黑曲霉Aspergillus niger因能够产生大量的木质纤维素降解酶而在木质纤维素资源利用中发挥重要作用。目前,有关黑曲霉基因组中与木质纤维素降解相关的基因是否存在可变剪接的情况尚不清楚。本研究以黑曲霉CBS513.88菌株为研究对象,采用rMATS和ABLas两种方法对黑曲霉在葡萄糖为唯一碳源(G组)和小麦秸秆为唯一碳源(WS组)下的56个木质纤维素降解酶基因的可变剪接事件进行分析,并通过RT-PCR扩增和内含子特异性扩增对3个典型基因的可变剪接体进行了验证。结果表明,ABLas可变剪接分析算法相较于rMATS分析算法更为准确,ABLas分析算法显示G组和WS组共有21个木质纤维素降解酶基因出现了可变剪接,可变剪接类型以内含子保留(IR)为主,占所有可变剪接事件的82.85%。另外,G组和WS组发生可变剪接的木质纤维素降解酶基因也有所不同:G组发生可变剪接的基因为13个,WS组发生可变剪接的基因为14个,两组都发生可变剪接的基因为6个,这表明黑曲霉木质纤维素降解酶基因的可变剪接在不同生长条件下存在差异,另一方面,黑曲霉中众多可变剪接体的存在也为开发新型的木质纤维素降解酶资源提供基础。  相似文献

The effects of salinity on growth, water relations, glycinebetainecontent, and ion accumulation in the perennial halophyte Atriplexgriffithii var. stocksii were determined. The following questionswere addressed: (1) What effect does salinity have on growthresponses at different ages? (2) Is A. griffithii an ion accumulator?(3) Does A. griffithii accumulate glycinebetaine in responseto salinity? Atriplex griffithii plants were grown in pots at0, 90, 180 and 360  m M NaCl in sand culture in a plantgrowth chamber and plants were harvested after 30, 60 and 90d. Plant total dry weight was significantly inhibited at 360m M NaCl. Root growth showed a substantial promotion at 90 mM NaCl. The water potential and osmotic potential of shootsbecame more negative with increasing salinity and time of growth.The Na+and Cl-content in both shoots and roots increased withincreases in salinity. Increased treatment levels of NaCl induceddecreases in Ca+, K+and Mg2+in plants. Atriplex griffithii accumulateda large quantity of ions, with the ash content reaching 39%of the dry weight in leaves. Inorganic ion accumulation is significantin osmotic adjustment and facilitates water uptake along a soil-plantgradient. Glycinebetaine concentration was low in roots, andin stems it increased with increases in salinity. AJMAL, S. & IQBAL, M., 1992. Structure of the vascular cambium of varying age and its derivative tissues in the stem of Ficus rumphii Blume. The cambial cells of Ficus rumphii and their derivatives vary in size and number with the age of the tree. The fusiform initials, vessel elements, sieve-tube members, xylem fibres and phloem fibres are relatively short in the terminal part of the stem axis, consistently longer down the stem reaching a maximum, and short again in the main trunk. The width of the respective cells shows a similar variation. The ray cell initials increase both in number and size, and form rays of varying dimensions. Uniseriate and biseriate cambial rays are abundant, while multiseriate rays are especially sparse in the young shoot. The proportion of uni- and biseriate rays falls in the lower part of the stem whereas that of multiseriate rays increases. Likewise, short cambial rays are abundant in young branches but their frequency decreases in the trunk region, leading to a rise of the tall ray population. The rays in the phloem and xylem regions reflect a similar variation pattern with certain fluctuations. The overall proportion of the rays increases in the older part of the axis, reaching a maximum tangential area in the trunk. The transectional area of vessel elements remains more or less constant down to the base after an initial increase in the terminal region, while the sieve-tube proportion tends to be constant in the basal region after a consistent gain from the tree top basewards.  相似文献

The stem succulent perennial halophyteHaloxylon recurvum Bungeex. Boiss. grows and produces seed under highly saline conditionsand seeds are deposited in saline soils. Experiments were conductedto determine the effect of salinity and temperature on the germinationof seeds. Results indicate that seeds can germinate at veryhigh salt concentrations (500 mM). However, highest germinationpercentages were obtained in distilled water. Cooler thermoperiodspromoted germination, while high temperatures significantlyinhibited the germination of seeds at all NaCl concentrationstested. Rate of germination decreased with increases in salinity.At higher thermoperiods the rate of germination was significantlylower in comparison to lower thermoperiods. Seeds recover afterbeing transferred to distilled water and recovery was higherfrom higher salinity concentrations and lower thermoperiods.Final recovery germination percentages in high salt treatmentswere significantly lower than non-saline controls, indicatingthat exposure to high concentration of NaCl permanently inhibitedgermination. Cambial structure and activity of Ficus rumphii Blume vary withthe changes in local climate. The cambial cells start swellingearly in April prior to the onset of periclinal divisions whichare most frequent in August. Cell division stops in October.During the growth season, initiation as well as cessation ofthe phloem production precedes that of xylem. A moderately hightemperature is correlated with the cambial reactivation. Onceinitiated, the activity continues at relatively low temperatures.Hot and dry environment favours the phloem production, whereashot and moderately humid conditions induce xylogenesis. Thesize and relative proportion of cambial initials also changewith season. Fusiform initials are shorter and broader duringthe rainy season (July–September) than for the rest ofthe year. Zygophyllum simplexL. is a succulent annual that grows on thecoastal and inland saline flats around Karachi, Pakistan. Theseeds are moderately salt tolerant during germination. GerminationofZygophyllum simplexseeds under various salinity, proline,betaine, GA and kinetin treatments was determined. Proline (0.1and 1 mM) and betaine (0.1 and 1 mM) alleviated the innate dormancyof seeds, and germination reached 60–70% compared to 12%in the control set. At low salinity compatible osmotica alleviatedsome effects of salinity, but at higher NaCl concentrationsboth proline and betaine were ineffective. Gibberellic acid(0.3 and 3 mM) and kinetin (0.05 and 0.5 mM) substantially alleviatedboth innate as well as salinity-induced seed dormancy. At highersalinity (125 mM), low concentrations of kinetin (0.05 mM) andhigh concentrations of GA (3 mM) were more effective. GA completelyalleviated the effect of salinity at all concentrations used.
