Much research has been conducted about different types of fermentation at high temperature, but only a few of them have studied cell viability changes during high-temperature fermentation. In this study, Acetobacter senegalensis, a thermo-tolerant strain, was used for gluconic acid production at 38 °C. The influences of different carbon sources and physicochemical conditions on cell viability and the resuscitation of viable but nonculturable (VBNC) cells formed during fermentation were studied. Based on the obtained results, A. senegalensis could oxidize 95 g l− 1 glucose to gluconate at 38 °C (pH 5.5, yield 83%). However, despite the availability of carbon and nitrogen sources, the specific rates of glucose consumption (qs) and gluconate production (qp) reduced progressively. Interestingly, gradual qs and qp reduction coincided with gradual decrease in cellular dehydrogenase activity, cell envelope integrity, and cell culturability as well as with the formation of VBNC cells. Entry of cells into VBNC state during stationary phase partly stemmed from high fermentation temperature and long-term oxidation of glucose, because just about 48% of VBNC cells formed during stationary phase were resuscitated by supplementing the culture medium with an alternative favorite carbon source (low concentration of ethanol) and/or reducing incubation temperature to 30 °C. This indicates that ethanol, as a favorable carbon source, supports the repair of stressed cells. Since formation of VBNC cells is often inevitable during high-temperature fermentation, using an alternative carbon source together with changing physicochemical conditions may enable the resuscitation of VBNC cells and their use for several production cycles.

Background

Mathematical modeling has achieved a broad interest in the field of biology. These models represent the associations among the metabolism of the biological phenomenon with some mathematical equations such that the observed time course profile of the biological data fits the model. However, the estimation of the unknown parameters of the model is a challenging task. Many algorithms have been developed for parameter estimation, but none of them is entirely capable of finding the best solution. The purpose of this paper is to develop a method for precise estimation of parameters of a biological model.

Methods

In this paper, a novel particle swarm optimization algorithm based on a decomposition technique is developed. Then, its root mean square error is compared with simple particle swarm optimization, Iterative Unscented Kalman Filter and Simulated Annealing algorithms for two different simulation scenarios and a real data set related to the metabolism of CAD system.

Results

Our proposed algorithm results in 54.39% and 26.72% average reduction in root mean square error when applied to the simulation and experimental data, respectively.

Conclusion

The results show that the metaheuristic approaches such as the proposed method are very wise choices for finding the solution of nonlinear problems with many unknown parameters.

Measles virus (MV) causes small and large outbreaks in Iran. Molecular assays allow identifying and the sources of measles imported from neighboring countries. We carried out a phylogenetic analysis of measles virus circulating in Iran over the period 2010–2012. Specimens from suspected cases of measles were collected from different regions of Iran. Virus isolation was performed on urine and throat swabs. Partial nucleoprotein gene segments of MV were amplified by RT-PCR. PCR products of 173 samples were sequenced and analyzed. The median age of confirmed cases was 2 years. Among all confirmed cases, 32% had unknown vaccination status, 20% had been vaccinated, and 48% had not been vaccinated. Genotypes B3 and D8 (for the first time), H1 and D4 were detected mainly in unvaccinated toddlers and young children. Genotype B3 became predominant in 2012 and was closely related to African strains. H1 strains were also found in small and large outbreaks during 2012 but were not identical to Iranian H1-2009 strains. A majority of the Iranian D4 strains during 2010–2012 outbreaks were linked to the D4 strain identified in the Pakistan in 2007. We identified a single case in 2010 belonging to D8 genotype with 99.7% identity to Indian isolates. Although the vaccination program is currently good enough to prevent nationwide epidemics and successfully decreased measles incidence in Iran, the fraction of protected individuals in the population was not high enough to prevent continuous introduction of cases from abroad. Due to increasing number of susceptible individuals in some areas, sustained transmission of the newly introduced viral genotype remains possible.  相似文献

The silverleaf whitefly is one of the most important pests found commonly in Iran and in the world. The development of alternative methods instead of chemical applications is necessary in pest management for human health and environment protection. The objective of this research was to assess the effects of extracts of two medicinal plant species: Calotropis procera and Allium sativum, and a formulation containing azadirachtin on fecundity and fertility of the silverleaf whitefly, grown on greenhouse tomato plants. The effects were compared to that of pymetrozine, a synthetic insecticide. According to the results, there was a significant difference among treatments for all reproductive parameters. Gross fecundity rates for pesticides control, herbal extract control, C. procera extract, A. sativum extract, azadirachtin and pymetrozine were 184.75, 146.72, 80.11, 82.18, 63.06 and 55.96 eggs, respectively. These herbal extracts were effective against this pest and they can be the suitable choices for replacing these chemical insecticides.  相似文献
