide concentration on the structural properties of the antimicrobial hybrid peptide Cecropin A–Magainin 2. In TFE/water mixtures, the structure of the peptide retained α‐helix contents and an average hinge angle in close agreement with the experimental NMR and CD measurements reported in literature. Compared to the TFE/water mixture, the peptide simulated at the same ionic concentration lost most of its α‐helix structure. The increase of peptide concentration at both 300 and 310 K resulted in the peptide aggregation. The peptides in the complex retained the initial N‐ter α‐helix segment during all the simulation. The α‐helix stabilization is further enhanced in the high salt concentration simulations. The peptide aggregation was not observed in TFE/water mixture simulations and, the peptide aggregate, obtained from the water simulation, simulated in the same conditions did dissolve within few tens of nanoseconds. The results of this study provide insights at molecular level on the structural and dynamics properties of the CA‐MA peptide at physiological and membrane mimic conditions that can help to better understand its delivery and interaction with biological interfaces. © 2014 Wiley Periodicals, Inc. Main conclusion

Vavilovia formosa (Stev.) Fed. is a scientifically valuable common ancestor of the plant tribe Fabeae and also important in breeding and agronomy studies of the cultivated Fabeae, but it is close to extinction. A concerted academic and geovernmental effort is needed to save it.

Abstract

Since 2007, an informal international group of researchers on legumes has been working to increase awareness of Vavilovia formosa (Stev.) Fed., a relict and endangered wild-land relative to crop plant species. A majority of the modern botanical classifications place it within the tribe Fabeae, together with the genera vetchling (Lathyrus L.), lentil (Lens Mill.), pea (Pisum L.) and vetch (Vicia L.). V. formosa is encountered at altitudes from 1,500 m up to 3,500 m in Armenia, Azerbaijan, Georgia, Iran, Iraq, Lebanon, Russia, Syria and Turkey. This species may be of extraordinary importance for broadening current scientific knowledge on legume evolution and taxonomy because of its proximity to the hypothetical common ancestor of the tribe Fabeae, as well as for breeding and agronomy of the cultivated Fabeae species due to its perenniality and stress resistance. All this may be feasible only if a concerted and long-term conservation strategy is established and carried out by both academic and geovernmental authorities. The existing populations of V. formosa are in serious danger of extinction. The main threats are domestic and wild animal grazing, foraging, and early frosts in late summer. A long-term strategy to save V. formosa from extinction and to sustain its use in both basic and applied research comprises much improved in situ preservation, greater efforts for an ex situ conservation, and novel approaches of in vitro propagation.  相似文献

Vavilovia formosa is a relict, endangered species from the highlands of the Caucasus and the Near East. Described in 1812, it has had an uncertain status and was finally recognized as a separate genus of tribe Fabeae (Fabaceae). Our informal international group was established in 2007 to revive the interest in this species as it had been seriously neglected for decades. Here, we provide an overview of the accumulated knowledge on V. formosa and present the results of the most recent multidisciplinary research. Three expeditions were made to two locations in Armenia in 2009, providing the material for anatomical, morphological, chemical and molecular analysis. Unlike previous attempts, ex situ conservation in Yerevan and in vitro propagation, important for potential interspecific hybridization, were successful. Molecular tools were used to clarify the taxonomic position of V. formosa, often considered the closest to the extinct ancestor of the whole tribe. The analysis of four informative regions of plastid and nuclear DNA showed that V. formosa belongs to the same clade as Lathyrus and Pisum, with a distinct status. Preservation and maintenance of V. formosa remains the only basis for further development of all other scientific aspects, especially breeding and uses in agronomy. © 2013 The Linnean Society of London, Botanical Journal of the Linnean Society, 2013, 172 , 524–531.  相似文献

The properties of hypothalamic pathways activating sympathetic preganglionic neurons in the lateral horns of the spinal cord were studied in cats. This activation was shown to take place through reticulospinal sympathetic-activating neurons in the medulla. The possible zone of location of hypothalamic sympathetic-activating output neurons was shown by the scanning stimulation method to be in the posterolateral hypothalamus. Reticulospinal sympathetic-activating neurons in the medulla are probably excited monosynaptically by hypothalamo-sympathetic activating fibers projected on them.L. A. We studied evoked potentials (EP) and responses of single neurons in fields 24 and 25 of the cingulate gyrus of the cat brain cortex. The responses were elicited by stimulations of a parasympathetic, a sympathetic, and a somatic nerve (the pelvic, splanchnic, and sciatic nerves). The configuration and amplitude/temporal characteristics of both EP and neuronal responses showed that the organization of the visceral and somatic afferent inputs to the cortical fields under study is to a great extent similar. Localization of the foci of maximum activity for EP within field 24 and their overlapping, the higher responsiveness of the neurons of this field to afferent volleys as compared with that in field 25, the dominance of short-latency neuronal responses in field 24, and the presence of polysensory neurons with modally specific opposite responses in this field allow us to conclude that field 24 (compared with field 25) plays a more significant role in the perception and analysis of viscerosomatic afferent information. Taking into account earlier published data that the zones of concentration of the efferent neurons (forming a descending limbico-sympathetic discharge in the L2-L3 white connective branches and a limbico-parasympathetic discharge in the pelvic nerve) are localized within field 25 and overlap each other, we discuss the functional roles of fields 24 and 25 of the cingulate gyrus in viscerosensory and visceromotor control.  相似文献

It was discovered by making simultaneous recordings of evoked hypothalamic-parasympathetic and hypothalamic-sympathetic firing that sympathetic discharges in the splanchnic nerve and parasympathetic discharges in the pelvic nerve with minimum latency and lowest threshold level could be obtained by stimulating the posterolateral hypothalamus. It was also found that the focus of maximum neuronal activity produced by stimulating afferent visceral nerve fibers as well as the highest concentration of evoked efferent response in sympathetic and parasympathetic nerves were located in the same hypothalamic area. A working hypothesis was put forward that convergent polysensory hypothalamic neurons also act as divergent polyeffector constituents of the integrative system of the hypothalamovisceral reflex arc.L.
