Background aimsThe aim was to investigate the therapeutic effect of granulocyte–colony-stimulating factor (G-CSF) administration following implantation of autologous bone marrow mononuclear cells (BM MNC) for patients with lower limb ischemia.MethodsThe design was a randomized controlled trial. Fifteen patients with severe chronic limb ischemia were treated with autologous BM MNC [without G-CSF (MNC–G-CSF) or combined with G-CSF administration for 5 days following transplantation (MNC+G-CSF)].ResultsAll clinical parameters, including ankle brachial index, visual analog scale and pain-free walking distance, showed a mean improvement from baseline, which was measured at 4 and 24 weeks after transplantation in both groups. However, in three (20%) patients, the clinical course did not improve and limb salvage was not achieved. No significant difference was observed among the patients treated in the MNC–G-CSF and MNC+G-CSF groups. No severe adverse reactions were reported during the study period. No relationship was observed between both the numbers of viable MNC or CD34+ cells and the clinical outcome.ConclusionsAutologous transplantation of BM MNC into ischemic lower limbs is safe, feasible and efficient for patients with severe peripheral artery disease. However, the administration of G-CSF following cell transplantation does not improve the effect of BM MNC implantation and therefore would not have any beneficial value in clinical applications of such cases.  相似文献

In arid and semi‐arid regions of the world, including Iran, soil salinity is one of the major abiotic stresses. One of the ways to achieve high performance in these areas is to use salt‐tolerant varieties of wheat. Iran is known as one of the places where the D‐genome originated and evolved. In order to evaluate the salt tolerance of Iranian genotypes based on the eight indices using analysis of variance, regression and an artificial neural network (ANN), 41 Iranian wheat varieties (Trticum aestivum L.) were planted in a randomised complete block design with three replications under two saline irrigation conditions, 0.631 and 11.8 dS m?1, in Kerman, Iran. Significant differences between the varieties were observed, and the significant two‐way interaction of environment × varieties in combined analysis and non‐significant correlation, 0.07, between the yield in two environments (yield in non‐stress conditions, Yp, and yield in stress conditions, Ys) indicates the existence of genetic variation among varieties and the different responses of the varieties in both the environments. The indices of tolerance, geometric mean product (GMP), mean product (MP), harmonic mean (HM) and stress tolerance index (STI) were calculated based on grain yield evidence of positive significant correlation with Yp and Ys. Based on the ANN results, yield stability index (YSI), MP, GMP and STI were the best indices to predict salinity‐tolerant varieties. The varieties selected based on these indices, such as Bolani, Sistan, Ofogh, Pishtaz, Karchia and Arg, produced high yield in both the environments. These results show that bread wheat originating from Iran has salt tolerance potential and can also be used in studies related to salinity tolerance mechanisms.  相似文献

Background aimsRecalcitrant diabetic wounds are not responsive to the most common treatments. Bone marrow-derived stem cell transplantation is used for the healing of chronic lower extremity wounds.MethodsWe report on the treatment of eight patients with aggressive, refractory diabetic wounds. The marrow-derived cells were injected/applied topically into the wound along with platelets, fibrin glue and bone marrow-impregnated collagen matrix.ResultsFour weeks after treatment, the wound was completely closed in three patients and significantly reduced in the remaining five patients.ConclusionsOur study suggests that the combination of the components mentioned can be used safely in order to synergize the effect of chronic wound healing.  相似文献
