Finite element method (FEM) simulations have been carried out on free-standing and finite dielectric substrate-supported eccentric (i) silica core-gold nanoshell dimers and (ii) gold core-silica nanoshell dimers for understanding their near- and far-field plasmonic properties. In the case of eccentric silica core-gold nanoshell dimers, multiple peaks are observed in the near- and far-field spectra due to the plasmon hybridization. The number of peaks is found to be sensitive to the core offset parameters of the nanoshells forming nanodimer. The wavelength locations of the peaks due to the constructive coupling of the lower order modes found relatively more sensitive to the dielectric substrate. The number of peaks in the near- and far-field spectra found the same presence and absence of the dielectric substrate. The values of full width at half maximum (FWHM) of the peaks observed in the near-field spectra are found larger as compared to those observed in the far-field spectra. In contrast, in the case of eccentric gold core-silica nanoshell dimers, multiple peaks have not been observed. The FWHM of the observed peak is found sensitive to the core offset parameters of the nanoshells, and the number of peaks in the near field- and far-field spectra found not same in the presence and absence of the dielectric substrate. Moreover, the differences in near- and far-field spectra of plasmonically coupled (i) concentric nanoshells, (ii) eccentric nanoshells, and (iii) concentric and eccentric nanoshells also investigated numerically.

Exposure of intact cells of Spirulina to high temperature (HT) stress (40–60 °C) caused decrease in absorption spectrum and fluorescence emission spectrum. Low temperature emission spectra were altered at phycocyanin (PC) level. Preterm birth is a major contributor to neonatal mortality and morbidity. Infection results in elevation of inflammation‐related cytokines followed by infiltration of immune cells into gestational tissue. CXCL12 levels are elevated in preterm birth indicating it may have a role in preterm labour (PTL); however, the pathophysiological correlations between CXCL12/CXCR4 signalling and premature labour are poorly understood. In this study, PTL was induced using lipopolysaccharide (LPS) in a murine model. LPS induced CXCL12 RNA and protein levels significantly and specifically in myometrium compared with controls (3‐fold and 3.5‐fold respectively). Highest levels were found just before the start of labour. LPS also enhanced the infiltration of neutrophils, macrophages and T cells, and induced macrophage M1 polarization. In vitro studies showed that condition medium from LPS‐treated primary smooth muscle cells (SMC) induced macrophage migration, M1 polarization and upregulated inflammation‐related cytokines such as interleukin (IL)‐1, IL‐6 and tumor necrosis factor alpha (TNF‐α). AMD3100 treatment in pregnant mice led to a significant decrease in the rate of PTL (70%), prolonged pregnancy duration and suppressed macrophage infiltration into gestation tissue by 2.5‐fold. Further, in‐vitro treatment of SMC by AMD3100 suppressed the macrophage migration, decreased polarization and downregulated IL‐1, IL‐6 and TNF‐α expression. LPS treatment in pregnant mice induced PTL by increasing myometrial CXCL12, which recruits immune cells that in turn produce inflammation‐related cytokines. These effects stimulated by LPS were completely reversed by AMD3100 through blocking of CXCL12/CXCR4 signalling. Thus, the CXCL12/CXCR4 axis presents an excellent target for preventing infection and inflammation‐related PTL.  相似文献

Hyaluronidase (Hyaluronate lyase, E.C. 3.2.1.35) has been isolated from Heterometrus fulvipes scorpion venom by a combination of gel filtration on Sephadex G-75 and ion exchange chromatography on DEAE-cellulose. The enzyme preparation showed a single band on polyacrylamide gel electrophoresis and a molecular weight of 82,000. The final preparation was purified 27-fold. The optimum pH for enzyme activity was 4.0. No loss of activity was observed up to 30 degrees C and showed a sharp decrease in activity at 50 degrees C. Abstract

Speciation of binary complexes of Co(II), Ni(II), and Cu(II) with L-phenylalanine (Phe) in the presence of water–anionic surfactant mixtures in the concentration range of 0.0–2.5% w/v SLS has been studied pH-metrically at a temperature of 303 K and at an ionic strength of 0.16 mol L?1. The selection of best fit chemical models is based on statistical parameters and residual analysis. The predominant species detected were ML, ML2, and ML2H2 for Co(II), Ni(II), and Cu(II). The trend in the variation of stability constants with the mole fraction of SLS was explained on the basis of electrostatic and non-electrostatic forces. Distribution of the species with pH at different compositions of SLS–water media was also presented.  相似文献

The calcium-sensing receptor (CaR) is a G-protein-coupled receptor that signals in response to extracellular calcium and regulates parathyroid hormone secretion. The CaR is also expressed on normal mammary epithelial cells (MMECs), where it has been shown to inhibit secretion of parathyroid hormone-related protein (PTHrP) and participate in the regulation of calcium and bone metabolism during lactation. In contrast to normal breast cells, the CaR has been reported to stimulate PTHrP production by breast cancer cells. In this study, we confirmed that the CaR inhibits PTHrP production by MMECs but stimulates PTHrP production by Comma-D cells (immortalized murine mammary cells) and MCF-7 human breast cancer cells. We found that changes in intracellular cAMP, but not phospholipase C or MAPK signaling, correlated with the opposing effects of the CaR on PTHrP production. Pharmacologic stimulation of cAMP accumulation increased PTHrP production by normal and transformed breast cells. Inhibition of protein kinase A activity mimicked the effects of CaR activation on inhibiting PTHrP secretion by MMECs and blocked the effects of the CaR on stimulating PTHrP production in Comma-D and MCF-7 cells. We found that the CaR coupled to Galpha(i) in MMECs but coupled to Galpha(s) in Comma-D and MCF-7 cells. Thus, the opposing effects of the CaR on PTHrP production are because of alternate G-protein coupling of the receptor in normal versus transformed breast cells. Because PTHrP contributes to hypercalcemia and bone metastases, switching of G-protein usage by the CaR may contribute to the pathogenesis of breast cancer.  相似文献

BRAF inhibitors have revolutionized treatment of mutant BRAF metastatic melanomas. However, resistance develops rapidly following BRAF inhibitor treatment. We have found that BRAF‐mutant melanoma cell lines are more sensitive than wild‐type BRAF cells to the small molecule tyrosine kinase inhibitor dovitinib. Sensitivity is associated with inhibition of a series of known dovitinib targets. Dovitinib in combination with several agents inhibits growth more effectively than either agent alone. These combinations inhibit BRAF‐mutant melanoma and colorectal carcinoma cell lines, including cell lines with intrinsic or selected BRAF inhibitor resistance. Hence, combinations of dovitinib with second agents are potentially effective therapies for BRAF‐mutant melanomas, regardless of their sensitivity to BRAF inhibitors.  相似文献
