In this study, acetone extracts and acidic extracts were prepared from skin mucus, gill, kidney, liver and spleen of the Japanese eel, Anguilla japonica, and they exhibited different levels of antibacterial activities against three strains of Gram-negative bacteria, Edwardsiella tarda, Aeromonas hydrophila, Aeromonas sp. and one Gram-positive bacterium Micrococcus leteus. The mucus was chosen as the source of antibacterial peptide for further purification of antibacterial peptides. Following the intraperitoneal injection of A. hydrophila, one of the main pathogenic bacteria of Japanese eel and many other fish, a peptide was purified from acetic acid extraction of the skin mucus, by using cationic exchange liquid chromatography and reverse-phase high-performance liquid chromatography (RP-HPLC). The isolated antibacterial peptide, named as AJN-10, exhibited antibacterial activity against A. hydrophila. The AJN-10 is a heat-tolerant and hydrophilic peptide. The molecular weight of this peptide is 6,044.28 Da, as determined by matrix-assisted laser desorption ionisation time of flight mass spectrometry. The 20 N-terminal amino acid sequences were clarified by Edman degradation, and based on results of homology search by BLAST analysis of the 20 N-terminal sequences, the AJN-10 showed little similarity to other proteins in databases.  相似文献

探究从油樟(Cinnamomum longipaniculatum)叶中提取出的不同极性黄酮样品对大肠杆菌(Escherichia coli)、志贺杆菌(Shigella)、金黄色葡萄球菌(Staphylococcus aureus)、稻瘟病菌(Magnaporthe grisea)和黄曲霉菌(Aspergillus flavus)五种病原菌的抑制作用。以五种病原菌为指示菌,将不同的黄酮样品滴加在滤纸片上,观察抑菌圈的大小。结果显示,样品Fr1、Fr2、Fr3、Fr4和Fr7对大肠杆菌有抑菌作用;样品Fr2、Fr3、Fr4、Fr5、Fr7和Fr8对志贺杆菌有抑菌作用;样品Fr4和Fr8对金黄色葡萄球菌有抑菌作用;样品过孔和Fr4对黄曲霉菌有抑菌作用;样品过孔对稻瘟病菌有抑菌作用。故不同极性的黄酮样品有良好的抑菌性。  相似文献

植物内生菌关系到植物的健康生长,且与植物产生的相关功效产物紧密关联。以我国四川宜宾油樟叶片分离筛选得到的内生细菌作为研究对象,利用传统微生物培养方法,基于形态学观察,16S rRNA和gyrA基因系统发育分析的多相分类学方法,将其鉴定为贝莱斯芽胞杆菌(Bacillus velezensis);并通过对从油樟叶片中分离的油樟真菌YZP-01以及常见植物病原真菌绳状篮状菌(Talaromyces funiculosus)、拟轮枝镰孢菌(Fusarium verticillioides)、草酸青霉(Penicillium oxalicum)四种致病菌株进行了拮抗试验,发现了该菌株具有广谱且良好的拮抗病原菌活性。使用特定引物扩增功能基因(bioA,bmyB,ituC,fenD,srfAA,srfAB,yngG和yndJ),并成功从菌株YZ12中扩增出这些基因。  相似文献

Background

More and more HIV therapeutic vaccines will enter clinical trials; however, little is known about the willingness to participate (WTP) in HIV therapeutic vaccine trials among HIV-positive individuals.

Objective

To investigate the WTP in HIV therapeutic vaccine trials among Chinese HIV-infected patients.

Methods

We conducted a cross-sectional survey on HIV-positive inpatients and outpatients at Shanghai Public Health Center. A total of 447 participants were recruited into this study. Following an introduction with general information on HIV therapeutic vaccine and its potential effectiveness and side effects, each participant completed a questionnaire in a self-administered form. The questionnaires covered demographics, high-risk behaviors, clinical characteristics and willingness to participate in HIV therapeutic vaccine trial.

Results

The overall willingness to participate in HIV therapeutic vaccine trials was 91.5%. Interestingly, multivariate logistic regression analyses demonstrated that the willingness was higher for those sexually infected by HIV (odds ratio [OR]: 4.36; 95% confidence interval [CI]: 1.53–12.41), diagnosed as HIV-1 infection for greater than 5 years (OR: 7.12, 95% CI: 1.83–27.76), and with the presence of infectious complications (OR: 2.75; 95% CI: 1.02–7.45). The primary reason for participation was to delay or reduce antiretroviral treatment (ART) and to avoid ART side effects (76.6%), and then followed by delaying disease progression (74.9%), increasing immune response to suppress opportunistic infections (57.7%) and preventing the development of drug resistance (37.1%). Reasons for unwillingness to participate mainly included concern for safety (37.0%), lack of knowledge on therapeutic vaccine (33.3%), and satisfaction with ART effectiveness (22.2%).

Conclusions

The WTP in HIV therapeutic vaccine trials was high among HIV-infected Chinese patients. HIV+ subjects who acquired infection through sexual contact and who were diagnosed for more than 5 years may represent a good candidate population for enrollment in therapeutic vaccine trials.  相似文献

溶磷菌和固氮菌溶解磷矿粉时的互作效应   总被引:13,自引:0,他引:13  
采用4株溶磷菌(Lx81、Dm84、Jm92、Lx191)、和3株固氮菌(ChW5、ChW6、ChO6)单独和混合接种后测定培养液有效磷含量、pH值及总有机酸含量的方法,研究溶磷菌和固氮菌溶解磷矿粉时的互作效应。结果表明,相对于单独接种溶磷菌:Lx81与3株固氮菌分别混合培养能提高磷矿粉的溶解能力,4株溶磷菌与ChW6,Lx81、Dm84、Lx191与Ch06分别混合培养及Jm92+ChW5组合溶磷量极显著增加(P〈0.01);Dm84+ChW5、Lxl91+ChW5、Jm92+Ch06组合的溶磷量下降(P〈0.01)。除Lx81+ChW6、Lx81+Ch06培养液pH值降低外,混合培养的其它组合培养液pH值均较单独接种溶磷菌时升高。有机酸测定结果表明,Lx81、Jm92与ChW5、Ch06分别混合培养、ChW6+Lx81组合有机酸含量升高(P〈0.01),其它7种组合的有机酸含量均较单独接种溶磷菌的值下降(P〈0.01)。溶磷菌和固氮菌单菌培养时溶磷量与pH值、溶磷量与总有机酸含量及pH值与总有机酸含量之间呈现线性相关;Dm84、Lx191与3株固氮菌分别混合培养溶磷量与pH值之间、Lx81与3株固氮菌分别混合培养溶磷量与总有机酸含量之间呈现线性相关,其它组合的溶磷量与pH值、总有机酸含量间没有相关性。溶磷菌和固氮菌混合培养对溶解磷矿粉既有协同作用也有拮抗作用。  相似文献

Age and growth of Anguilla japonica sampled from the Qiulu River in southern China were studied. Mean body lengths and weights of the eels at each age were back-calculated from their otoliths. The growth parameters for the von Bertalanffy function gave L = 98–2cm, K =0·07 and t 0=− 1·94. Scales were observed to form firstly at the middle part of the lateral line, at a minimum body length of 160 mm and were inappropriate for ageing.  相似文献

鳗鲡病原性维氏气单胞菌的分离与鉴定   总被引:6,自引:0,他引:6  
从养殖患病的日本鳗鲡(Anguilla japonica)肾脏分离得到1株优势菌株,形态特征和生理生化测试结果显示,分离菌在4%羊血平板培养基上呈α型溶血,革兰氏阴性,有运动性,氧化酶阳性,在pH6.0和1%NaCl中生长等。结合Biolog自动微生物鉴定系统和16S rRNA、gyrB基因序列分析结果,确定该分离菌为维氏气单胞菌(Aeromonas veronii)。人工腹腔注射感染鳗鲡的试验结果表明,分离菌对鳗鲡的半数致死量LD50为2.15×107CFU/mL。药敏试验结果显示,在13种抗菌类药物中,分离菌对左氧氟沙星、氟哌酸、阿奇霉素和萘啶酸等8种药物敏感,而对利福平和新生霉素等5种药物敏感度低。  相似文献

To develop a vaccine, which can simultaneously prevent the diseases caused by various pathogenic bacteria in fish, we try to find a conserved outer membrane protein (OMP) antigen from different bacterial pathogens. In this study, an OMP fragment of 747 bp (named as Omp-G), which was highly conserved in seven Aeromonas OMP sequences from the NCBI database, was amplified by PCR from one Aeromonas sobria strain (B10) and two Aeromonas hydrophila strains (B27 and B33) with the designed specific primers. The sequence was cloned into pGEX-2T (6 × His-tag) vector, expressed in Escherichia coli system, and then the recombinant protein (named as rOmp-G) was purified with nickel chelating affinity chromatography. The purified rOmp-G showed a good immunogenicity in rabbits and well-conserved characteristics in these three pathogens by enzyme-linked immunosorbed assay. Furthermore, the rOmp-G also showed good immunogenicity in eels (Anguilla anguilla) for eliciting significantly increased specific antibodies (P < 0.01), and providing higher protection efficiencies (P < 0.05) after the pathogens challenge. The values of the relative percent survival in eels were 70% and 50% for two A. hydrophila strain challenge, and 75% for A. sobria strain challenge. This is the first report of a potential vaccination in eels that simultaneously provide protectiveness against different Aeromonas pathogens with a conserved partial OMP.  相似文献

Elymus natans is a dominant native species widely planted to restore the heavily degraded alpine meadows in Qinghai-Tibetan plateau. The objective of this study was to determine how E. natans establishment affected the quality and fertility of a heavily degraded soil. Soil samples (at depths of 0–10, 10–20 and 20–30 cm) were collected from the 3- and 7-year-old E. natans re-vegetated grasslands, and in the heavily degraded alpine meadow (control). The establishment of E. natans promoted plant cover and aboveground biomass. Compared to the non-reseeded meadow, the concentration of total organic C increased by 13% in the soil under 3-year-old reseeded E. natans grassland at 0–10 cm, and by 7–33% in the soil under 7-year-old reseeded E. natans grassland at 0–10, 10–20 and 20–30 cm depths. Rapid increases in total and available N were also observed in two E. natans re-vegetated grasslands, especially in the 0–10 cm soil layer. Across three sampling depths, total P concentration was increased by 17–35% and 18–54% in 3- and 7-year-old reseeded soil respectively, compared to the soil of control. After 3 years of E. natans growth, microbial biomass C increased by 13–58% at 0–10 and 10–20 cm layers; while it increased by 43–87% in 7-year-old reseeded treatment at 0–10, 10–20 and 20–30 cm depths relative to control. A similar increasing trend was observed for microbial biomass N and P generally. Significant increase in neutral phosphatase, urease, catalase and dehydrogenase was also found in 3- and 7-year-old re-vegetated grasslands compared with heavily degraded meadow. Our results suggest a significant positive impact of E. natans establishment on soil quality. Thus, E. natans establishment could be an effective and applicable measure in restoring heavily degraded alpine meadow in the region of Qinghai-Tibetan Plateau.  相似文献
