Properties of cationic peptides bearing amino or guanidino groups with various side chain lengths that bind to double stranded RNAs (dsRNAs) were investigated. Peptides with shorter side chain lengths effectively bound to dsRNAs (12mers) increasing their thermal stability. NMR measurements suggested that the cationic peptide binds to the inner side of the major groove of dsRNA. These peptides also increased the thermal stability of siRNA and effectively protected from RNase A digestion. On the other hand, both peptides containing amino groups and guanidine groups did not disturb RNAi activity. 相似文献

The relaxation effect of cilostazol, a phosphodiesterase III inhibitor, on the thoracic aorta was investigated. Cilostazol induced the relaxation of the thoracic aorta precontracted by phenylephrine in a concentration-dependent manner. The concentration-dependent relaxation was shifted to the right in the endothelium denuded aorta compared with that of intact endothelium, suggesting that this relaxation was partly dependent on endothelium. Cilostazol-induced relaxation of thoracic aorta tone was reversed by treatment with N(G)-nitro L-arginine (L-NNA), a competitive inhibitor of nitric oxide (NO) synthase. Cilostazol also significantly increased the NO level in the porcine thoracic aorta. In rats treated with cilostazol, the urinary excretion of nitrites, a stable metabolite of NO, and basal production of NO of the aortic ring were significantly greater than in those without treatment. These findings indicate that cilostazol-induced vasodilation of the rat thoracic aorta was dependent on the endothelium, which released NO from aortic endothelial cells. 相似文献

Background
In April 2009, a novel swine-derived influenza A virus (H1N1pdm) emerged and rapidly spread around the world, including Japan. It has been suggested that the virus can bind to both 2,3- and 2,6-linked sialic acid receptors in infected mammals, in contrast to contemporary seasonal H1N1 viruses, which have a predilection for 2,6-linked sialic acid.Methods/Results
To elucidate the existence and transmissibility of α2,3 sialic acid-specific viruses in H1N1pdm, amino acid substitutions within viral hemagglutinin molecules were investigated, especially D187E, D222G, and Q223R, which are related to a shift from human to avian receptor specificity. Samples from individuals infected during the first and second waves of the outbreak in Japan were examined using a high-throughput sequencing approach. In May 2009, three specimens from mild cases showed D222G and/or Q223R substitutions in a minor subpopulation of viruses infecting these individuals. However, the substitutions almost disappeared in the samples from five mild cases in December 2010. The D187E substitution was not widespread in specimens, even in May 2009.Conclusions
These results suggest that α2,3 sialic acid-specific viruses, including G222 and R223, existed in humans as a minor population in the early phase of the pandemic, and that D222 and Q223 became more dominant through human-to-human transmission during the first and second waves of the epidemic. A new catshark,Scyliorhinus tokubee sp. nov., is described based on specimens from the coast of Shirahama, eastern Izu Peninsula, southern Japan. The present species is distinguished from other congeners in having a particular coloration with dark saddles, blotches and numerous small light spots, a wide oral cleft, the anterior nasal flap not reaching the oral cleft, a short interspace between the dorsal fins, developed clasper hooks, and some meristic characters (number of vertebrae, jaw teeth, and spiral valve turns). Spinach chloroplasts in the light and in the dark were treated with several reagents for protein modification to see the effect of light on their resistivity toward modification. The reagents were p-diazobenzenesulfonic acid, diazonium-1-H-tetrazole, sodium-2,4,6-trinitrobenzenesulfonate, sodium-β-naphtoquinone-4,6-disulfonate and diazonium-1,2,4-triazole. No difference in the absorption spectrum was found between chloroplasts treated with these reagents in the light and those treated in the dark. However, these light- and dark-treated samples when solubilized with a nonionic detergent showed a difference in turbidity. Diazonium-1,2,4-triazole was the most suitable of the above reagents, and the solubilized sample of chloroplasts treated with diazonium-1,2,4-triazole in the light showed a turbidity which was about 2-fold higher than that of the same sample treated in the dark. This increase in turbidity was interpreted as being due to a change in the resistivity toward chemical modification of chloroplasts caused by illumination. In the presence of 3-(p-chlorophenyl)-1,1-dimethylurea, pentachlorophenol and 2-methylthio-4,6-bis-isopropylamino-s-triazine, which are inhibitors of the Hill reaction, the light-induced increase of turbidity was suppressed by 72, 78 and 62%, respectively. The addition of ATP caused a much greater increase of turbidity both in the light and in the dark. We report a novel approach for direct on-membrane glycoproteomics by digestion of membrane-blotted glycoproteins with multiple enzymes using piezoelectric chemical inkjet printing technology and on-membrane direct MALDI-TOF mass spectrometry. With this approach, both N-linked glycan analyses and peptide mass fingerprinting of several standard glycoproteins were successfully performed using PNGase F and trypsin microscale digestions of the blotted spots on membrane from an SDS-PAGE gel. In addition, we performed a similar analysis for 2-DE separated serum glycoproteins as a demonstration of how the system could be used in human plasma glycoproteomics. 相似文献
