Transferring fine-scale ecological knowledge into an understanding of earth system processes presents a considerable challenge to ecologists. Our objective here was to identify and quantify heterogeneity of, and relationships among, vegetation and soil properties in terra firme rain forest ecosystems in eastern Amazonia and assess implications for generating regional predictions of carbon (C) exchange. Some of these properties showed considerable variation among sites; soil textures varied from 11% to 92% clay. But we did not find any significant correlations between soil characteristics (percentage clay, nitrogen [N], C, organic matter) and vegetation characteristics (leaf area index [LAI], foliar N concentration, basal area, biomass, stem density). We found some evidence for increased drought stress on the sandier sites: There was a significant correlation between soil texture and wood δ13C (but not with foliar δ13C); volumetric soil moisture was lower at sandier sites; and some canopy foliage had large, negative dawn water potentials (ψld), indicating limited water availability in the rooting zone. However, at every site at least one foliage sample indicated full or nearly full rehydration, suggesting significant interspecific variability in drought vulnerability. There were significant differences in foliar δ15N among sites, but not in foliar % N, suggesting differences in N cycling but not in plant access to N. We used an ecophysiological model to examine the sensitivity of gross primary production (GPP) to observed inter- and intrasite variation in key driving variables—LAI, foliar N, and ψld. The greatest sensitivity was to foliar N; standard errors on foliar N data translated into uncertainty in GPP predictions up to ±10% on sunny days and ±5% on cloudy days. Local variability in LAI had a minor influence on uncertainty, especially on sunny days. The largest observed reductions in ψld reduced GPP by 4%–6%. If uncertainty in foliar N estimates is propagated into the model, then GPP estimates are not significantly different among sites. Our results suggest that water restrictions in the sandier sites are not enough to reduce production significantly and that texture is not the key control on plant access to N. - Amazonian goliath catfishes are widespread in the Amazon Basin. Recently, otolith 87Sr:86Sr analyses using laser ablation–multi-collector–inductively coupled plasma mass spectrometry (LA-MC-ICPMS) revealed a >8,000 km trans-Amazonian natal homing in Brachyplatystoma rousseauxii among fish caught and hatched in the largest Amazon River tributary, the upper Madeira basin. Although also suspected for fish in the upper Amazon, homing could not be demonstrated owing to less distinct environmental 87Sr:86Sr gradients along the Amazon mainstem. Using scanning X-ray fluorescence microscopy (SXFM), a separate study provided evidence that Se:Ca and Sr:Ca are useful markers for identifying migration into Andean headwaters and the estuarine environment.
- We analysed otoliths of known 87Sr:86Sr profiles using SXFM mapping to test if Sr:Ca and Se:Ca patterns could demonstrate natal homing for three fish caught in the upper Amazon, using as reference two individuals that were natal homers and two forced residents (hatched after the construction of hydroelectric dams on the Madeira River) from the upper Madeira River.
- As hypothesised, although the Sr isotope profiles of the upper Amazon individuals were uninformative, two of them presented similar alternating mirror patterns of Sr:Ca and Se:Ca to those of the upper Madeira natal homers, indicating migrations out of the Andean region and into the estuary area. Both were therefore natal homers from the upper Amazon.
- The third individual from the upper Amazon presented similar Sr:Ca and Se:Ca patterns to those of the upper Madeira residents, suggesting it was a natural resident from the upper Amazon.
- By combining the results of 87Sr:86Sr analyses (LA-MC-ICPMS) and Sr:Ca and Se:Ca mappings (SXFM) that are completely independent of one another, we demonstrated that B. rousseauxii also performs natal homing in the upper Amazon. Our results indicate that the life cycle of B. rousseauxii is more complex than previous literature hypothesised, with the existence of partial migration, even in absence of physical barriers. Quantifying the relative importance of these different life-history strategies will have important implications for fisheries management. Our results also lay the groundwork for conservation efforts in the context of hydropower development in the Amazon Basin and set testable hypotheses of the potential impacts of the Madeira River dams.

Langerin is a C-type lectin expressed by a subset of dendritic leukocytes, the Langerhans cells (LC). Langerin is a cell surface receptor that induces the formation of an LC-specific organelle, the Birbeck granule (BG). We generated a langerin(-/-) mouse on a C57BL/6 background which did not display any macroscopic aberrant development. In the absence of langerin, LC were detected in normal numbers in the epidermis but the cells lacked BG. LC of langerin(-/-) mice did not present other phenotypic alterations compared to wild-type littermates. Functionally, the langerin(-/-) LC were able to capture antigen, to migrate towards skin draining lymph nodes, and to undergo phenotypic maturation. In addition, langerin(-/-) mice were not impaired in their capacity to process native OVA protein for I-A(b)-restricted presentation to CD4(+) T lymphocytes or for H-2K(b)-restricted cross-presentation to CD8(+) T lymphocytes. langerin(-/-) mice inoculated with mannosylated or skin-tropic microorganisms did not display an altered pathogen susceptibility. Finally, chemical mutagenesis resulted in a similar rate of skin tumor development in langerin(-/-) and wild-type mice. Overall, our data indicate that langerin and BG are dispensable for a number of LC functions. The langerin(-/-) C57BL/6 mouse should be a valuable model for further functional exploration of langerin and the role of BG. 相似文献

Imaging of biofilms on opaque surfaces is a challenge presented to researchers especially considering pathogenic bacteria, as those typically grow on living tissue, such as mucosa and bone. However, they can also grow on surfaces used in industrial applications such as food production, acting as a hindrance to the process. Thus, it is important to understand bacteria better in the environment they actually have relevance in. Stainless steel and titanium substrata were line structured and dotted surface topographies for titanium substrata were prepared to analyze their effects on biofilm formation of a constitutively green fluorescent protein (GFP)‐expressing Escherichia coli strain. The strain was batch cultivated in a custom built flow cell initially for 18 h, followed by continuous cultivation for 6 h. Confocal laser scanning microscopy (CLSM) was used to determine the biofilm topography. Biofilm growth of E. coli GFPmut2 was not affected by the type of metal substrate used; rather, attachment and growth were influenced by variable shapes of the microstructured titanium surfaces. In this work, biofilm cultivation in flow cells was coupled with the most widely used biofilm analytical technique (CLSM) to study the time course of growth of a GFP‐expressing biofilm on metallic surfaces without intermittent sampling or disturbing the natural development of the biofilm. 相似文献

We introduce a new experimental system combining adenovirus-mediated gene transfer and fetal thymic organ culture (FTOC). This system allowed us to efficiently express in developing thymocytes a mutant form of the NF-kappa B inhibitor I kappa B alpha (mut-I kappa B) and to study the maturation defects occurring when NF-kappa B activation is inhibited during fetal development. Fetal thymocytes infected with adenovirus containing mut-I kappa B were found to develop normally until the CD44-CD25+, CD4-CD8- double-negative stage, while production of more mature double-positive and single-positive populations was strongly decreased. Proliferation, as measured by the percentage of cells in cycle appeared normal, as did rearrangement and expression of the TCR beta-chain. However, apoptosis was much higher in FTOC infected with adenovirus containing mut-I kappa B than in FTOC infected with a control virus. Taken together, these results suggest that NF-kappa B plays a crucial role in ensuring the differentiation and survival of thymocytes in the early stages of their development. 相似文献

TLRs function as molecular sensors to detect pathogen-derived products and trigger protective responses ranging from secretion of cytokines that increase the resistance of infected cells and chemokines that recruit immune cells to cell death that limits microbe spreading. Viral dsRNA participate in virus-infected cell apoptosis, but the signaling pathway involved remains unclear. In this study we show that synthetic dsRNA induces apoptosis of human breast cancer cells in a TLR3-dependent manner, which involves the molecular adaptor Toll/IL-1R domain-containing adapter inducing IFN-beta and type I IFN autocrine signaling, but occurs independently of the dsRNA-activated kinase. Moreover, detailed molecular analysis of dsRNA-induced cell death established the proapoptotic role of IL-1R-associated kinase-4 and NF-kappaB downstream of TLR3 as well as the activation of the extrinsic caspases. The direct proapoptotic activity of endogenous human TLR3 expressed by cancerous cells reveals a novel aspect of the multiple-faced TLR biology, which may open new clinical prospects for using TLR3 agonists as cytotoxic agents in selected cancers. 相似文献

Chloroperoxidase (CPO) is a versatile enzyme, which is secreted by the marine fungus Caldariomyces fumago (Leptoxyphium fumago). However, the application of the enzyme is hampered by its high price, which is due to the costly, labor‐intensive purification process. One challenge of the downstream process is the removal of a coproduced black pigment that forms a complex with the active enzyme. While strain development can be considered as an option to reduce the synthesis of the interfering pigment, the metabolism of the microorganism can be altered alternatively by using the biofilm growth mode of the fungus. The aim of this study was to reduce pigment formation during CPO synthesis. We investigated for the first time CPO production during C. fumago biofilm growth initiated through the presence of different microstructured stainless steel surfaces (material number: 1.4571; AISI 316Ti). CPO production by C. fumago was similar when grown as a biofilm or in suspension, whereas pigment formation was drastically reduced by cells grown on moderately structured surfaces (Ra = 0.13 ± 0.02 μm). The possibilities of biofilm growth for changing cell properties and for continuous fermentation are discussed. 相似文献

Titanium is known to possess excellent biocompatibility as a result of corrosion resistance, lack of allergenicity when compared with many other metals. Fluoride is well known as a specific and effective caries prophylactic agent and its systemic application has been recommended widely over recent decades. Nevertheless, high fluoride concentrations impair the corrosion resistance of titanium. The purpose of this article is to summarize the current data regarding the influence of fluoride on titanium corrosion process in the last 5?years. These data demonstrate noxious effects induced by high fluoride concentration as well as low pH in the oral cavity. Therefore, such conditions should be considered when prophylactic actions are administrated in patients containing titanium implants or other dental devices. 相似文献

Staphylococcal enterotoxins are bacterial products that display superantigen activity in vitro as well as in vivo. For instance, staphylococcal enterotoxin B (SEB) polyclonally activates T cells that bear the Vbeta8 gene segment of the TCR. SEB-activated T cells undergo a burst of proliferation that is followed by apoptosis. Using an in vivo adaptation of a fluorescent cell division monitoring technique, we show here that SEB-activated T cells divide asynchronously, and that apoptosis of superantigen-activated T cells is preferentially restricted to cells which have undergone a discrete number of cell divisions. Collectively, our data suggest that superantigen-activated T cells are programmed to undergo a fixed number of cell divisions before undergoing apoptosis. A delayed death program may provide a mechanistic compromise between effector functions and homeostasis of activated T cells. 相似文献

On the basis of faunistic and floristic inventories, various authors have developed the forest refuge theory to explain species diversity in humid tropical regions. Renno et al. (1990) used electrophoretic markers to study the genetic structure of Leporinus friderici and suggested the existence of an aquatic refuge on the Guiana shield. In the present study, mitochondrial markers (RFLP) confirmed and complemented the previous electrophoretic study. Four multimorphs were evidenced, allowing the populations to be separated into two sets on either side of the eastern edge of the Guiana refuge, i.e. the Kourou river region in French Guiana. 相似文献
