The dispersal pattern of microorganisms has been a debated topic in the last decade, because for microscopic organisms with dispersing stages, biogeographic barriers may not exist and geographic distances may have little effect on the probability of their occurrence. In this study we tested whether a group of microscopic animals, the bdelloid rotifers, sampled in a tropical country, could indeed be cosmopolitan, strictly tropical, and/or have endemic species. We sampled freshwater habitats throughout Thailand in dry and rainy seasons between 2014 and 2016, focusing on permanent water bodies, where the prevalence of species producing resting stages acting as propagules for dispersal should be minimal and thus dispersal limitation should exist, with several species being narrowly distributed. We used traditional morphological identification, geometric morphometric analyses on the shape and size of the jaws, and DNA taxonomy to identify species and to quantify the morphological and genetic distances of Thai samples in comparison with data on bdelloids from other areas in the world. We found a total of 12 species of planktonic and periphytic bdelloid rotifers, and all approaches we used, qualitative and quantitative, confirmed a group of nine potentially cosmopolitan species already known from other continents, one tropical oriental species, and two new species with a potentially narrow geographic distribution, one of which we here formally describe. We thus support the idea that for microscopic animals the moderate endemicity hypothesis suggested for protists may also hold true: most of the species we found indeed have a wide geographic distribution, but other species with narrower distributions do exist.

Blastocystis is a unicellular protist most commonly detected in humans and a variety of animals. The predominant mode of its transmission is the fecal–oral route, but its zoonotic potential is not completely understood. The objective of this study was to determine the presence and genetic diversity of Blastocystis on pig farms in Nakhon Pathom Province, Central Thailand. A total of 154 human and 90 pig stool samples were collected and analyzed. Nested PCR detected Blastocystis in 35.55% of the pig samples and 6.49% of the human samples. Subtyping based on regions of the small-subunit ribosomal RNA (SSU rRNA) gene identified three Blastocystis subtypes in pigs and humans: ST1, ST3, and ST5. Blastocystis ST5 was the predominant subtype, followed by ST1 and then ST3. All the sequences from the Blastocystis-positive samples from both pigs and humans were closely related. This study reveals a possibility of low host specificity of Blastocystis STs (ST1, ST3 and ST5) on pig farms in Thailand. We tentatively suggest that close contact with or exposure to pig stools may be a significant source of Blastocystis detected in pig handlers. Further studies are required to confirm the zoonotic transmission of this organism in Thailand, because pigs may play an important role in the transmission of Blastocystis.  相似文献

Background

Rapid easy-to-use HIV tests offer opportunities to increase HIV testing among populations at risk of infection. We used the OraQuick Rapid HIV-1/2 antibody test (OraQuick) in the Bangkok Tenofovir Study, an HIV pre-exposure prophylaxis trial among people who inject drugs.

Methods

The Bangkok Tenofovir Study was a randomized, double-blind, placebo-controlled trial. We tested participants’ oral fluid for HIV using OraQuick monthly and blood using a nucleic-acid amplification test (NAAT) every 3 months. We used Kaplan-Meier methods to estimate the duration from a positive HIV NAAT until the mid-point between the last non-reactive and first reactive oral fluid test and proportional hazards to examine factors associated with the time until the test was reactive.

Results

We screened 3678 people for HIV using OraQuick. Among 447 with reactive results, 436 (97.5%) were confirmed HIV-infected, 10 (2.2%) HIV-uninfected, and one (0.2%) had indeterminate results. Two participants with non-reactive OraQuick results were, in fact, HIV-infected at screening yielding 99.5% sensitivity, 99.7% specificity, a 97.8% positive predictive value, and a 99.9% negative predictive value. Participants receiving tenofovir took longer to develop a reactive OraQuick (191.8 days) than participants receiving placebo (16.8 days) (p = 0.02) and participants infected with HIV CRF01_AE developed a reactive OraQuick earlier than participants infected with other subtypes (p = 0.04).

Discussion

The oral fluid HIV test performed well at screening, suggesting it can be used when rapid results and non-invasive tools are preferred. However, participants receiving tenofovir took longer to develop a reactive oral fluid test result than those receiving placebo. Thus, among people using pre-exposure prophylaxis, a blood-based HIV test may be an appropriate choice.
