A promising producer of bioactive compounds isolated from a Brazilian tropical soil was tested for its range of antimicrobial activities. Strain 606, classified as Streptomyces sp., could not be identified up to species level, suggesting a possible new taxon. The supernatant and 10 extracts and fractions, obtained by extraction and chromatographic techniques, presented antimicrobial activity using antibiograms. The methanolic fraction was highly active against pathogenic bacteria, phytopathogenic fungi and the human pathogenic yeast Candida albicans. It also possessed high antiviral activity inhibiting the propagation of an acyclovir-resistant herpes simplex virus type 1 strain on HEp-2 cells at non-cytotoxic concentration. The strong cytotoxic effect suggests an antitumour action. This revised version was published online in August 2006 with corrections to the Cover Date.  相似文献

Foundation seaweed species are experiencing widespread declines and localized extinctions due to increased instability of sea surface temperature. Characterizing temperature thresholds are useful for predicting patterns of change and identifying species most vulnerable to extremes. Existing methods for characterizing seaweed thermal tolerance produce diverse metrics and are often time-consuming, making comparisons between species and techniques difficult, hindering insight into global patterns of change. Using three kelp species, we adapted a high-throughput method – previously used in terrestrial plant thermal biology – for use on kelps. This method employs temperature-dependent fluorescence (TF0) curves under heating or cooling regimes to determine the critical temperature (Tcrit) of photosystem II (PSII), i.e., the breakpoint between slow and fast rise fluorescence response to changing temperature, enabling rapid assays of photosynthetic thermal tolerance using a standardized metric. This method enables characterization of Tcrit for up to 48 samples per two-hour assay, demonstrating the capacity of TF0 curves for high-throughput assays of thermal tolerance. Temperature-dependent fluorescence curves and their derived metric, Tcrit, may offer a timely and powerful new method for the field of phycology, enabling characterization and comparison of photosynthetic thermal tolerance of seaweeds across many populations, species, and biomes.  相似文献

Hemicellulose polysaccharides influence assembly and properties of the plant primary cell wall (PCW), perhaps by interacting with cellulose to affect the deposition and bundling of cellulose fibrils. However, the functional differences between plant cell wall hemicelluloses such as glucomannan, xylan, and xyloglucan (XyG) remain unclear. As the most abundant hemicellulose, XyG is considered important in eudicot PCWs, but plants devoid of XyG show relatively mild phenotypes. We report here that a patterned β-galactoglucomannan (β-GGM) is widespread in eudicot PCWs and shows remarkable similarities to XyG. The sugar linkages forming the backbone and side chains of β-GGM are analogous to those that make up XyG, and moreover, these linkages are formed by glycosyltransferases from the same CAZy families. Solid-state nuclear magnetic resonance indicated that β-GGM shows low mobility in the cell wall, consistent with interaction with cellulose. Although Arabidopsis β-GGM synthesis mutants show no obvious growth defects, genetic crosses between β-GGM and XyG mutants produce exacerbated phenotypes compared with XyG mutants. These findings demonstrate a related role of these two similar but distinct classes of hemicelluloses in PCWs. This work opens avenues to study the roles of β-GGM and XyG in PCWs.

Patterned β-GGM resembles xyloglucan in structure, biosynthesis, and function.

In a Nutshell Background: Plant primary cell walls (PCWs) need to be rigid enough to define the plant shape and yet allow cell expansion at the same time. Plants achieve this by forming a complex network that is composed of cellulose and various non-cellulosic polysaccharides, such as hemicelluloses. Cell walls differ in the abundance of the various hemicelluloses, and their roles are poorly understood. In contrast to xyloglucan (XyG), which has been the most extensively studied hemicellulose in the PCWs, neither the structure nor functions of glucomannan has been resolved. Question: Are the functions of the glucomannan in PCWs distinct from the roles of the most abundant hemicellulose, XyG? Findings: We discovered a type of glucomannan in eudicot PCWs, which we named β-galactoglucomannan (β-GGM) because of its distinctive structures: disaccharide side chains of β-Gal-α-Gal and alternating repeats of Glc-Man in the backbone. Similarity to XyG in structure and biosynthesis led us to identify a β-galactosyltransferase for the β-GGM biosynthesis. We found that β-GGM contributed to normal cell expansion, in a way that was masked by the presence of XyG. These results suggest related functions of β-GGM to XyG, highlighting the necessity to consider the contribution of multiple hemicelluloses in the functional study of plant cell walls. Next steps: We would like to know how β-GGM binds to cellulose, and how this differs to cellulose binding of XyG. Investigation of the precise arrangements and interactions of cellulose and hemicelluloses including β-GGM and XyG will help further understanding of the enigmatic functions of hemicelluloses.  相似文献

Zusammenfassung Da in den letzten Jahren die Beobachtungen über ein einkerniges Tapetum weiter zugenommen haben, liegt die Vermutung nahe, daß dieses eine weitere Verbreitung hat, als bisher angenommen wurde. In der vorliegenden Arbeit wurden zur Prüfung der jüngst aufgeworfenen Frage, ob dem Vorkommen des einkernigen Tapetums eine gewisse systematische Bedeutung zukäme, nicht nur die Angaben über das einkernige Tapetum (vgl. die Zusammenstellung S. 15–17) kritisch gesichtet, sondern auch alle Angaben über ein mehrkerniges Tapetum gesammelt und beide Vorkommen einander gegenübergestellt (vgl. den systematischen Teil). Es schien auch angezeigt, auf die verschiedenartigen Abweichungen von der normalen Tapetumentwicklung hinzuweisen, da in einzelnen solchen Fällen (bei weiblichen Pflanzen und pollensterilen Apomikten) ein Unterbleiben oder Verzögern von Kernteilungen in den Tapetumzellen beobachtet werden konnte, wodurch eine Einkernigkeit vorgetäuscht sein kann. — Das Vorhandensein sowohl eines inneren als auch eines mehrschichtigen Tapetums ist in den vorliegenden Fällen nicht an eine bestimmte Kernzahl geknüpft.  相似文献

The Opal multiplex technique is an established methodology for the detection of multiple biomarkers in one section. The protocol encompasses iterative single stainings and heating-mediated removal of the primary and secondary antibodies after each staining round, leaving untouched the Opal fluorophores which are deposited onto the antigen of interest. According to our experience, repetitive heating of skin sections often results in tissue damage, indicating an urgent need for milder alternatives to strip immunoglobulins. In this study, we demonstrate that considerable heating-related damage was found not only in skin but also in tissues of different origin, mostly characterized by low cell density. Importantly, the morphology remained fully intact when sections were repetitively exposed to β-mercaptoethanol-containing stripping buffer instead of multiple heating cycles. However, target epitopes appeared sensitive at a differential degree to multiple treatments with stripping buffer, as shown by loss in staining intensity, but in all cases, the staining intensity could be restored by increment of the primary antibody concentrations. Application of β-mercaptoethanol-containing stripping buffer instead of heating for antibody removal markedly improved the quality of the Opal multiplex technique, as a substantial higher number of differently colored cells could be visualized within a well-conserved morphological context:  相似文献

Gene(s) for the autosomal dominant endocrine cancer syndromes, multiple endocrine neoplasia type 2A (MEN2A), multiple endocrine neoplasia type 2B (MEN2B), and familial medullary thyroid carcinoma (MTC1) all map to the pericentromeric region of chromosome 10. Predictive testing for the inheritance of mutant alleles in individuals at risk for these disorders has been limited by the availability of highly informative and closely linked flanking markers. We describe the development of eight new markers, including two PCR-based dinucleotide repeat polymorphisms and six RFLPs that flank the disease loci. One of the dinucleotide repeat markers (sJRH-1) derives from the RBP3 locus on 10q11.2 and has a PIC of .88. The other dinucleotide repeat (sTCL-1) defines a new locus, D10S176, that maps by in situ hybridization to 10p11.2 and has a PIC of .68. We have constructed a new genetic linkage map of the pericentromeric region of chromosome 10, on the basis of 13 polymorphisms at six loci, which places the MEN2A locus between the dinucleotide repeat markers, with odds of 5,750:1 over the next most likely position. Using this set of markers, predictive genetic testing of 130 at-risk individuals from six families segregating MEN2A revealed that 95% were jointly informative with flanking markers, representing a significant improvement in genetic testing capabilities.  相似文献

The accumulation of lipofuscin in the retinal pigment epithelium (RPE) has been implicated in the development of age-related macular degeneration (AMD) in humans. The exact composition of lipofuscin is not known but its best characterized component is N-retinylidene-N-retinylethanolamine (A2E), a byproduct of the retinoid visual cycle. Utilizing our recently developed matrix-assisted laser desorption/ionization imaging mass spectrometry (MALDI–IMS)-based technique to determine the spatial distribution of A2E, this study compares the relationships of lipofuscin fluorescence and A2E in the murine and human RPE on representative normal tissue. To identify molecules with similar spatial patterns, the images of A2E and lipofuscin were correlated with all the individual images in the MALDI–IMS dataset. In the murine RPE, there was a remarkable correlation between A2E and lipofuscin. In the human RPE, however, minimal correlation was detected. These results were reflected in the marked distinctions between the molecules that spatially correlated with the images of lipofuscin and A2E in the human RPE. While the distribution of murine lipofuscin showed highest similarities with some of the known A2E-adducts, the composition of human lipofuscin was significantly different. These results indicate that A2E metabolism may be altered in the human compared to the murine RPE.  相似文献
