Abstract: Bioelectrical impedance analysis (BIA) measures resistance and reactance of a current as it passes through an organism. The validity of using BIA as a tool to measure body water content, and hence body composition and condition, was tested on harp and ringed seals. The resistance and reactance readings from BIA were compared to estimates of total body water (TBW) determined via tritiated water dilution. The relationship between resistance and TBW (% of body mass) was linear after logarithmic transformation and the two variables were highly correlated. We describe the electrode configuration and placements which provide reliable results in these seals. Our findings indicate that BIA has considerable potential as an inexpensive, rapid, and reliable technique for estimating body composition of phocid seals.  相似文献

A comparison of identified faunal assemblages from the Dorset site of Phillip's Garden indicates that harp seal hunting was the main focus of activity throughout the site's occupation. Despite the highly specialized nature of site use, it appears that reliance on harp seal decreased over time while fish and birds became increasingly important. These changes may reflect longer seasonal occupations at the site in later centuries, and/or a decrease in the local availability of harp seal. The observed shift coincides with the onset of a local climatic warming trend, which might have affected harp seal movements in the area. Dorset subsistence and settlement patterns in Newfoundland are still poorly understood due to a lack of preserved faunal assemblages in the region. The temporal trend illustrated here indicates that we cannot assume that these patterns were static throughout the Dorset occupation of the island.  相似文献

AIMS: Wine is the product of complex interactions between yeasts and bacteria in grape must. Amongst yeast populations, two groups can be distinguished. The first, named non-Saccharomyces (NS), colonizes, with many other micro-organisms, the surface of grape berries. In the past, NS yeasts were primarily considered as spoilage micro-organisms. However, recent studies have established a positive contribution of certain NS yeasts to wine quality. Amongst the group of NS yeasts, Brettanomyces bruxellensis, which is not prevalent on wine grapes, plays an important part in the evolution of wine aroma. Some of their secondary metabolites, namely volatile phenols, are responsible for wine spoilage. The other group contributing to wine aroma, which is also the main agent of alcoholic fermentation (AF), is composed of Saccharomyces species. The fermenting must is a complex microbial ecosystem where numerous yeast strains grow and die according to their adaptation to the medium. Yeast-yeast interactions occur during winemaking right from the onset of AF. The aim of this study was to describe the interactions between B. bruxellensis, other NS and Saccharomyces cerevisiae during laboratory and practical scale winemaking. METHODS AND RESULTS: Molecular methods such as internal transcribed spacer-restriction fragment length polymorphism and polymerase chain reaction and denaturing gradient gel electrophoresis were used in laboratory scale experiments and cellar observations. The influence of different oenological practices, like the level of sulphiting at harvest time, cold maceration preceding AF, addition of commercial active dry yeasts on B. bruxellensis and other yeast interactions and their evolution during the initial stages of winemaking have been studied. Brettanomyces bruxellensis was the most adapted NS yeast at the beginning of AF, and towards the end of AF it appeared to be more resistant than S. cerevisiae to the conditions of increased alcohol and sugar limitation. CONCLUSIONS: Among all NS yeast species, B. bruxellensis is better adapted than other wild yeasts to resist in must and during AF. Moreover, B. bruxellensis appeared to be more tolerant to ethanol stress than S. cerevisiae and after AF B. bruxellensis was the main yeast species in wine. SIGNIFICANCE AND IMPACT OF THE STUDY: Brettanomyces bruxellensis interacts with other yeast species and adapts to the wine medium as the dominant yeast species at the end of AF. Contamination of B. bruxellensis might take place at the beginning of malolactic fermentation, which is a critical stage in winemaking.  相似文献

This report describes a novel strategy for isolating Drosophila mutants with conditional eye phenotypes that should be generally applicable for identifying genes required for cellular responses to specific drugs. To test the strategy, we screened 3 of the 5 major chromosome arms for hydroxyurea- and (or) caffeine-sensitive (huc) mutants, and isolated mutations affecting 5 different complementation groups. Most of these were represented by single alleles; however, we also isolated multiple alleles of huc(29DE) gene, an essential gene that is also associated with a nonconditional pupal lethal phenotype. We also identified huc(95E) mutants, which are extremely sensitive to caffeine. Although huc(95E) is a nonessential gene, mutant imaginal disc cells undergo caffeine-dependent apoptosis, and huc(95E) gene function is required for the viability of the organism when mutant larvae are exposed to levels of caffeine that controls can easily tolerate. We have mapped the cytological positions of huc(29D) and huc(95E) as a first step toward molecularly characterizing the relevant genes.  相似文献

Urban areas (especially cities) are challenged in meeting their direct water needs from local sources. They also exert strain on global water resources through their indirect (virtual) water use. Agencies concerned with urban water management have visions and goals for managing direct water use, but indirect use is only inferred in more global visions for sustainable consumption. There is limited quantification of “urban water performance” at the macro urban scale (whole of city) to monitor progress toward these goals. It is constrained by a lack of clarity about the evaluation approaches that best serve them. We ask, How can the evaluation approaches described in literature advance urban water management goals? We reviewed the utility of eight evaluation approaches, including urban water system modeling, urban metabolism (territorial and mass balance), consumption (life cycle assessment, water footprinting, and input‐output analysis), and complex systems (ecological network analysis and systems dynamics) approaches. We found that urban metabolism based on water mass balance is a core method for generating information to inform current goals for direct urban water use, with potential for being “coupled” with the other approaches. Consumption approaches inform the management of indirect water use. We describe this in a framework for urban water evaluation to give greater clarity to this field and flag the further research that would be needed to progress this. It includes the recommendation to differentiate the evaluation of direct and indirect urban water, but to also interpret them together.  相似文献

Inventory and monitoring of wine microbial consortia   总被引:2,自引:0,他引:2  
The evolution of the wine microbial ecosystem is generally restricted to Saccharomyces cerevisiae and Oenococcus oeni, which are the two main agents in the transformation of grape must into wine by acting during alcoholic and malolactic fermentation, respectively. But others species like the yeast Brettanomyces bruxellensis and certain ropy strains of Pediococcus parvulus can spoil the wine. The aim of this study was to address the composition of the system more precisely, identifying other components. The advantages of the polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) approach to wine microbial ecology studies are illustrated by bacteria and yeast species identification and their monitoring at each stage of wine production. After direct DNA extraction, PCR-DGGE was used to make the most exhaustive possible inventory of bacteria and yeast species found in a wine environment. Phylogenetic neighbor-joining trees were built to illustrate microbial diversity. PCR-DGGE was also combined with population enumeration in selective media to monitor microbial changes at all stages of production. Moreover, enrichment media helped to detect the appearance of spoilage species. The genetic diversity of the wine microbial community and its dynamics during winemaking were also described. Most importantly, our study provides a better understanding of the complexity and diversity of the wine microbial consortium at all stages of the winemaking process: on grape berries, in must during fermentation, and in wine during aging. On grapes, 52 different yeast species and 40 bacteria could be identified. The diversity was dramatically reduced during winemaking then during aging. Yeast and lactic acid bacteria were also isolated from very old vintages. B. bruxellensis and O. oeni were the most frequent.  相似文献

Glycopeptides of desired structure can be conveniently prepared by the coupling of reducing oligosaccharides to aspartic acid of peptides via their glycosylamines formed in the presence of saturated aqueous ammonium hydrogen carbonate. The resulting oligosaccharide chains are N-linked to asparagine as in natural glycoproteins, allowing different peptide oligosaccharide combinations to be analysed for conformational effects. In the present paper, a pentapeptide of ovalbumin was coupled to Man5GlcNAc2 oligosaccharide and the glycopeptide and the two parent compounds compared by NMR ROESY experiments and molecular dynamics simulations. Despite the small size of the peptide, conformational effects were observed suggestive of the oligosaccharide stabilising the peptide in solution and of the peptide influencing oligosaccharide conformation. These effects are relevant to the function of glycosylation and the enzymic processing of oligosaccharide chains.  相似文献

The applicability of using stomach temperature telemetry to detect and estimate milk intake throughout lactation was investigated in harbour seals (Phoca vitulina). Observations of a mother-pup pair were conducted daily while stomach temperature was concurrently logged. Milk intake caused a decrease in the pup's stomach temperature. The duration of stomach temperature change was related to the length of the nursing bout (r 2=0.82, P<0.001). Laboratory stomach simulations suggested a strong relationship between milk volume and duration of temperature change (r 2=0.98, P<0.001). This relationship was used to estimate the volume of milk consumed by the pup. Suckling bout length and the estimated milk intake per bout increased, as a weekly average, with pup age (one-way ANOVA; F 3.67=11.66, P<0.001) over the 5-week lactation period. A diel change in nursing time was noted, with a transition from largely nocturnal to daytime feedings. Although not visually confirmed, stomach temperature data collected from the mother provided evidence of seawater ingestion. Methods that could improve estimates of milk intake are discussed. Stomach temperature telemetry proved to be a useful technique for detecting ingestion events in harbour seals, and may provide a valuable tool for investigating lactation energetics and aspects of maternal investment in a variety of nursing species.  相似文献

Purpose

To promote eco-efficient sugarcane products, there is a need for life cycle assessment (LCA) methods that enable rapid assessment of the environmental implications of alternative agricultural practices. In response, a customised LCA method for sugarcane growing was developed and operationalized in the CaneLCA tool. The aim of the paper was to describe the CaneLCA method in detail and to test the effectiveness of the tool’s parameterisation for evaluating the environmental implications of cane growing practice alternatives.

Methods

CaneLCA (Version 1.03) was developed over 6 years (2011–2017) in conjunction with the Australian sugarcane sector. The LCA process was customised for sugarcane growing by focusing on ‘cradle to farm gate’ operations and relevant impact categories, and by parameterising practice variables. To evaluate the effectiveness of the tool, we used it to assess a case study of actual practice changes in the Wet Tropics region of Australia, in terms of the scope of practice variables and environmental implications that can be accounted for.

Results and discussion

The case study results generated by CaneLCA were consistent with those generated by past studies using LCA software. The parameterisation of practice variables allowed for all the practice changes represented in the case study to be assessed. It is suitable for evaluating such known practice alternatives, but less suited to evaluating very innovative practice alternatives, as it is constrained by the underlying algorithms and factors. Most of the environmental implications could be considered, except for effects on soil quality. This will be an area for future tool development to understand the full implications of agricultural practice change, along with the introduction of dynamic models to better estimate emissions.

Conclusions

CaneLCA makes the LCA process more rapid for evaluating alternative sugarcane growing practices, thereby speeding up progress towards devising more eco-efficient sugarcane products. It provides a model that could be adapted for other sugarcane growing regions, and for other perennial cropping systems. The novelty of the method is the detailed parameterisation of practice variables so that a wide range of alternative practices can be evaluated.
