An attempt to induce shoot regeneration from leaf disc explants from Allium sativum L., A. porrum L., and A. schoenoprasum L. and the induction of shoot regeneration from single flower-bud receptacles in A. porrum is presented. While the regeneration rate from leaf disc explants was low, an efficient method for propagating A. porrum in vitro was obtained by cultivating single flower-bud receptacles. The shoot regeneration ability was strongly controlled by the genotype. Up to 294 shoots per leek plant could be harvested. Simultaneously the same plant could be used for seed production and bulbil formation in vivo. The efficiency of the in vitro multiplication method described allows the integration of this procedure into breeding programmes of A. porrum

Microenvironment and activation signals likely imprint heterogeneity in the lymphatic endothelial cell (LEC) population. Particularly LECs of secondary lymphoid organs are exposed to different cell types and immune stimuli. However, our understanding of the nature of LEC activation signals and their cell source within the secondary lymphoid organ in the steady state remains incomplete. Here we show that integrin alpha 2b (ITGA2b), known to be carried by platelets, megakaryocytes and hematopoietic progenitors, is expressed by a lymph node subset of LECs, residing in medullary, cortical and subcapsular sinuses. In the subcapsular sinus, the floor but not the ceiling layer expresses the integrin, being excluded from ACKR4+ LECs but overlapping with MAdCAM-1 expression. ITGA2b expression increases in response to immunization, raising the possibility that heterogeneous ITGA2b levels reflect variation in exposure to activation signals. We show that alterations of the level of receptor activator of NF-κB ligand (RANKL), by overexpression, neutralization or deletion from stromal marginal reticular cells, affected the proportion of ITGA2b+ LECs. Lymph node LECs but not peripheral LECs express RANK. In addition, we found that lymphotoxin-β receptor signaling likewise regulated the proportion of ITGA2b+ LECs. These findings demonstrate that stromal reticular cells activate LECs via RANKL and support the action of hematopoietic cell-derived lymphotoxin. 相似文献

MPI collective communication operations to distribute or gather data are used for many parallel applications from scientific computing, but they may lead to scalability problems since their execution times increase with the number of participating processors. In this article, we show how the execution time of collective communication operations can be improved significantly by an internal restructuring based on orthogonal processor structures with two or more levels. The execution time of operations like MPI_Bcast() or MPI_Allgather() can be reduced by 40% and 70% on a dual Xeon cluster and a Beowulf cluster with single-processor nodes. But also on a Cray T3E a significant performance improvement can be obtained by a careful selection of the processor structure. The use of these optimized communication operations can reduce the execution time of data parallel implementations of complex application programs significantly without requiring any other change of the computation and communication structure. We present runtime functions for the modeling of two-phase realizations and verify that these runtime functions can predict the execution time both for communication operations in isolation and in the context of application programs. 相似文献

The adzuki bean (Vigna angularis (Wild.) Ohwi and Ohashi) and common bean (Phaseolus vulgaris L.) have a high physiological demand for N. A 2-year field study was conducted to investigate the seasonal change of available soil N and symbiotic N2 fixation usage. The beans were seeded at two densities, 22.2 plants m–2 with a row spacing of 0.3 m and 11.1 plants m–2 with a row spacing of 0.6 m. The amount of fixed N2 in the shoot was calculated using the 15N natural abundance method. The common bean demonstrated low N2 fixation and the ability to accumulate high levels of soil N. Soil nitrate under the common bean was continually absorbed. The adzuki bean, on the other hand, had a remarkable peak of N accumulation in the early reproductive stage. This was mainly due to N2 fixation, though the soil nitrate level was high. Narrowing the plant row spacing increased the dry matter yield of both species, but the origin of the increased N differed between the species. For the first 77 DAP in 1999 (73 DAP in 2000) the N increase for both beans was due to both soil and atmospheric N2. At harvest, though, the increase of N in common bean was mainly due to soil N, while that in adzuki bean was mainly due to atmospheric N2. It can be concluded that the low symbiotic N2 fixation ability of common bean was due to its high soil N uptake ability and constant N accumulation, which enabled an efficient soil N absorption. Adzuki bean absorbed N mainly for a short period and depended more on symbiotically fixed N2 and, in contrast to common bean, left a high level of NO3-N remaining in the soil after cropping. 相似文献

A hydroponic experiment was carried out to characterize the oxidative stress responses of two potato cultivars (Solanum tuberosum L. cvs. Asterix and Macaca) to cadmium (Cd). Plantlets were exposed to four Cd levels (0, 50, 100, 150 and 200 μM) for 7 days.
Cd concentration was increased in both roots and shoot. Number of sprouts and roots was not decreased, whereas Cd treatment
affected the number of nodal segments. Chlorophyll content and ALA-D activity were decreased in both cultivars, whereas carotenoids
content was decreased only in Macaca. Cd caused lipid peroxidation in roots and shoot of both cultivars. Protein oxidation
was only verified at the highest Cd level. H2O2 content was increased in roots and shoot of Asterix, and apparently, a compensatory response between roots and shoot of Macaca
was observed. SOD activity was inhibited in roots of Asterix at all Cd treatments, whereas in Macaca it was only increased
at two highest Cd levels. Shoot SOD activity increased in Asterix and decreased in Macaca. Root CAT activity in Asterix decreased
at 100 and 150 μM, whereas in Macaca it decreased only at 50 μM. Shoot CAT activity was decreased in Macaca. Root AsA content
in Macaca was not affected, whereas in shoot it was reduced at 100 μM and increased at 200 μM. Cd caused increase in NPSH
content in roots and shoot. Our results suggest that Cd induces oxidative stress in both potato cultivars and that of the
two cultivars, Asterix showed greater sensitivity to Cd levels. 相似文献

Some sulphonium salts derived from lysine were synthesized with the general structure R-Lys-CH2S+-(alkyl)2. They were examined as inhibitors of the cysteine proteinase clostripain, which has a preference for cleaving peptide bonds at the carboxy group of basic amino acids, and of a number of trypsin-related serine proteinases. Clostripain was irreversibly inactivated by all reagents examined, but in the case of the serine proteinases, depending on the reagent structure, irreversible and reversible inhibitions were observed. Biological invasions have important ecological impacts at both local and global levels, affecting biodiversity, ecosystem
functioning, and economic sustainability. The study of invasions requires specific methodological approaches to gain rapid
insight into the spatial and temporal dynamics of the system. We studied a recent invasion by the exotic herb Hieracium pilosella L. (Mouse-ear hawkweed, Asteraceae) in the Argentinean portion of the northern part of Tierra del Fuego Island in Southern
Patagonia. To assess the extent of this invasion and the related ecological and land use factors, we performed an extensive
field sampling at a regional scale and used a spatial pattern approach using geostatistical techniques to build a map of the
invasion. Our results showed that the invading species is widely distributed across the entire Fuegian steppe, in general
with low cover (<2%) and a particular spatial structure having hot spots (10–70%) related to specific land uses. We found
regional-latitudinal and also local-community level variability in the frequency, cover and spatial distribution of the invasive
species, each of them associated with particular ecological factors, and no association with regular domestic animal grazing.
Region-wide invasion may be accounted for by wind dispersal of the seeds and the latitudinal variability of the precipitation;
while local-variability was associated with the susceptibility of different plant communities to invasion. Hot spots were
found to be related to massive soil disturbances (e.g., road building, shrub removal, physical impacts of long-term and intense
domestic animal use, etc.), probably due to the presence of bare soil favoring colonization by the invading species. Scrublands,
lawns and grasslands were more invaded than wetlands, heathlands or salt grasslands. Since the invasion is apparently at initial
stages due to the low cover values found, we believe that information about the extent and environmental or management factors
involved in this exotic plant’s expansion should be used to design appropriate control and mitigation tools. In this sense,
our results point to management actions focused on reducing the presence of hot spots of invasion by means of quick revegetation
of bare soil after a massive disturbance event takes place. 相似文献

The specificity of peptidyl chloromethyl ketones has been used to label proteases in complex biological systems by incorporating tyrosine into the structure for eventual radioiodination. Contrary to results with iodination of proteins, a mild reagent, that is, one which iodinates at neutrality, was unsuitable, giving complex mixtures with poor reproducibility, apparently because of side reactions at the chloromethyl ketone group. On the other hand, iodine monochloride in acetic acid provided clean products. In the cases examined where a tyrosine residue was not appropriate for the specificity of the target protease, this residue was located well displaced from the primary specificity site. The resultant diiodotyrosine-containing derivatives were generally highly active as protease inhibitors. The p-aminobenzoyl group was used as an alternative to tyrosine as an iodinatable component. 相似文献
