The sterol composition of seven strains of marine peridinioid dinoflagellates comprising the four known species of Heterocapsa Stein was examined by gas chromatography-mass spectrometry to determine the utility of these compounds in systematics. Cholest-5-en-3β-ol (cholesterol), 24-methyl-cholest-5-en-3β-ol (24-methylcholesterol), 4α,24(S)-dimethyl-5α-cholestan-3β-ol (4,24-dimethylcholestanol), 4α,23,24(R)-trimethyl-5α-cholest-22-en-3β-ol (dinosterol), 4α,23ξ,24ξ-trimethyl-5α-cholestan-3β-ol (dihydrodinosterol), and an unknown sterol were detected. Sterol composition does not vary significantly from species to species within the genus Heterocapsa and thus cannot be used for species differentiation. Sterols may, however, have value in defining the properties of dinoflagellate taxa above the family level. Over the course of the growth curve for Heterocapsa niei (Loeblich) Morrill & Loeblich 4,24-dimethylcholestanol and dinosterol covaried, suggesting that 4,24-dimethylcholestanol is converted into dinosterol by a previously proposed bioalkylation scheme. 相似文献

Transforming growth factor β (TGF-β) is a potent growth regulator and tumor suppressor in normal intestinal epithelium. Likewise, epithelial cell growth is controlled by rapid decay of growth-related mRNAs mediated through 3′ untranslated region (UTR) AU-rich element (ARE) motifs. We demonstrate that treatment of nontransformed intestinal epithelial cells with TGF-β inhibited ARE-mRNA expression. This effect of TGF-β was promoted through increased assembly of cytoplasmic RNA processing (P) bodies where ARE-mRNA localization was observed. P-body formation was dependent on TGF-β/Smad signaling, as Smad3 deletion abrogated P-body formation. In concert with increased P-body formation, TGF-β induced expression of the ARE-binding protein tristetraprolin (TTP), which colocalized to P bodies. TTP expression was necessary for TGF-β-dependent P-body formation and promoted growth inhibition by TGF-β. The significance of this was observed in vivo, where colonic epithelium deficient in TGF-β/Smad signaling or TTP expression showed attenuated P-body levels. These results provide new insight into TGF-β''s antiproliferative properties and identify TGF-β as a novel mRNA stability regulator in intestinal epithelium through its ability to promote TTP expression and subsequent P-body formation. 相似文献

The present study was undertaken to determine the mechanism by which phorbol ester stimulates eicosanoid synthesis in endothelial cells. We observed that phorbol 12-myristate 13-acetate (PMA) actively stimulated eicosanoid synthesis over a prolonged period of time, and the stimulatory effect was abolished by cycloheximide and actinomycin D. Western blot was employed to test the hypothesis that PMA elicited sustained eicosanoid synthesis via the stimulation of de novo synthesis of prostaglandin G/H synthase (cyclooxygenase, EC 1.14.99.1). Treatment of cultured human umbilical vein endothelial cells resulted in an enhancement of the 70-kDa immunoreactive prostaglandin G/H synthase band over the control cells treated with medium alone. The enhancement was abolished by cycloheximide. Human umbilical vein endothelial cells were then metabolically labeled with L-[35S]methionine, and the effect of PMA on methionine incorporation was evaluated by immunoblotting. PMA increased the synthetic rate of prostaglandin G/H synthase over the control cells. By pulse-chase experiments, we further showed that prostaglandin G/H synthase has a rapid turnover rate (t1/2 less than 10 min) in control cells, and PMA had no effect on the enzyme turnover. Our data indicate that PMA increases the synthesis of prostaglandin G/H synthase which is required for circumventing the autoinactivation of prostaglandin G/H synthase and hence permit sustained conversion of arachidonic acid into eicosanoids. 相似文献
