It is shown that non-covalent attachment of streptavidin, as well as of avidin, to biotinylated human erythrocytes induces homologous hemolysis by complement. Rabbit antiserum against human C3 is found to inhibit the lysis specifically as compared with non-immune rabbit serum. Efficiency of lysis inhibition is greater for avidin- and streptavidin-induced lysis of biotinylated human erythrocytes than for antibody-sensitized sheep erythrocytes. In contrast to positively charged avidin (pI 11), streptavidin is a neutral protein. Hence, hemolysis of streptavidin-carrying erythrocytes is inconsistent with the suggestion on the crucial role of avidin charge in lysis. Membrane alterations (cross-linking and clusterization of biotinylated components) induced by avidin (streptavidin) seem to be a more plausible explanation for the lysis. 相似文献

The activity of Ca(2+)-dependent neutral proteinases in rats under cobalt and mercury chloride injection was investigated. The calpains activity increase in the lungs, heart, liver and kidneys was revealed after 2 h cobalt chloride action. The mercury chloride gives a reliable increase of calcium-dependent neutral proteinases only in the kidneys. 相似文献

Dimerization of two copies of genomic RNA is a necessary step of retroviral replication. In the case of human immunodeficiency virus type 1 (HIV-1) the process is explored in many details. It is proved that conserved stem-loop structure is an essential element in RNA dimerization. Similar model of two-step dimerization mechanism can be considered for avian sarcoma and leukosis virus group (ASLV) in spite of the absence of homology between dimer initiation site (DIS) of ASLV and that of HIV-1. In this paper, short RNA fragments of two viruses: avian sarcoma virus CT-10 and avian leukosis virus HPRS-103 have been chosen in order to investigate the structural requirements of dimerization process and compare them to that of HIV-1. The rate of spontaneous transition from loose to tight dimer was studied as a function of stem length and temperature. Although both types of dimers were observed for both avian retroviruses chosen, fragments of CT-10 requires much higher RNA concentration to form loose dimer. In spite of identical sequence of the loops (5'-A-CUGCAG-3') avian sarcoma virus CT-10 RNA fragments dimerization was greatly impaired. The differences can be explained by deletion of adenine 271 in avian sarcoma virus CT-10 in the stem and by resulting shortening of the self-complementary loop. 相似文献

Results are presented from experimental studies of the implosion of wire (tungsten, aluminum, and molybdenum) and fiber (kapron) arrays at the Angara 5-1 facility at currents of up to 4 MA. The radial distributions of the azimuthal magnetic field in the stage of plasma production in such arrays are measured in detail for the first time, and the radial profiles of the current density are evaluated. The data obtained using an optical streak camera are compared with the results of measurements of the current and voltage. Using the obtained experimental results, the rate of plasma production in the arrays made of different materials is estimated. 相似文献

Prolonged plasma creation in heterogeneous liners, in which the liner substance is separated into two phase states (a hot plasma and a cold skeleton), is investigated both experimentally and theoretically. This situation is typical of multiwire, foam, and even gas liners in high-current high-voltage facilities. The main mechanisms governing the rate at which the plasma is created are investigated, and the simplest estimates of the creation rate are presented. It is found that, during prolonged plasma creation, the electric current flows through the entire cross section of the produced plasma shell, whose thickness is comparable with the liner radius; in other words, a current skin layer does not form. During compression, such a shell is fairly stable because of its relatively high resilience. It is shown that, under certain conditions, even a thick plasma shell can be highly compressed toward the discharge axis. A simplified numerical simulation of the compression of a plasma shell in a liner with prolonged plasma creation is employed in order to determine the conditions for achieving regimes of fairly compact and relatively stable radial compression of the shell. 相似文献

The rate of cell-cycle initiation was studied in a-cells of S. cerevisiae in the presence of the synthetic analogue of α-factor [N-Trp, Arg7]-α-factor (TA-αF). It was shown that TA-αF lowers the rate constant of cell-cycle initiation (or transition probability) for each separate cell. It was concluded on the basis of these results that the term ‘arrest’ in G1 by α-factor should be interpreted in a quantitative sense as the decrease in the probability of the emergence from ‘start’ per unit time and not as being equivalent to an ‘all-or-none’ response. The dependence of the rate constant of the cell-cycle initiation on the concentration of TA-αF can be described by the Hill equation where n = 1.01 + 0.05 and K = 14.5 ± 2.5 nM (±S.E.). It is demonstrated that after transfer of cells into the medium with a higher or a lower concentration of TA-αF, the rate constant of cell-cycle initiation changes abruptly from one value to another after a lag-period of 30 and 40 min respectively. This suggests a multistep mechanism of action for α-factor. The difference in the lag-periods allows us to suggest that α-factor exerts its action by two independent pathways. Since the Hill coefficient is practically equal to unit, no cooperative interactions are likely to be involved at least in one of these pathways. The inhibition of cell-cycle initiation can be used as a more adequate and sensitive test for biological activity of α-factor as compared to morphological measurements. 相似文献

Neurotoxic beta-amyloid peptide plays an important role in the pathology of Alzheimer’s disease. In aggregated form it binds to several proteins on the surface of the brain cells leading to their death. p75 receptor involved in supporting of cell balance is one of the targets for toxic beta-amyloid. We proposed that induction of antibodies against potential binding sites of p75 with beta-amyloid can be a promising approach towards development of new anti-Alzheimer’s disease treatment. Four potentially immunoactive fragments of p75 were chosen and chemically synthesized. Investigation of immunoprotective effect of the peptide fragments carried out in mice with experimentally induced form of Alzheimer’s disease helped to reveal two fragments effectively preserving murine memory from impairment. Results obtained by ELISA biochemical analysis showed that only immunization with fragment p75 155–164 led to significant decrease in beta-amyloid level in the brain of the experimental mice. Thus, immunization with both fragments of p75 receptor pro-vides a new insight into anti-Alzheimer’s disease drug design. 相似文献

One of the factors limiting the transmission of the electromagnetic pulse to the load in high-power electrophysical facilities is the current leakage in magnetically insulated transmission lines (MITLs). In this paper, the Angara-5-1 eight-module facility with an output power up to 6 TW is considered. The experimental and calculated time profiles of the leakage current for eight-module shots with a dynamic load (cylindrical arrays made of 40 tungsten wires) and single-module shots with a solid cylindrical metal load are compared. When interpreting the results, the contribution of vacuum electrons to the leakage current at the transition from the cylindrical to the conical section of the MITL is taken into account. 相似文献
