An investigation of bacterial diversity in compost was performed using molecular chronometer in order to reveal its phylogeny. Thirty-three bacterial isolates isolated from compost were analyzed by 16S rRNA gene sequencing which revealed phylogenetic lineage of class Bacilli, γ, β-Proteobacteria, and Actinobacteria. Among these lineages, isolates belonging to class Bacilli consisted of species from genera Staphylococcus, Bacillus, Terribacillus, and Lysinibacillus. From phylum Actinobacteria: Microbacterium barkeri and Kocuria sp. were identified. Other bacterial groups had phylogenetic linkage with genera Comamonas and Acidovorax (class β-Proteobacteria); Serratia, Klebsiella, and Enterobacter (class γ-Proteobacteria). Similar isolates were analyzed through ARDRA. Amplified product of 16S rRNA gene from each isolates was subjected to cleavage by enzymes HpaII, HinfI, and MspI in separate reaction tubes. HpaII generated 2–6 bands ranging from 90–688 bp, HinfI generated 2–5 bands of 71–1,038 bp, and MspI 2–7 bands of 69–793 bp. The restriction patterns from HpaII, HinfI, and MspI were normalized separately and combined by means of pattern recognition software “Diversity Database.” HpaII had highest discrimination index (0.72) than HinfI (0.68) and MspI (0.65), and the combination of all three showed discrimination index (0.69). Numerical analysis of ARDRA patterns demonstrated sufficient phylogenetic information for characterizing bacterial diversity. Phylogenetic relationship obtained among isolates through ARDRA was compared with 16S rRNA gene sequence and ARDRA results showed sufficiently similar 16S rRNA gene sequence analysis, but not an overlapping. It has been observed that ARDRA technique facilitates the identification of bacteria in less than 36 h as compared to traditional 16S rRNA gene sequencing. 相似文献

Abstract. Natural regeneration of Pinus resinosa (red pine) seedlings around mature trees was studied in burned and unburned stands. Growth inhibitory effects of the forest organic matter on red pine seedlings was tested by a stair-step experiment using leachate of forest soil monoliths and also by a seed germination bio-assay using forest floor substrates. To test if higher burning temperatures can remove the allelopathic effects of red pine-Kalmia organic matter, a laboratory bio-assay was conducted by germinating red pine seeds on the organic matter burned at 200, 400, 600 and 800°C. Deposition of dry needles and a thick duff layer under red pine stands affected seedling establishment. Red pine seedling establishment increased with the decreasing thickness of duff layer away from the stump of the seed-bearing trees. Wildfire helped in removing the duff layer and increased seedling establishment. A high fuel load within a 0 - 1 m radius around the tree stump caused a deep burn of the organic matter including part of the soil seed reserve. On a burned-over surface, more seedlings established in a band between 1 and 2 m around the stump than inside and outside the band. Primary root growth of red pine was severely inhibited when the seedlings were grown in unburned forest floor organic matter where Kalmia was the principal understory species. Water leachate of a Pinus resinosa-Kalmia soil monolith was inhibitory to red pine seedling growth. In greenhouse conditions, the seedlings grew well in burned-over soil from a Pinus resinosa stand. Burned organic matter from a red pine forest showed an increase in pH with a burning temperature of 600°C. Primary root growth of red pine seedlings was similarly increased with increasing temperature up to 600°C; at higher temperatures the root length of seedlings did not increase any further. 相似文献

Shoot regeneration after prescribed burning or following the freezing temperatures of winter was monitored for nineteen heathland species present in an Arctostaphyleto-Callunetum community in northeast Scotland. Species whose renewal buds were near the surface of the ground started to grow earlier in the spring than species with renewal buds above the surface, but grouping species according to the position of their renewal bud (i.e. their life-form) did not account for all of the interspecific variation apparent. In the case of shoot regeneration after fire, species whose renewal buds were destroyed by fire because they were above-ground started to regenerate about the same time as species with belowground buds, protected from fire, but reached their maximum frequency of occurrence later. Grouping species by life-form was of limited value as a means of interpreting this interspecific variation in the timing of shoot regeneration after fire. It would be unwise to use plant life-form as the sole basis for interpreting or predicting a species' response to temperature stress when extreme temperatures occur regularly, as they do in heathland. The possible use of other plant traits to interpret and predict interspecific variation in the regeneration rate of heathland plants is discussed.Nomenclature follows Tutin et al. Isopongaglabol and 6-methoxyisopongaglabol, two new hydroxyfuranoflavones, together with two furanoflavones 5-methoxyfurano(8,7-4″,5″)flavone and 5-methoxy-3′,4′-methylenedioxyfurano(8,7-4″,5″)flavone, two simple flavones, desmethoxykanugin and fisetin tetramethyl ether, a chromenoflavanone, ovalichromene B, two triterpenes, cycloart-23-ene-3β,25-diol and friedelin, and β-sitosterol-β-d-glucoside were isolated from the petrol and CHCl3 extracts of the flowers of Pongamia glabra. On adsorption of some electron-acceptor molecules on the solid films of all-trans-beta-carotene, beta-apo-8'-carotenal, astacene and methylbixin a new absorption band appears on the longer-wavelength side of the spectrum in addition to the original bands. The position of this new band is dependent on the electron affinity (EA) of the acceptor molecules, and the intensity of this band increases with the amount of adsorbed acceptor molecules. A linear relationship between the vmax. of the new band and EA was observed. The value of the ionization potential of the polyenes estimated from such linear relationship agrees satisfactorily with the value obtained by other methods. It has been concluded that the polyenes behave as electron donor and first form molecular charge-transfer complexes (of type [polyene . I2] with iodine) with electron acceptors, these finally dissociating to yield ionic complexes (of type [polyene . I+] with iodine). 相似文献

Background
A disintegrin and metalloproteinase-12 (ADAM12) is a member of the greater ADAM family of enzymes: these are multifunctional, generally membrane-bound, zinc proteases for which there are forty genes known (21 of these appearing in humans). ADAM12 has been implicated in the pathogenesis of various cancers, liver fibrogenesis, hypertension, and asthma, and its elevation or decrease in human serum has been linked to these and other physiological/pathological conditions.Scope
In this review, we begin with a brief overview of the ADAM family of enzymes and protein structure. We then discuss the role of ADAM12 in the progression and/or diagnosis of various disease conditions, and we will conclude with an exploration of currently known natural and synthetic inhibitors.Major conclusion
ADAM12 has potential to emerge as a successful drug target, although targeting the metalloproteinase domain with any specificity will be difficult to achieve due to structural similarity between the members of the ADAM and MMP family of enzymes. Overall, more research is required to establish ADAM12 being as a highly desirable biomarker and drug target of different diseases, and their selective inhibitors as potential therapeutic agents.General significance
Given the appearance of elevated levels of ADAM12 in various diseases, particularly breast cancer, our understanding of this enzyme both as a biomarker and a potential drug target could help make significant inroads into both early diagnosis and treatment of disease. 相似文献7.
Fibrobacter succinogenes S85 grew rapidly on cellobiose (0.31 h−1) and the absolute rate of increase in fermentation acids was 0.68 h−1. Cultures that were provided with ball-milled cellulose initially produced fermentation acids and microbial protein as fast
as those provided with cellobiose, but the absolute cellulose digestion rate eventually declined. If the inoculum size was
increased, the kinetics decayed from first to zero order (with respect to cells) even sooner, but in each case the absolute
rate declined after only 20 to 30% of the cellulose had been fermented. Congo red binding indicated that the cellulose surface
area of individual cellulose particles was not decreasing, and the transition of ball-milled cellulose digestion corresponded
with the appearance of unbound cells in the culture supernatant. When bound cells from partially digested cellulose were removed
and the cellulose was re-incubated with a fresh inoculum, the initial absolute fermentation rate was as high as the one observed
for undigested cellulose and cellobiose. Based on these results, cellulose digestion by F. succinogenes S85 appears to be constrained by cellulose surface area rather than cellulase activity per se.

Post kala-azar dermal leishmaniasis (PKDL) is a rare disease. This is a solitary case report from Orissa, India. We describe a case of PKDL in a 55-year-old male who presented with multiple nodular lesions over face, trunk, and extremities. The patient had been to an endemic area of kala-azar and had a previous history of leishmaniasis. Fine needle aspiration cytology samples from skin nodules revealed Leishmania amastigotes. 相似文献

Several species of Lolium and other cool-season grasses (Poaceae subfamily Pooideae) tend to harbor symbiotic, seed-transmitted, fungi that enhance their fitness by various means. These fungal endophytes--species of Neotyphodium or Epichlo? (Clavicipitaceae)--are known for production of antiherbivore metabolites such as the bioprotective loline alkaloids. Lolines are saturated pyrrolizidines with an exo-1-amine and an ether bridge between C-2 and C-7. The ether bridge is an unusual feature for a biogenic compound in that it links two bridgehead carbon atoms. Much of the loline-biosynthetic pathway has been elucidated by administering isotopically labeled precursors to fungal cultures and by comparisons of loline biosynthesis genes to known gene families. The first step appears to be an unusual gamma-substitution reaction involving an enzyme related to O-acetylhomoserine (thiol) lyase, but which uses the secondary amine of L-proline rather than a sulfhydryl group as the nucleophile. The strained ether bridge is added after formation of the pyrrolizidine rings. Lolines with dimethylated or acylated 1-amines have insect antifeedant and insecticidal activities comparable to nicotine, but little or no toxicity to mammals. Considering the surprising abundance of lolines in some grass-endophyte symbiota, possible additional effects on plant stress tolerance and physiology are worth future consideration. In this review, we discuss the history of loline discovery, methods of analysis, biological activities and distribution in nature, as well as progress on the genetics and biochemistry of their biosynthesis, and on the chemical synthesis of these alkaloids. 相似文献

Hirsutella thompsonii Fisher is a host-specific fungal pathogen of the diverse group of arthropods known as the Acari. It particularly affects
eriophyid mites, and the identification of its isolates is a difficult task due to its pleomorphic nature. Seven isolates
of H. thompsonii var. thompsonii and H. thompsonii var. synnematosa collected from various agro-climatic regions of India were subjected to PCR analysis using random decamer primers to differentiate
between them. Random amplified polymorphic DNA analysis revealed that all of the isolates from the citrus rust mite, Phyllocoptruta oleivora Ashmead (Acari: Eriophyidae), clustered together, with the exception of one, HtCRMB. The only isolate of H. thompsonii from Aceria guerreronis keifer (Acari: Eriophydae), HtPDBC, clustered separately. 相似文献
