In order to study saxitoxin (STX) production bymicro-algae in the laboratory, a defined algal culture medium which supports
optimum growth over a longtime-period is a requirement. In the development of such a medium, a number of modifications were
made to a standard algal culture medium (GP) and growth of a STX-producing isolate of Alexandrium minutum in the different formulations was assessed by measuring maximum cell densities and mean generation times (MGT). All experiments
were carried out under controlled conditions in an aerobic atmosphere with increased CO2. Whilst maximum cell densities in the different modifications were similar, the MGT was significantly shortened by the addition
of Tris buffer and the trace metals strontium, selenium and molybdenum. Replacement of natural with artificial seawater and
removal of soil extract did not adversely affect algal growth. Five of the six media formulations supported the growth of
A. minutumover a 9-month period.

The present study is aimed at determining some haematological and biochemical parameters in the wild Indian bonnet monkeys as also the microscopic and ultrastructural characteristics of their pancreatic islets. Adult wild Indian bonnet monkeys (Macaca radiata radiata) of both sexes weighing between 2.5 and 4 kg were used in these experiments. Their platelet, reticulocyte and total leukocyte counts and the blood concentrations of hemoglobin and plasma proteins and the serum concentrations of aspartate amino transferase, alanine amino transferase and calcium are similar to the values reported for M. mulatta. Plasma glucose is lower when compared with reported values of M. mulatta and M. fascicularis. Insulin levels are comparable with those of M. mulatta and M. nigra. Histology of islets is similar to that of humans. Ovoid cell collections of islet cells are scattered throughout the pancreas. Ultrastructure of A, B and D cells is similar to humans. These findings suggest that this relatively underutilized macaques may be a suitable model for biomedical research. 相似文献

We analysed monthly samples collected in Guanabara Bay, with a conical net of 200 m mesh during 1985. The bay was divided into three areas: an outer region (area A), influenced by oceanic waters; an inner region (area C), influenced by fluvial inflow; and a transition region (area B) with intermediate features. Penilia avirostris and Evadne tergestina were observed in the three areas, with greatest densities, however, in the outermost region, which had the highest salinities and lowest temperatures. Penilia avirostris was more abundant in summer (March), a period with the greatest relative densities of nanoplankton. Evadne tergestina was also abundant in summer, but its peak fell in November, a period with a relative increase in microphytoplankton density in the bay. The two species disappeared in winter: Penilia avirostris was absent from May to August, whereas Evadne tergestina disappeared in August and September. 相似文献

Calcium (Ca) and manganese (Mn) are essential nutrients required for normal plant growth and development, and transport processes play a key role in regulating their cellular levels. Arabidopsis (Arabidopsis thaliana) contains four P(2A)-type ATPase genes, AtECA1 to AtECA4, which are expressed in all major organs of Arabidopsis. To elucidate the physiological role of AtECA2 and AtECA3 in Arabidopsis, several independent T-DNA insertion mutant alleles were isolated. When grown on medium lacking Mn, eca3 mutants, but not eca2 mutants, displayed a striking difference from wild-type plants. After approximately 8 to 9 d on this medium, eca3 mutants became chlorotic, and root and shoot growth were strongly inhibited compared to wild-type plants. These severe deficiency symptoms were suppressed by low levels of Mn, indicating a crucial role for ECA3 in Mn nutrition in Arabidopsis. eca3 mutants were also more sensitive than wild-type plants and eca2 mutants on medium lacking Ca; however, the differences were not so striking because in this case all plants were severely affected. ECA3 partially restored the growth defect on high Mn of the yeast (Saccharomyces cerevisiae) pmr1 mutant, which is defective in a Golgi Ca/Mn pump (PMR1), and the yeast K616 mutant (Deltapmc1 Deltapmr1 Deltacnb1), defective in Golgi and vacuolar Ca/Mn pumps. ECA3 also rescued the growth defect of K616 on low Ca. Promoter:beta-glucuronidase studies show that ECA3 is expressed in a range of tissues and cells, including primary root tips, root vascular tissue, hydathodes, and guard cells. When transiently expressed in Nicotiana tabacum, an ECA3-yellow fluorescent protein fusion protein showed overlapping expression with the Golgi protein GONST1. We propose that ECA3 is important for Mn and Ca homeostasis, possibly functioning in the transport of these ions into the Golgi. ABSTRACT We describe the deleterious effects of using an antimony potassium tartrate emetic to obtain diet samples from birds, and compare information obtained from regurgitated samples versus fecal samples in describing diets of autumn migrants. We also examined dose effectiveness in captive Dark‐eyed Juncos (Junco hyemalis) subjected to the same emetic technique used in the field. Over 70% of migrants given an emetic at a study site in Idaho regurgitated useful samples. For 5 of 7 species analyzed, regurgitated samples produced significantly more arthropods per sample than fecal samples, and one species, Warbling Vireo, showed higher numbers of distinct arthropod taxa per sample. In most species, regurgitated samples accumulated arthropod taxa more quickly than fecal samples. However, increasing the number of fecal samples by 5–17 produced a similar number of taxa. Diet composition based on fecal versus regurgitated samples was generally similar, but there were significant differences. Two of 130 treated migrants died soon after treatment. Recapture frequency for treated birds was less than half that for untreated birds, but it is not clear whether this difference was due to treatment‐related mortality or emigration. Each treated bird that we recaptured had lost mass and this suggests a deleterious effect because untreated migrants tended to gain mass. In captivity, 18 Dark‐eyed Juncos were treated with emetic: 6 with the full mass‐specific recommended dose, 6 with half the recommended dose, and the final 6 with one quarter the recommended dose. All were alive 15–20 min posttreatment (recommended release time), but 17 of 18 died within 30 min after receiving the emetic. Together, our data suggest that although the emetic technique may be slightly more information‐rich in assessing diet, it is more harmful than previously reported especially in certain species and should be used only after adequate consideration of the immediate mortality and short‐term physiological effects on birds to be studied. 相似文献

For decades, the search for new vaccine adjuvants has been largely empirical. A series of new adjuvants and related formulations are now emerging that are acting through identified immunological mechanisms. Understanding adjuvant mechanism of action is crucial for vaccine design, since this allows for directing immune responses towards efficacious disease-specific effector mechanisms and appropriate memory. It is also of great importance to build new paradigms for assessing adjuvant safety at development stages and at regulatory level. This report reflects the conclusions of a group of scientists from academia, regulatory agencies and industry who attended a conference, organized by the International Association for Biologicals (IABS), on the mode of action of adjuvants on 29–30 April 2010 in Bethesda, Maryland, USA, particularly focusing on how understanding adjuvants mode of action can impact on the assessment of vaccine safety and help to develop target-specific vaccines. A high level of human development is dependent on energy consumption (roughly 4 kW per person), and most developed countries that have reached this level have done so through the extensive use of fossil energy. However, given that fossil resources are finite, in order for developed countries to maintain their level of development and simultaneously allow developing countries to reach their potential, it is essential to develop viable renewable energy alternatives. Of particular importance are liquid fuel replacements for petroleum, the fossil resource that primarily drives commerce and economic growth. The intent of this article is to remind our fellow biofuel researchers, particularly those involved in lignocellulosic pretreatment, of these global issues and the serious nature of our work. We hope that this will inspire us to generate and report higher quality and more thorough data than has been done in the past. Only in this way can accurate comparisons and technoeconomic evaluations be made for the many different pretreatment technologies that are currently being researched. The data that primarily influence biorefinery economics can be subdivided into three main categories: yield, concentration, and rate. For these three categories we detail the specific data that should be reported for pretreatment research. In addition, there is other information that is needed to allow for a thorough comparison of pretreatment technologies. An overview of these criteria and our comparison of the current state of a number of pretreatment technologies with respect to these criteria are covered in the last section. © 2012 American Institute of Chemical Engineers Biotechnol.
