Infection of Nile tilapia Oreochromis niloticus by monogeneans of the genus Cichlidogyrus is harmful. Currently, diagnosis of this infection is based on invasive techniques and the identification of isolated parasites by their morphology. To facilitate diagnosis, we have developed a non-lethal polymerase chain reaction (PCR) test for detection of Cichlidogyrus spp. DNA in the gill mucus of O. niloticus, using 5 pairs of specific primers based on Cichlidogyrus sclerosus 28S rRNA (Cicly 1 to Cicly 5) which generate fragments of approximately 188, 180, 150, 159 and 189 bp, respectively. PCR specificity was tested using genomic DNA extracted individually from 175 isolated Cichlidogyrus spp., 75 Gyrodactylus cichlidarum and 75 endopararasitic Enterogyrus spp., as well as from 75 protozoans Trichodina spp. The Cicly primers were used to detect Cichlidogyrus spp. DNA in mucus from the gills of 23 Nile tilapia confirmed to be infected with the parasite. Negative controls consisted of 45 uninfected Nile tilapia. The limit of sensitivity of the assay was 1.2 ng of purified parasite DNA. The Cicly primers did not amplify DNA from the mucus of non-infected Nile tilapia, G. cichlidarum, Trichodina spp. or Enterogyrus spp. In all cases, the sensitivity and specificity of the test were 100%. The sequences of all the amplified fragments showed a high similarity to that of the 28S rRNA region of C. sclerosus (93 to 100% identical to GenBank Accession No. DQ157660.1). We provide evidence for a safe and non-invasive DNA-based diagnostic method for the presence of Cichlidogyrus in the gill mucus of O. niloticus.  相似文献

The pink conch Strombus gigas is an important fisheries resource in the Caribbean region, including the Yucatán Peninsula. We analyzed the genetic diversity and genetic structure of two populations (Alacranes Reef and Chinchorro Bank) with the use of five microsatellite molecular markers. The results indicate that the two populations are in the same rank of genetic diversity (He), from 0.613 to 0.692. Significant deviation from H-WE was observed in the both populations due to deficit to heterozygotes, this was attributed to inbreeding as a consequence of over-fishing; nevertheless, other possible causes considered are mixing of individuals from two or more populations, and the existence of null alleles. Levels of genetic differentiation indicated the existence of a single homogenous population in the Yucatan Peninsula (F(ST) de 0.003, p = 0.49), which fits with highest levels of gene flow is significant (2.3 individuals) between both populations. Results from this study support the hypothesis that S. gigas is part of a single panmictic population in the Yucatan Peninsula; therefore, this fishery resource should be regulated the same way for both areas.  相似文献

We surveyed protozoan and metazoan parasites as well as white spot syndrome virus (WSSV) and infectious hypodermal hematopoietic necrosis virus (IHHNV) in white shrimp Litopenaeus setiferus and the palaemonid prawn Macrobrachium acanthurus native to the lower Jamapa River region of Veracruz, Mexico. The presence of parasites and the infection parameters were evaluated in 113 palaemonid prawns collected during the northwind (n = 45), rainy (n = 38) and dry seasons (n = 30) between October 2007 and July 2008, and in 91 shrimp collected in the rainy season between May and June 2008. In L. setiferus, ciliates of the subclass Apostomatia (Ascophrys sp.) were evident in gills, and third-stage larvae of the nematode Physocephalus sexalatus were evident in the stomach. Cestodes of the genus Prochristianella were evident in the hepatopancreas, while some gregarines of the genus Nematopsis, as well as unidentified larval cestodes, were observed in the intestine. Histology identified Ascophrys sp. in association with gill necrosis and tissue melanization. Slight inflammation was observed in intestinal epithelium near cestode larvae. In M. acanthurus, epibionts of the protozoans Epistylis sp., Acineta sp. and Lagenophrys sp. were observed under uropods, periopods and pleopods. An unidentified ciliate of the Apostomatia was also found in the gills, and Nematopsis was identified in the intestine. No histopathology was observed in association with these parasites. Moreover, neither WSSV nor IHHNV were detected by the polymerase chain reaction (PCR) in any of the L. setiferus or M. acanthurus analysed.  相似文献

Between February and June 2010, 113 fiddler crabs Uca spinicarpa were collected from the Chuburna lagoon system on the northern coast of the Yucatan Peninsula, México. Of the 68 crabs gathered outside their burrows, 13 were infected with 25 cystacanths of Hexaglandula corynosoma (intensity of infection from 1 to 5) and the remaining 55 crabs were uninfected. The other 45 crabs were found inside their burrows and only one was found infected with 1 cystacanth of H. corynosoma. Serotonin (5-HT) levels were higher in the group of crabs infected with H. corynosoma in contrast to the group of uninfected crabs and the group of those infected with other parasites. A redundancy analysis corroborated a positive relationship between 5-HT and the intensity of infection with H. corynosoma. In contrast, dopamine levels remained similar among different groups of crabs.  相似文献

The present study documents the first finding of Panulirus argus Virus 1 (PaV1) in spiny lobster Panulirus argus from Cuba. Samples originated from 2 nursery sites, Matias Keys and Bocas de Alonso Keys, and 2 fishing sites, La Grifa and El Ramajo. Lobsters from the nursery sites (artificial reefs) were collected by SCUBA diving, while those from the fishing sites were collected from artificial shelters known as 'casitas cubanas'. In these shelters it was observed that healthy lobsters tended to avoid infected lobsters. Prevalence of PaV1 in the sampling sites was assessed by using clinical signs such as lethargy, an opaque reddish shell coloration, and milky white hemolymph with loss of clotting activity. The presence of PaV1 was subsequently confirmed by histology and PCR of tissues and hemolymph samples from suspected individuals. Histological sections of the hepatopancreas, gills, gonads, and gut showed infected hemocytes with hypertrophied nuclei and eosinophilic intranuclear Cowdry type A inclusions. A 499 bp band was observed by PCR. The sequence of the amplified fragments was 96% similar to the PaV1 sequence in GenBank. The overall mean prevalence of PaV1 was 4.48% (range: 0 to 9.3%) after pooling the results of the 4 sampling sites.  相似文献

Experimental results showed that the gill monogenean Cichlidogyrus sclerosus and its host, the Nile tilapia Oreochromis niloticus, exhibited significant numerical and physiological responses after exposure to sediments polluted with polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls, and heavy metals in comparison with control fishes. After 15 days of exposure, C. sclerosus abundance significantly increased in treatments with low to fairly high sediment pollutant concentrations, but declined at high sediment pollutant concentrations. Hypertrophy and hyperplasia in secondary gill lamellae and the spleen melanomacrophage centers were significantly higher at extremely high sediment pollutant concentrations compared with the controls. Spleen lymphocyte and monocyte counts were significantly lower at extremely high sediment pollutant concentrations and were significantly correlated with high fluorescent aromatic compound concentrations measured as PAH exposure indicators. A multivariate redundancy analysis showed significant statistical association between sediment pollutant concentration, C. sclerosus abundance, and tilapia physiological variables. The polluted sediments negatively affected monogenean abundance and induced immunosuppression in hosts, consequently increasing histological damage in hosts and allowing persistent C. sclerosus infection. This study documents evidence suggesting that C. sclerosus and its host are indeed excellent models to test environmental quality in tropical freshwater ecosystems.  相似文献

The humoral immune response of the tilapia Oreochromis niloticus was evaluated using a direct ELISA. Serum was tested from fish infected with Cichlidogyrus spp. (Monogenea) and from fish injected intraperitoneally with the Cichlidogyrus spp. antigenic extract, i.e., 150 microl of the Cichlidogyrus spp. saline extract diluted in Freund's complete adjuvant (FCA) (1:1) were inoculated intraperitoneally at day 0, followed by 2 dosages of 50 microl of the same Cichlidogyrus spp. saline extract diluted in Freund's incomplete adjuvant (FIA) (1:1) at weeks 2 and 4, respectively. The humoral response was also evaluated by the double immunodiffusion test (DID) and by serum protein and total immunoglobulin (Ig) determinations. The IgM OD values in the hyperimmune fish were significantly higher than in the infected and uninfected fish groups. In the DID test, a precipitation (antigen-antibody) band was observed between the Cichlidogyrus spp. saline extract and hyperimmune sera, but not with the other groups. Increases in serum protein concentration and total Igs were observed in the immunized fish at weeks 2 and 10 postinjection. Results from this study suggest that tilapia is capable of producing an induced humoral immune response against an antigenic extract of Cichlidogyrus spp.  相似文献

The nuclear architecture plays an important role in the temporal and spatial control of complex functional processes within the nucleus. Alterations in nuclear structures are characteristic of cancer cells and the mechanisms underlying these perturbations may directly contribute to tumor development and progression. In this review, we will highlight aspects of the nuclear microenvironment that are perturbed during tumorigenesis and discuss how a greater understanding of the role of nuclear structure in the control of gene expression can provide new options for cancer diagnosis and treatment.  相似文献

The results of a study on the use of Hansch's series design, cluster methodology, for the generation of new benzo[1,2-c]1,2,5-oxadiazole N-oxide derivatives as antitrypanosomal compounds are described. In vitro activity of these compounds was tested against Tulahuen 2 strain of Trypanosoma cruzi. Clearly, the Hansch methodology allowed identifying two cluster-substituents suitable for further structural modifications. The most effective drugs, derivatives 11, 18, and 21, with 50% inhibitory concentration (IC(50)) of the same order as that of the reference drug, represent an excellent structural point of chemical modifications for the design of future drugs. Preliminary results from the study of the mechanism of action of these benzofuroxans point to perturbation of the mitochondrial electron chain, inhibiting parasite respiration.  相似文献

Benzofuroxan derivatives have been shown to inhibit the growth of Trypanosoma cruzi, the etiological agent of Chagas' disease. Therefore, 2D- and 3D-QSAR models of their in vitro antichagasic activity were developed. Six new derivatives were synthesized to complete a final set of 26 structurally diverse benzofuroxans. The 2D-QSAR model (r = 0.939, r(adj)(2) = 0.849) was generated using multiple regression analysis of tabulated substituents' physicochemical properties and indicator variables. In addition, a 3D-QSAR model (r(2) = 0.997, q(2) = 0.802) was obtained using a comparative molecular field analysis (CoMFA). Due to the well-known benzofuroxan tautomerism, in both approaches (2D- and 3D-QSAR) it was necessary to include an indicator variable to consider the N-oxide position (I(6)). This parameter was established using low-temperature NMR experiments. Both QSAR models identified the electrophilic character of the substituent alpha-atom as a requirement for activity. Further support was found using a density functional theory (DFT) approach.  相似文献
