In this paper we demonstrate the study of plant water balanceby the non-invasive measurement of tissue water content andwater flow using proton nuclear magnetic resonance (NMR). Sapvelocity and flux were measured independently in the presenceof an excess of stationary tissue water. The instrumentationdescribed allows automated and unattended measurement of flow-and water content-variables in a well-defined region of theplant over periods of several days, with a time resolution betweensuccessive measurements of c. 5 s. Using this apparatus theeffect of changes in light intensity (day/night rhythm) andrelative humidity on stem tissue water content as well as onthe velocity and flux of xylem sap in the stem were investigatedin a cucumber plant. The results are in agreement with predictionsfrom a simple model for plant water balance, which is basedon water potential, flow rate and resistance to flow. As longas only transpiration is varied, flow rate and water content(or potential) are affected in opposite ways as demonstratedin this paper. In contrast, the model predicts that changesin uptake (resulting from changes in, for example, root resistance)will induce changes in water content and flow in the same direction.An experimental verification of this prediction is given ina subsequent paper, where, in addition, the NMR results arecompared to those obtained with a dendrometer. Evidence of the mechanical advantage of subsidiary cells wasobtained by simultaneous measurements of turgor pressure potentialsin adjacent subsidiary and guard cells using injection circuitswith two separate needles. In Tradescantia virginiana the mechanicaladvantage approaches two. Using the same technique evidencewas obtained that the Spannungsphase is, in the first place,a turgor relations phenomenon due to the mechanical advantageof epidermal or subsidiary cells. In addition, the evidenceindicated that the elastic properties of guard cell walls mayundergo changes during the Spannungsphase when potassium iontransport commences. During these measurements it was confirmedthat the optimum leaf water deficit for maximum stomatal openingoccurs when the epidermal turgor is near zero. Under these conditionsthe width of the stomatal pore is a function of the turgor pressureof the guard cells, since at zero turgor of the subsidiary cellstheir mechanical advantage has disappeared.  相似文献

Abstract. Experiments were carried out on Pinus radiata (D. Don) trees grown as cuttings from clonal parent stock. Some of these trees were about 0.4 m high while others were about 5 m high; all were grown in containers. The stem diameters at the tops and at the bottoms of the large trees, rates of photosynthesis, and needle water potentials were measured both when the trees were well watered and as they dehydrated after water was withheld. The water potentials of well-watered plants was highest in the small trees and lowest at the top of the large trees. When water was withheld, photosynthesis was in most cases unaffected by a small reduction in water potential, but the rate of photosynthesis fell as water potentials declined further. The stems of the large trees expanded at a constant rate when the trees were well watered and for part of the dehydration period, while subsequent stem shrinkage and the fall in photosynthesis both occurred at approximately the same time.
Water potentials increased little in the 24 h after rewatering, and significant rates of photosynthesis were not measured until 2 or 3 d later while renewed stem expansion was not measured until 2 d after rewatering.
Water deficits reduced the lumen diameter of newly matured stem tracheids, but increased the thickness of their walls. After 1 month of water potentials of about −2.4 MPa, tracheid lumen diameter and wall thickness were both much reduced, and this reduction continued in tracheids maturing shortly after rewatering.  相似文献

Factors connected with the formation of water droplets in leavesby distillation from the mesophyll to the epidermis were investigatedin a number of species. It was concluded that in illuminatedleaves water droplets form principally on the inner walls ofguard and subsidiary cells, and sometimes below the anticlinalwalls of epidermal cells, because these sites are cooler thanthe rest of the leaf. Under more isothermal conditions any waterdroplets that had formed disappeared. With increasing waterstress water droplets did not form so readily, though distillationwas occurring. Few water droplets were observed in leaves outof doors that had open stomata. Significant temperature gradientswere measured across leaves with thermocouples, but these werelarger than were gradients calculated from measured thermalconductivities of leaves. The evaporation resistances of theinner walls of the epidermis and of the mesophyll were foundto be similar. This led to the conclusion that the hydrophobicityof the surfaces of these tissues is similar. Abstract Baited and unbaited hoop-nets commonly are used to capture catfish in lotic and lentic systems. Turtle bycatch and post-capture mortality has been problematic during catfish surveys in Missouri, USA, most recently in the Gasconade River, Gasconade and Osage counties. We evaluated 3 modified hoop-net designs that would reduce turtle bycatch without reducing catfish capture in the Gasconade River during 15 May-15 July 2006 after pilot study evaluation of 5 hoop-net designs in April 2006. We deployed modified and control-nets in blocks for 48 hours to evaluate differences in turtle and catfish catch rate, as well as abundance, size, and mortality rate of turtle bycatch. The chimney design reduced turtle bycatch by 84% when compared to the control, without decreasing the number or average size of captured flathead catfish (Pylodictis olivaris). Environmental conditions that affected turtle mortality included Secchi disc transparency, temperature, dissolved oxygen, and stream river depth. This is the first known attempt to create turtle exclusion or escapement devices for hoop-nets deployed in freshwater systems. Biologists using hoop-nets to sample aquatic vertebrates in moderate to large river systems will benefit from our study. The application of this methodology will reduce turtle bycatch mortality, especially when sampling is conducted in high water temperatures.  相似文献

The hydraulic system in leaves was investigated in some detailand analysed in terms of an electrical transmission line analogy.The equivalent ‘circuit elements’ in the leaf wereidentified and their values calculated. The values of the ‘capacitative’and ‘inductive’ terms were calculated for variousleaf water potentials and for various changes in potential.They both exhibited considerable non-linearity. The calculatedvalues were used to derive values for the ‘impedance’of the pathways through the leaf xylem, and through the mesophyllcell walls, and to investigate the characteristics of thesetwo pathways when oscillations, or fluctuations, in leaf waterstatus were occurring.  相似文献

A new electronic system for effecting feedback control of temperaturefrom 3 to 50 °C and humidity from approximately 15 to 99per cent is described. This system uses thermistor sensors tocontrol, through electronic control units, the temperature ofwater baths which determined the temperature and humidity ofthe airstream passing through leaf or small plant chambers.The control has been found to be precise, and the temperatureand humidity can each be reset from a separate single controlknob.  相似文献

The apparatuscaused minimal disturbance to, and distortion of, the stem beingmeasured. A magnetohydrodynamic sap flux meter with a mains energisedelectromagnet is described, and its calibration discussed. Thewhole apparatus was designed to operate from the mains supply,to obviate the need for batteries when working in the laboratory.The calibration curve for the instrument was not that expectedfor a single pipe. If the stem was treated as a large numberof pipes in parallel a theoretical curve was obtained that wasvery similar to the empirical one.  相似文献
