Corn (Zea mays L.) and sorghum (Sorghum vulgare, Pers.) plants were grown in a vermiculite-gravel mixture in controlled environment chambers until they were 40 days old. Water was withheld until they were severely wilted, and they were then rewatered. During drying and after rewatering stomatal resistance was measured with a diffusion porometer each morning, and water saturation deficit and water potential were measured on leaf samples. The average resistance of the lower epidermis of well watered plants was lower for corn than for sorghum. When water stress developed, the stomata began to close at a higher water potential in corn than in sorghum. The stomata of both species began to reopen normally soon after the wilted plants were rewatered, and on the 2nd day the leaf resistances were nearly as low as those of the controls. The average leaf water potential of well watered corn was −4.5 bars; that of sorghum, −6.4 bars. The lowest leaf water potential in stressed corn was −12.8 bars at a water saturation deficit of 45%. The lowest leaf water potential in stressed sorghum was −15.7 bars, but the water saturation deficit was only 29%. At these values the leaves of both species were tightly rolled or folded and some injury was apparent. Thus, although the average leaf resistance of corn is little lower than that of sorghum, corn loses much more of its water before the stomata are fully closed than does sorghum. The smaller reduction in water content of sorghum for a given reduction in leaf water potential is characteristic of drought-resistant species.  相似文献

There are several studies in the literature dealing with the effect of metal-humic complexes on plant metal uptake, but none of them correlate the physicochemical properties of the complexes with agronomic results. Our study covers both aspects under various experimental conditions. A humic extract (SHE) obtained from a sapric peat was selected for preparing the metal–humic complexes used in plant experiments. Fe–, Zn– and Cu–humic complexes with a reaction stoichiometry of 2:0.25 (humic:metal, w/w) were chosen after studying their stability and solubility with respect to pH (6–9) and the humic:metal reaction stoichiometry. Wheat and alfalfa plants were greenhouse cultured in pots containing one of three model soils: an acid, sandy soil and two alkaline, calcareous soils. Treatments were: control (no additions), SHE (53 mg kg–1 of SHE), and metal (Cu, Zn and Fe)–SHE complexes (2.5 and 5 mg kg–1 of metal rate and a SHE concentration to make 53 mg kg –1). Cu- and Zn–humic complexes significantly (p0.05) increased the plant uptake and the DTPA-extractable soil fraction of complexed micronutrients in most plant–soil systems. However, these effects were associated with significant increases (p0.05) of shoot and root dry weight only in alfalfa plants. In wheat, significant increases of root and shoot dry matter were only observed in the Cu–humic treated plants growing in the acid soil, where Cu deficiency was more intense. The Fe–humic complex did not increase Fe plant assimilation in any plant–soil system, but SHE increased Fe-uptake and/or DTPA-extractable soil Fe in the wheat–calcareous soil systems. These results, taken together with those obtained from the study of the pH- and SHE:metal ratio-dependent SHE complex solubility and stability, highlight the importance of the humic:Fe complex stoichiometry on iron bioavailability as a result of its influence on complex solubility.  相似文献

Polarisation of the actin cytoskeleton must cease during cytokinesis, to support efficient assembly and contraction of the actomyosin ring at the site of cell division, but the underlying mechanisms are still understood poorly in most species. In budding yeast, the Mitotic Exit Network (MEN) releases Cdc14 phosphatase from the nucleolus during anaphase, leading to the inactivation of mitotic forms of cyclin-dependent kinase (CDK) and the onset of septation, before G1-CDK can be reactivated and drive re-polarisation of the actin cytoskeleton to a new bud. Here, we show that premature inactivation of mitotic CDK, before release of Cdc14, allows G1-CDK to divert the actin cytoskeleton away from the actomyosin ring to a new site of polarised growth, thereby delaying progression through cytokinesis. Our data indicate that cells normally avoid this problem via the MEN-dependent release of Cdc14, which counteracts all classes of CDK-mediated phosphorylations during cytokinesis and blocks polarised growth. The dephosphorylation of CDK targets is therefore central to the mechanism by which the MEN and Cdc14 initiate cytokinesis and block polarised growth during late mitosis.  相似文献

Several carbon metabolism enzymes were measured in cultured cells and bacteroids of Rhizobium meliloti 102F51 and in alfalfa root nodule cytosol. The enzyme activity levels of the pentose phosphate pathway were much higher than those of the Embden-Meyerhof-Parnas or Entner-Doudoroff pathways in extracts of cultured cells. The pattern of enzyme activities in the bacteroids was different from that of cultured cells.  相似文献

Although responsiveness of N2-fixing plants to elevated CO2 conditions have been analyzed in previous studies, important uncertainties remain in relation to the effect enhanced CO2 in nodule proteomic profile and its implication in leaf responsiveness. The aim of our study was to deepen our understanding of the relationship between leaf and nodule metabolism of N2-fixing alfalfa plants after long-term exposure to elevated CO2. After 30-day exposure to elevated CO2, plants showed photosynthetic down-regulation with reductions in the light-saturated rate of CO2 assimilation (A sat) and the maximum rate of rubisco carboxylation (Vcmax). Under elevated CO2 conditions, the rubisco availability limited potential photosynthesis by around 12 %, which represented the majority of the observed fall in Vcmax. Photosynthetic down-regulation has been associated with decreased N availability even if those plants are capable to assimilate N2. Diminishment in shoot N demand (as reflected by the lower rubisco and leaf N content) suggests that the lower aboveground N requirements affected negatively nodule performance. In this condition, specific nodule activity was reduced due to an effect on nodule metabolism that manifested as a lower amount of nitrogenase reductase. Moreover, the nodule proteomic approach also revealed that nodule functioning was altered simultaneously in various enzyme quantity apart from nitrogenase. At elevated CO2, the tricarboxylic acid cycle was also altered with a reduced amount of isocitrate synthase protein. The nodule proteome analysis also revealed the relaxation of the antioxidant system as shown by a decline in the amount of catalase and isoflavone reductase protein.  相似文献

The plant fraction of alfalfa ( Medicago sativa L. cv. Aragon) nodules contained both nitrate reductase (NR) and nitrite reductase (NiR). Specific activity of NADH-NR from the cytosol of nodules not treated with NO3- was about 30 nmol (mg protein)-1-h-1 and was not basically affected by NO3 addition. In contrast, typical specific activity for cytosolic NiR was 1.5 umol (mg protein)-1h-1 using methyl viologen as electron donor. This activity strongly increased with NO3 concentration, probably due to substrate induction. Maximal activity was 3.5 μmol (mg protein)-1h-1 at 50 to 200 mM NO3.
Estimates indicate that the contribution of cytosol to the overall NR and NiR activities of alfalfa nodules is distinctly different: less than 10% and about 70%, respectively. The increasing amounts of NO2 accumulating in the cytosol upon NO3, supply, and the different response to NO3 of bacteroid and cytosolic NRs support the concept that most of this NO2 comes from the bacteroids.  相似文献

The evolution of Chlamydia trachomatis is mainly driven by recombination events. This fact can be fuelled by the coincidence in several European regions of the high prevalence of non-invasive urogenital genotypes and lymphogranuloma venereum (LGV) outbreaks. This scenario could modify the local epidemiology and favor the selection of new C. trachomatis variants. Quantifying the prevalence of co-infection could help to predict the potential risk in the selection of new variants with unpredictable results in pathogenesis or transmissibility. In the 2009-2013 period, 287 clinical samples with demonstrated presence of C. trachomatis were selected. They were divided in two groups. The first group was constituted by 137 samples with C. trachomatis of the LGV genotypes, and the second by the remaining 150 samples in which the presence of LGV genotypes was previously excluded. They were analyzed to detect the simultaneous presence of non-LGV genotypes based on pmpH and ompA genes. In the first group, co-infections were detected in 10.9% of the cases whereas in the second group the prevalence was 14.6%, which is the highest percentage ever described among European countries. Moreover, bioinformatic analyses suggested the presence among men who have sex with men of a pmpH-recombinant variant, similar to strains described in Seattle in 2002. This variant was the result of genetic exchange between genotypes belonging to LGV and members of G-genotype. Sequencing of other genes, phylogenetically related to pathotype, confirmed that the putative recombinant found in Madrid could have a common origin with the strains described in Seattle. Countries with a high prevalence of co-infections and high migration flows should enhance surveillance programs in at least their vulnerable population.  相似文献
