The reversible hydrolysis of maltose to glucose by immobilized glucoamylase entrapped in spherical solid particles is studied theoretically. For this purpose a known kinetic model taking into account these reversible reactions and the competitive synthesis of iso-maltose was adopted. The mass transfer limitations in the bulk liquid and in the pores of the particles containing the enzyme are considered, using Fick's law. On the basis of mathematical modelling the optimum conditions for biocatalyst performance are established. An appropriate combination of particle size and initial substrate concentration may lead to reduction of undesirable mass transfer resistance and therefore product inhibition and to an improved selectivity of the biocatalyst with respect of glucose formation.List of Symbols

Three novel antimicrobial peptides designated ToAMP1, ToAMP2 and ToAMP3 were purified from Taraxacum officinale flowers. Their amino acid sequences were determined. The peptides are cationic and cysteine-rich and consist of 38, 44 and 42 amino acid residues for ToAMP1, ToAMP2 and ToAMP3, respectively. Importantly, according to cysteine motifs, the peptides are representatives of two novel previously unknown families of plant antimicrobial peptides. ToAMP1 and ToAMP2 share high sequence identity and belong to 6-Cys-containing antimicrobial peptides, while ToAMP3 is a member of a distinct 8-Cys family. The peptides were shown to display high antimicrobial activity both against fungal and bacterial pathogens, and therefore represent new promising molecules for biotechnological and medicinal applications. 相似文献

Catalase activity of the methanol-assimilating yeast Candida boidinii M-363 was determined cytochemically and biochemically. Electron microscopic investigations on ultrathin sections were made on cells from 16, 24, and 48h batch cultures in nutrient medium with methanol (or glucose as a control) as the sole source of carbon and energy. The electron-dense oxidation product of 3,3′-diaminobenzidine was found predominantly in the mitochondrial cristae and membranes. The mitochondria were increased in number, enlarged, sometimes aggregated, with variable form and size and they characteristically developed when the strain was grown on methanol. The significant development of these organelles and their intensive DAB staining correlated with the considerable increase in catalase activity. Biochemically, catalase in the cell-free extract was determined to be maximal along the exponential growth phase of the strain during its incubation on methanol. Enzyme analysis of the heavy mitochondrial fraction showed that it possessed catalase activity but not peroxidase activity. The results showed that not only peroxisomes but also mitochondria may be structurally and functionally responsible for the high catalase activity of some methanol-assimilating yeasts. What is more, the contribution of the mitochondria to the utilization of methanol may be significant. 相似文献

We tried to answer the question to what extent neurons in the tectum of the superior colliculus (SC) of the cat and in regions of the mesencephalic reticular formation (MRF) localized more ventrally are involved in the
control of movements of the limbs. The impulse activity of neurons of the above-mentioned structures was recorded in animals
performing targeted operated food-procuring movements by their forelimbs (pressing the pedal). As was found, neurons with
impulse reactions correlating with forelimb movements are rather numerous in the SC and adjacent MRF, and there are several
groups of neurons, whose impulse responses reached their maxima within different phases of the movements. These were neurons
with peaks of the discharge frequency coinciding with the target-reaching movement, with the moment of touching the pedal,
with pressing the pedal, and with the development of the muscle force counteracting the forced withdrawal of the limb toward
the initial position. Such specific patterns of the responses of different neurons of the SC and neighboring MRF are indicative
of a rather specific involvement of the above structures in the control of forelimb movements in cats.

Two strains (KM3 and KM5) of halophilic methylobacteria isolated from Red Sea algae do not require vitamin B12 for growth and can use methanol, methylamine, dimethylamine, trimethylamine, dimethyl sulfide, and fructose as sources of carbon and energy. The cells of these strains are gram-negative motile monotrichous (strain KM3) or peritrichous (strain KM5) rods. The strains are strictly aerobic and require Na+ ions but not growth factors. They are oxidase-and catalase-positive and reduce nitrates to nitrites. Both strains can grow in a temperature range of 4 to 37°C (with optimal growth at 29–34°C), at pH between 5.5 and 8.5 (with optimal growth at pH 7.5–8.0), and in a range of salt concentrations between 0.5 and 15% NaCl (with optimal growth at 5–9% NaCl). The phospholipids of these strains are dominated by phosphatidylethanolamine and phosphatidylglycerol and also include phosphatidylcholine, phosphatidylserine, and cardiolipin. The dominant fatty acids are C16:1ω7c and C16:0. The major ubiquinone is Q8. The cells accumulate ectoin, glutamate, and sucrose as intracellular osmoprotectants. The strains implement the 2-keto-3-deoxy-6-phosphogluconate-dependent variant of the ribulose monophosphate pathway. The G+C content of the DNA is 44.4–44.7 mol%. Analysis of the 16S rRNA genes showed that both strains belong to Gammaproteobacteria and have a high degree of homology (99.4%) to Methylophaga marina ATCC 35842T. Based on the data of polyphasic taxonomy, isolates KM3 and KM5 are identified as new strains M. marina KM3 (VKM B-2386) and M. marina KM5 (VKM B-2387). The ability of these strains to produce auxins (indole-3-acetic acid) suggests their metabolic association with marine algae. 相似文献

On the basis of the generalization and analysis of the results of bacteriological and immunological investigations the epidemic process of meningococcal infection (MI) in Mongolia was found to undergo definite changes during the last 20 years. Group A meningococci prevailing in the etiology of MI were replaced by strains belonging to group B affecting mainly young children (aged up to 3 years). MI morbidity rate caused by group B meningococci was found to be higher in Mongolia than in other countries of the world. These data substantiate the necessity of using more effective remedies for the control of this infection and, in particular, specific immunization with vaccines against group B meningococci; profound study of the properties of the circulating meningococcal strains is to be carried out. 相似文献
