Transferring fine-scale ecological knowledge into an understanding of earth system processes presents a considerable challenge to ecologists. Our objective here was to identify and quantify heterogeneity of, and relationships among, vegetation and soil properties in terra firme rain forest ecosystems in eastern Amazonia and assess implications for generating regional predictions of carbon (C) exchange. Some of these properties showed considerable variation among sites; soil textures varied from 11% to 92% clay. But we did not find any significant correlations between soil characteristics (percentage clay, nitrogen [N], C, organic matter) and vegetation characteristics (leaf area index [LAI], foliar N concentration, basal area, biomass, stem density). We found some evidence for increased drought stress on the sandier sites: There was a significant correlation between soil texture and wood δ13C (but not with foliar δ13C); volumetric soil moisture was lower at sandier sites; and some canopy foliage had large, negative dawn water potentials (ψld), indicating limited water availability in the rooting zone. However, at every site at least one foliage sample indicated full or nearly full rehydration, suggesting significant interspecific variability in drought vulnerability. There were significant differences in foliar δ15N among sites, but not in foliar % N, suggesting differences in N cycling but not in plant access to N. We used an ecophysiological model to examine the sensitivity of gross primary production (GPP) to observed inter- and intrasite variation in key driving variables—LAI, foliar N, and ψld. The greatest sensitivity was to foliar N; standard errors on foliar N data translated into uncertainty in GPP predictions up to ±10% on sunny days and ±5% on cloudy days. Local variability in LAI had a minor influence on uncertainty, especially on sunny days. The largest observed reductions in ψld reduced GPP by 4%–6%. If uncertainty in foliar N estimates is propagated into the model, then GPP estimates are not significantly different among sites. Our results suggest that water restrictions in the sandier sites are not enough to reduce production significantly and that texture is not the key control on plant access to N. The composition in segregated haploid sets of paternal and maternal chromosomes has been studied in order to verify whether their composition is uniparental of mixed, fixed or variable. Primary cultures where prepared using kidneys from hybrids of strains of Mus musculus in which the parental chromosomes are distinguishable; the maternal set consists of 20 teleocentric chromosomes, the paternal set of 9 metacentric chromosomes, derived by Robertsonian fusion and 2 telocentrics. Applying Seabright's banding technique, an analysis of segregated haploid and diploid cells, which have originated spontaneously through polyploidisation-segregation processes was carried out. It was concluded that the haploid sets have a variable composition of paternal and maternal chromosomes.  相似文献

Folding and insertion of β-barrel outer membrane proteins (OMPs) is essential for Gram-negative bacteria. This process is mediated by the multiprotein complex BAM, composed of the essential β-barrel OMP BamA and four lipoproteins (BamBCDE). The periplasmic domain of BamA is key for its function and contains five "polypeptide transport-associated" (POTRA) repeats. Here, we report the crystal structure of the POTRA4-5 tandem, containing the essential for BAM complex formation and cell viability POTRA5. The domain orientation observed in the crystal is validated by solution NMR and SAXS. Using previously determined structures of BamA POTRA1-4, we present a spliced model of the entire BamA periplasmic domain validated by SAXS. Solution scattering shows that conformational flexibility between POTRA2 and 3 gives rise to compact and extended conformations. The length of BamA in its extended conformation suggests that the protein may bridge the inner and outer membranes across the periplasmic space.  相似文献

Several projects investigating genetic function and evolution through sequencing and comparison of multiple genomes are now underway. These projects consume many resources, and appropriate planning should be devoted to choosing which species to sequence, potentially involving cooperation among different sequencing centres. A widely discussed criterion for species choice is the maximisation of evolutionary divergence. Our mathematical formalization of this problem surprisingly shows that the best long-term cooperative strategy coincides with the seemingly short-term “greedy” strategy of always choosing the next best single species. Other criteria influencing species choice, such as medical relevance or sequencing costs, can also be accommodated in our approach, suggesting our results' broad relevance in scientific policy decisions.  相似文献

As a crucial molecular chaperone in collagen biosynthesis, Hsp47 interacts with the nascent form as well as the mature triple-helical form of procollagen. The location(s) of Hsp47 binding sites on the collagen molecule are, as yet, unknown. We have examined the substrate specificity of Hsp47 in vitro using well-characterized CNBr peptide fragments of type I and type II collagen along with radiolabeled, recombinant Hsp47. Interaction of these peptides with Hsp47 bound to collagen-coated microtiter wells showed several binding sites for Hsp47 along the length of the alpha1 and alpha2 chains of type I collagen and the alpha1 chain of type II collagen, with the N-terminal regions showing the strongest affinities. The latter observation was also supported by the results of a ligand-blot assay. Except for two peptides in the alpha2(I) chain, peptides that showed substantial binding to Hsp47 did so in their triple-helical and not random-coil form. Unlike earlier studies that used peptide models for collagen, the results obtained here on fragments of type I and type II collagen identify, for the first time, binding of Hsp47 to specific regions of the collagen molecule. They also point to additional structural requirements for Hsp47 binding besides the known preference for third-position Arg residues and the triple-helical conformation.  相似文献

The EPR spectrum of sucrose irradiated by high-energy radiation is complex due to the presence of more than one radical species. In order to decompose the spectrum and elucidate the radical magnetic parameters a high-field (HF(-)EPR) study on stable free radicals in gamma-irradiated polycrystalline sucrose (table sugar) was performed at three different high frequencies--94, 190 and 285 GHz as well as at the conventional X-band. We suggest a presence of three stable radicals R1, R2 and R3 as the main radical species. Due to the increase of g-factor resolution at high fields the g-tensors of these radicals could be extracted by accurate simulations. The moderate g-anisotropy suggests that all three radicals are carbon-centred. Results from an earlier ENDOR study on X-irradiated sucrose single crystals (Vanhaelewyn et al., Appl Radiat Isot, 52, 1221 (2000)) were used for analyzing of the spectra in more details. It was confirmed that the strongest hyperfine interaction has a relatively small anisotropy, which indicates either the absence of alpha-protons or a strongly distorted geometry of the radicals.  相似文献
