The ultrastructure of cytolytic T lymphocytes adhered to the surface of target cells was investigated at different periods after start of interaction. Fifteen-minute incubation led to increase of number of Golgi apparatus cisternae and vacuoles. After 30 min incubation Golgi apparatus become oriented to the contact area. If several lymphocytes adhered to one target cell the Golgi apparatus of each of them was oriented toward the contact area. If one lymphocyte adhered simultaneously to two target cells its Golgi apparatus was oriented toward both target cells. Giant Golgi apparatus vacuoles were formed 30 to 60 min later and then moved to plasma membrane of lymphocyte and then the content of those vacuoles moved to the intercellular space between a cytolytic T lymphocyte and a target cell. The period required for the hypertrophy and change of orientation of Golgi apparatus is supposed to represent the “mobilization” step of a medium-sized and small killer lymphocyte.  相似文献

The effect of the ubiquitous downregulation of insulin receptor (InR) gene expression on the metabolism of juvenile hormone (JH) and dopamine (DA) in young females of D. melanogaster under normal conditions and heat stress is studied. The level of JH degradation and alkaline phosphatase activity (ALP, an enzyme regulating DA synthesis) were used as indicators of JH and DA levels, respectively. We demonstrated that, under normal conditions, the ubiquitous inhibition of the InR gene expression in D. melanogaster females induced an increase in the JH degradation and ALP activity. As we have already shown, this is indicative of the decrease in the concentration of the above hormones. It was also found that the total inactivation of InR does not affect the initiation of JH and DA metabolic system response to heat stress; however, it does affect its intensity. Thus, the involvement of the insulin signaling pathway in the regulation of the JH and DA metabolism in Drosophila females was demonstrated in vivo under normal and stress conditions.  相似文献

The effect of strong hypomorphic mutation of the insulin-like protein gene (dilp6) on metabolism of octopamine (one of the main biogenic amines in insects) was studied in Drosophila melanogaster males and females. The activity of tyrosine decarboxylase (the key enzyme of octopamine synthesis) and the activity of octopamine-dependent N-acetyltransferase (the enzyme of its degradation) were measured. It was demonstrated that the activity of both studied enzymes is decreased under normal conditions in the dilp641 mutants (as we previously demonstrated, this is correlated with an increased level of octopamine). It was also found that hypomorphic mutation of the dilp6 gene decreases the intensity of tyrosine decarboxylase response to heat stress. Thus, it was demonstrated for the first time that insulin-like DILP6 protein in drosophila influences the level of octopamine (regulating the activity of the enzyme degrading octopamine).  相似文献

It is known that prolactin (PRL) is the third pituitary hormone serving gonadotropic function in mammals. However, its role in the regulation of ovarian folliculogenesis and, in particular, its relationship to follicular atresia as well as the mechanism of its influence on follicular cells are poorly understood. We investigated PRL levels in follicular fluids (FFs) and intracellular store calcium ([Ca2+]is) in cell walls of bovine ovarian follicles with diameters of 10 to 20 mm and their relationship to follicular atresia. Ovarian follicles were categorized on the basis of macroscopic criteria and of microscopic examination of granulosa cell (GC) smears. Prolactin concentrations in FFs were measured by RIA and levels of [Ca2+]is in follicular cells were determined by using the fluorophore chlortetracycline. Compared to atretic follicles, morphologically normal follicles were characterized by higher concentrations of PRL in FFs (P < 0.001) and lower contents of [Ca2+]is in follicular cells (P < 0.01). Furthermore, follicles containing no more than 20% of pycnotic GCs had higher levels of PRL in their fluids than those containing over 40% of pycnotic GCs (P < 0.05). Finally, the direct effect of PRL on [Ca2+]is content in follicular cells was studied in vitro. Compared to control, PRL decreased (P < 0.001) the levels of [Ca2+]is in the cells after 24 h culture of follicular walls from morphologically normal follicles in TCM 199 supplemented by 10% fetal calf serum. Our findings suggest that the decline of PRL concentrations in FFs and the rise of [Ca2+]is contents in follicular cells are related to atresia of large bovine follicles and that there appears to be a relationship between the two biochemical parameters.  相似文献

“Secretory” granules, crystal-like structures, lysosema-like granules, ergastoplasmatic lamellae, electron-opaque ribosomes, and lipid vacuoles were detected in the cytoplasm of cytolytic T lymphocytes. After 30 to 60 min interaction with a target cell the fusion of “secretory” granules and crystal-like structures with lipids, mitochondrion degeneration, hypertrophy and reorientation of the Golgi apparatus toward the contact zone with a target cell were observed.  相似文献

Juvenile hormone hydrolysis, tyrosine decarboxylase activity, dopamine content and fitness (viability and fertility) were studied under normal and stress conditions in the adults of Drosophila melanogaster inactive strain carrying a mutation that decreases tyrosine decarboxylase activity and results in the lower contents of tyramine and octopamine. A sexual dimorphism of tyrosine decarboxylase activity, dopamine level and survival under heat stress in inactive flies was found. inactive adults showed higher dopamine levels and lower survival levels under heat stress as compared to wild type (Canton S) adults. Juvenile hormone degradation is decreased in young and increased in mature inactive females as compared to wild type. Fertility of the inactive strain did not differ from that of wild type strain under normal conditions, but after heat exposure the dynamics of its restoration was different. inactive females were found to develop the stress reaction, with juvenile hormone degradation, tyrosine decarboxylase activity, dopamine content and fertility levels used as the reaction indicators.  相似文献

The effect of Dipyridamole (10(-6)-10(-3) M) on the photomobilized electron transport in the system of quinone acceptors Q(A)-Q(B) of isolated photosynthetic reaction centers of Rhodobacter sphaeroides and on its temporary stabilization on Q(B) was studied. Depending on the type of the detergent present in the reaction center (lauryl dimethylamine oxide, Triton X-100, sodium dodecyl sulfate, and sodium cholate), dipyridamole could increase the time of the electron transfer to Q(B). The dipyridamole effect on the efficiency of the electron stabilization on Q(B) for reaction centers with different detergents was revealed in slowing down the process of dark reduction of photoactive bacteriochlorophyll from Q(B) at initial concentrations of added dipyridamole (10(-6)-10(-5) M) with following acceleration of the process at the dipyridamole concentrations of 10(-4)-10(-3) M. The pH lowering from 6.8-7.0 to 5.9-6.0 increased the dipyridamole effect. The possibility of the dipyridamole effect on the structural-dynamic state of the reaction center complex, including its hydrogen bond system, which influences the studied parameters of functional activity, is suggested.  相似文献

In this work, we evaluated the effect of adaptation to heat on the fall of blood pressure (BP) induced by heat shock (HS) and the interrelation between nitric oxide (NO) and heat shock protein, HSP70. Experiments were carried out on Wistar rats. It was shown that HS resulted in a generalized and transient increase in NO production (the electron paramagnetic resonance method) and a fall of BP from 113+/-3 to 88+/-1 mm Hg (p<0.05). Adaptation to heat itself did not affect BP, but completely prevented the NO overproduction and hypotension induced by HS. The adaptation simultaneously increased the brain NO-synthase content and induced HSP70 synthesis (the Western blot analysis) in various organs. Both the antihypotensive effects of adaptation and HSP70 accumulation were completely prevented by L-NNA, an inhibitor of NO synthesis, or quercetin, an inhibitor of HSP70 synthesis. The data suggest that adaptation to heat stimulates NO synthesis and NO activates synthesis of HSP70. HSP70, which hampers NO overproduction, thus restricts the BP fall induced by heat shock.  相似文献

The dopamine (DA) content and the level of juvenile hormone (JH) degradation were studied in females of the wild-type Canton S strain and the ecdysoneless 1 (ecd 1) mutant, which does not produce ecdysone at a restrictive temperature (29°C). Exposure at the restrictive temperature considerably increased the JH-hydrolyzing activity and the DA content in five-day ecd 1 females compared with flies of both strains growing at 19°C and Canton S females exposed at 29°C. In one-day ecd 1 females, the level of JH degradation also increased at the restrictive temperature, but the DA content was low. The effect of ecdysone deficiency on the stress reaction in Drosophila melanogaster females was studied using changes in DA content and JH degradation as the reaction indicators. The ecd 1 mutation did not prevent the initiation of the stress reaction in females exposed at the restrictive temperature, but changed its intensity (stress reactivity). The interaction of 20-hydroxyecdysone with JH and DA in regulating Drosophila reproduction under normal conditions and in stress is discussed.  相似文献
