The two surfaces of a membrane can be compared by isolating sealed vesicles which bare one side or the other to impermeable probe molecules. For this purpose, inside-out (IO) and right-side-out (RO) vesicles have been generated from human red blood cell membrane ghosts and partially resolved on dextran density gradients1. We now report further characterization of this system, showing (a) that vesicles can be made which are impermeable to small solutes, including Na+ and K+, even though the parent ghosts are quite unsealed; (b) that the dextran gradient fractionation serves to separate sealed vesicles (which happened to be IO and RO, respectively, in the original study1); (c) that impermeable RO vesicles form instead of IO vesicles if the published protocol is modified by the untimely addition of trace divalent cations; and (d) that the vesicle fractions can and must be accurately monitored for sealing and orientation to avoid misinterpretations such as those encountered in a previous sidedness study2.  相似文献

In Arabidopsis thaliana , the regulation of hexose levels by the large monosaccharide transporter (MST) gene family influences many aspects of plant growth. The cloning and transgenic expression of one family member (STP13) enabled the manipulation of carbon (C) and nitrogen (N) metabolism in Arabidopsis . Transgenic seedlings constitutively over-expressing STP13 (STP13OX) had increased rates of glucose uptake, higher endogenous sucrose levels and accumulated more total C and biomass per plant when grown on soil-less media supplemented with 55 m m glucose and sufficient N (9 m m nitrate). Furthermore, STP13OX seedlings acquired 90% more total N than the Col-0 seedlings, and had higher levels of expression of the nitrate transporter NRT2.2 . In addition, STP13OX seedlings were larger and had higher biomass than Col-0 seedlings when grown under a limiting N condition (3 m m nitrate). Transgene analysis of STP13 reveals that its gene product is localized to the plasma membrane (PM) in tobacco BY-2 suspension cells, that it encodes a functional MST in planta , and that the STP13 promoter directs GUS expression to the vasculature and to leaf mesophyll cells. This work highlights the link between C and N metabolism, demonstrating that a plant's N use may be improved by increasing the availability of C.  相似文献

NC92 #284 is a transposon mutant of Bradyrhizobium sp. (Arachis)strain NC92 and has a host-specific fixation phenotype. It appearsto be ineffective on the host pigeonpea (90% reduction in shootN compared to that of the wild type), but partially effectiveon two other host plants, groundnut and siratro (50% and 20%reduction in shoot N compared to the wild type, respectively).To understand this phenomenon of host-specific fixation, thephysiological basis of the phenotypes was investigated. Host-dependentdifferences in symbiotic effectiveness were largely explainedby the degree to which nitrogenase activity was impaired inthe various #284 symbioses. Nodulation and the onset of nitrogenfixation were found to be delayed on all three hosts, but tothe greatest degree on pigeonpea. The specific activity of nitrogenaseper gram nodule was also reduced on all three hosts, again tothe greatest extent on pigeonpea. By contrast, the carbon costsand relative efficiencies of each symbiosis were similar tothe wild type. The results indicate that the host-specific fixationphenotype of #284 is reflected in a quantitative reduction inthe amount of N2 fixed. Thus the phenotypes reflect the differentability of the three host plants to tolerate or support the#284 mutation, rather than a defect in a specific interactionbetween #284 and a particular host plant. 1. The reproductive fitness of a parasitoid depends on its mating and ovipositing success. Virgin haplodiploid females can reproduce, but produce only males, and may diminish fitness by producing more male offspring than required. Therefore, females must decide on whether to mate or oviposit first. 2. This study was conducted to assess the mating versus ovipositing decision and its impact on the reproductive fitness of Diaeretiella rapae (Hymenoptera: Aphididae), an endoparasitoid of the cabbage aphid Brevicoryne brassicae (Hemiptera: Aphididae). 3. When newly emerged females were given a choice between mating and ovipositing, about 62% of D. rapae females preferred to mate before ovipositing. Those females who oviposited before mating parasitised only 10% of the available aphids. After mating, females superparasitised their hosts with fertilised eggs, which resulted in a highly female‐biased sex ratio in the offspring. 4. Mating success was very high (91%) in the presence of hosts (cabbage aphid nymphs) compared with that in the absence of aphids. However, mating success was not influenced by the quality (size) of the hosts present in the mating arena, despite a parasitoid preference for larger hosts during oviposition. The time between pairing and mating was also shorter in the presence of host aphids. The mean number of aphids parasitised and the parasitism rate were significantly greater after mating.  相似文献
