Honeydew and leaf extracts from Salix babylonica indicate that large quantities of cytokinin are present in the leaves and are transported through the phloem of this plant during late autumn. The active compound in the extracts could be hydrolysed with β-glucosidase, whereafter it showed the same chromatographic behaviour as zeatin. It is proposed that cytokinins in the leaves are converted to the glucoside and then redistributed to the rest of the plant organs where it is stored. 相似文献

In many studies, reviewed in this paper, eluates of separatedplant extracts and various known endogenous and exogenous substanceshave promoted or inhibited adventitious rooting when appliedto stem cuttings. Some of the promoters are thought to act inconcert with auxin (the co-factors) and include various phenolics. The mode of action of most rooting promoters and inhibitorsis obscure. There is no good evidence that rhizocaline, theoriginally postulated root-forming hormone, consists of an auxin-phenolicconjugate, and other explanations for the action of phenolicsare not well substantiated. It is suggested that the action of many rooting promoters andinhibitors is mediated by chemical injury. The soybean callus assay was used to study the effect of stratification on the cytokinin levels of the embryo dormant seed of Protea compacta R.Br. and the seed of Leucadendron dapbnoides Meisn., where dormancy is coat imposed. Chilling the seed for 30 days increased germination significantly, and resulted in a simultaneous increase in the butanol soluble cytokinins of both species. It would appear as if these compounds are either synthesized or released from a bound form in embryo dormant seed. In contrast, an interconversion from water soluble to butanol soluble cytokïnins appears to account for the increase where dormancy is coat imposed. The results also indicate that for germination to take place a threshold concentration of cytokinin may be required. It is suggested that the increase in butanol soluble cytokinins may lead to the breaking of dormancy, probably by increasing radicle elongation and/or cotyledon expansion. 相似文献

The soybean callus assay was used to study the effect of high oxygen tensions on the cytokinin levels of Leucadendron daphnoides Meisn. seed, where dormancy is apparently due to the restricting effect of the seed coat on oxygen diffusion to the embryo. High oxygen tensions led to a six-fold increase in germination compared to seed incubated in air and resulted in significant increases in butanol soluble cytokinins prior to visible germination. It is suggested that the primary effect of oxygen is to increase the rate of respiration and thus, to provide the energy required for the synthesis of butanol soluble cytokinins which leads to cotyledon expansion and subsequent radicle elongation. Present indications are that untreated seeds remain dormant due to low concentrations of butanol soluble cytokinins in their embryos. 相似文献

The effect of micronutrient supply on the growth and seed production of guayule (Parthenium argentatum Gray) was investigated. A deficiency of boron, but no other trace element, significantly reduced the growth, seed production and the percentage of seeds that germinated. The optimum concentration of B for seed production was between 20 and 100 μM. Results indicate that guayule may benefit from B fertilisation. 相似文献

Nodal explants of Kalanchöe blossfeldiana Poellniz. werecultured in vitro on a low nutrient hormone-free medium. Floweringwas achieved in response to short-day inductive cycles. Thissystem was used to test the influence, on the flowering response,of a variety of culture conditions and media. Reduced vesseland medium volume both inhibited flowering, as did renderingthe vessel impervious to gasses. Nitrogen in the form of NH4NO3and KNO3 promoted flowering and vegetative growth in differentways. Increasing sucrose content in the medium caused some increasein the flowering response and in leaf anthocyanin production,but inhibited most aspects of vegetative growth. The growth of cytokinin-dependent soya bean callus has beenshown to be accelerated by adding N-(purin-6-yl)glycine to themedium. Two biologically active peaks were detected when thecallus was cultured with N-(purin-6-yl)glycine. These two peaksco-chromatographed with 6-(2, 3, 4-trihydroxy-3-methylbutylamino)purineand zeatin respectively. When 14C labelled N-(purin-6-yl)glycinewas applied to the callus, radioactivity was found with boththese compounds irrespective of whether or not the N-(purin-6-yl)glycinewas labelled in the side chain or in the 8-position of the purinering. Small amounts of zeatin appear to be produced from N-(purin-6-yl)glycinewhich could explain why this compound stimulates the divisionof soya bean callus. Zeatin and zeatinriboside were identified from the milk of mature Cocos nucifera fruits. It would appear as if the two compounds are present in roughly equal proportions. Ethyl acetate extraction in a liquid-liquid extractor at an alkaline pH (8.0) proved to be a very efficient method of extracting zeatinriboside. Partitioning with water-saturated n-butanol proved to be the best way of extracting zeatin. 相似文献

Using the soybean callus bioassay it has been shown that dormant lettuce seeds (Lactuca sativa L. cv. Grand Rapids) contain large amounts of water soluble cytokinins and small amounts of butanol soluble ones. When the seeds are irradiated with red light, or imbibed with 5 mg/1 gibberellic acid in the dark, the total cytokinin content of the seeds decreases, the level of water soluble cytokinins decreases, and the level of the butanol soluble cytokinins increases. Far-red light does not reverse this effect completely although cytokinin activity in the butanol extracts decreases following such irradiation. It is proposed that the interconversion of cytokinins initiated by red light, or gibberellic acid in the dark, is one of the primary events leading to radicle elongation in light-sensitive lettuce seed. 相似文献
