The rates of displacement of dimethyl sulfoxide from the cation [Pt(phen) (CH3) (Me2SO)]+ by a series of uncharged and negatively charged nucleophiles have been measured in a methanol/water (19:1 vol./vol.) mixture. The starting complex and the reaction products were characterized either as solids or in solution by their IR and 1H NMR spectra. The substitution reactions take place by way of a direct bimolecular attack of the ligand on the substrate. The sequence of reactivity observed is as expected on the basis of a nucleophilicity scale relevant for + 1 charged substrates ([Pt(en) (NH3)Cl]+ used as standard). The difference of reactivity between the first (t-BuNH2) and the last (SeCN) members of the series spans five orders of magnitude. The value measured for the nucleophilic discrimination (1.55) is the highest found so far for cationic substrates. This is a result of the easy transfer of some of the electron density brought in by the incoming ligand into the ancillary ligands. When the reaction is carried out in a series of protic and dipolar aprotic solvents, using chloride ion as nucleophile, the rate of formation of [Pt (phen) (CH3)Cl] is dominated by the extent of solvation of Cl, as measured by its values of the Gibbs molar energy of transfer ΔtG0. Conductivity measurements at 25°C in dichloromethane were fitted to the Fuoss equation and the values of the dissociation constants Kd for the ion pairs were calculated as follows: 2.27 × 10−5 M for Bu4NCl, 2.75 × 10−5 M for Bu4NSCN and 17.05 × 10−5 M for [Pt(phen) (CH3) (Me2SO)]PF6. The pseudo-first-order rate constants kobs for the reactions with Bu4NCl, Bu4NBr, Bu4NSCN and Bu4NI showed a curvilinear dependence on the concentration of the salt which levels off very soon (at concentrations higher than 0.005 M the kinetics are zero order in [Bu4NX]). On addition of the inert electrolyte Bu4NPF6 the rates slow down and the kinetics follow the rate law kobs = kKip[Bu4NX]/[Bu4NPF6] + Kip[Bu4NX]). These findings fit well with a reaction scheme which involves a pre-equilibrium Kip between ion pairs, followed by unimolecular substitution within the contact ion pair [Pt(phen) (CH3) (Me2SO)X]ip. Values of the equilibrium constants Kip for ion-pair exchange and of the internal substitution rates k were derived. The latter showed that the discrimination in reactivity between Cl, Br, SCN and I is greatly reduced with respect to aqueous solutions. The reason behind this may be desolvation of the ions coupled to the fact that a contact ion pair is already at a certain distance along the reaction coordinate in the direction of the transition state. Applications of the special salt effect and of ion pairing to synthesis are discussed.  相似文献

Abstract

Cobalt, nickel and chromium effects on germination of Alyssum, during after-ripening and aging. Cobalt (2.5–20mM), nickel (2.5–25mM) and chromium (0.2-1mM) effects have been studied in seeds of three species of Alyssum, of which two, A. bertolonii and A. argenteum, endemics to serpentine soils and one, A. nebrodense, closely related to the others but endemic to the Madonie (Sicily) where it grows on limestone.

The results, expressed as percentage of germinated seeds (p), Kotowski coefficient of velocity (v) and germination average ratio (), have been subjected to statistical analysis.

The effect per mM is generally inhibitory, not significant for cobalt, weak but significant for nickel, more marked and significant for chromium, linearly correlated to concentration. The inhibition reaches its highest level when seeds are ripe, being significantly related to the control germination capacity ().

The values of the specific inhibition (effect/mM modditppτ) for all the three species are higher for chromium, which is more toxic than nickel, and increase from A. nebrodense to A. argenteum, showing that of the two serpentine endemics A. bertolonii is more tolerant both to nickel and chromium.  相似文献

Background

Adjuvant chemotherapy (AC) in Stage II Colon Cancer (CC) is still under debate. Choice should be based on patients and disease characteristics. According to guidelines AC should be considered in high-risk T3N0 patients. No data are available for better option in low-risk patients. The aim of the study is to retrospectively evaluate relapse-free survival (RFS) and disease-free survival (DFS) according to treatment received in T3N0 CC.

Methods

RFS and DFS are evaluated with Kaplan-Meier method. Multivariate Cox proportional hazard model was developed using stepwise regression, enter limit and remove limit were p = 0.10 and p = 0.15, respectively.

Results

834 patients with T3N0 CC were recruited. Median age was 69 (29–93), M/F 463/371, 335 low-risk patients (40.2%), 387 high-risk (46.4%), 112 unknown (13.4%); 127 (15.2%) patients showed symptoms at diagnosis. Median sampled lymph nodes were 15 (1–76); 353 (42.3%) patients were treated with AC. Median follow up was 5 years (range 3–24). The 5-years RFS was 78.4% and the 5-years DFS was 76.7%. At multivariate analysis symptoms, lymph nodes, and adjuvant chemotherapy were prognostic factors for RFS. AC is prognostic factor for all endpoints.In low-risk group 5-years RFS was 87.3% in treated patients and 74.7% in non-treated patients (p 0.03); in high-risk group was respectively 82.7% and 71.4% (p 0.005).

Conclusions

Data confirmed the role of known prognostic factors and suggest the relevance of adjuvant chemotherapy also in low-risk stage II T3N0 CC patients. However, the highest risk in low-risk subgroup should be identified to be submitted to AC.  相似文献

In multi‐resolution simulations, different system components are simultaneously modeled at different levels of resolution, these being smoothly coupled together. In the case of enzyme systems, computationally expensive atomistic detail is needed in the active site to capture the chemistry of ligand binding. Global properties of the rest of the protein also play an essential role, determining the structure and fluctuations of the binding site; however, these can be modeled on a coarser level. Similarly, in the most computationally efficient scheme only the solvent hydrating the active site requires atomistic detail. We present a methodology to couple atomistic and coarse‐grained protein models, while solvating the atomistic part of the protein in atomistic water. This allows a free choice of which protein and solvent degrees of freedom to include atomistically. This multi‐resolution methodology can successfully model stable ligand binding, and we further confirm its validity by exploring the reproduction of system properties relevant to enzymatic function. In addition to a computational speedup, such an approach can allow the identification of the essential degrees of freedom playing a role in a given process, potentially yielding new insights into biomolecular function. Cardiac contraction and relaxation are regulated by conformational transitions of protein complexes that are responsible for calcium trafficking through cell membranes. Central to the muscle relaxation phase is a dynamic membrane protein complex formed by Ca2+-ATPase (SERCA) and phospholamban (PLN), which in humans is responsible for approximately 70% of the calcium re-uptake in the sarcoplasmic reticulum. Dysfunction in this regulatory mechanism causes severe pathophysiologies. In this report, we used a combination of nuclear magnetic resonance, electron paramagnetic resonance, and coupled enzyme assays to investigate how single mutations at position 21 of PLN affects its structural dynamics and, in turn, its interaction with SERCA. We found that it is possible to control the activity of SERCA by tuning PLN structural dynamics. Both increased rigidity and mobility of the PLN backbone cause a reduction of SERCA inhibition, affecting calcium transport. Although the more rigid, loss-of-function (LOF) mutants have lower binding affinities for SERCA, the more dynamic LOF mutants have binding affinities similar to that of PLN. Here, we demonstrate that it is possible to harness this knowledge to design new LOF mutants with activity similar to S16E (a mutant already used in gene therapy) for possible application in recombinant gene therapy. As proof of concept, we show a new mutant of PLN, P21G, with improved LOF characteristics in vitro.  相似文献

The twilight or mesophotic zone is amongst the less explored marine regions. In coastal areas, investigations and manipulative experiments on benthic biodiversity and ecosystem functioning at depths up to >50 m have been recently made possible by the progress of SCUBA techniques. In this study, we tested the effects of the presence of a gorgonian forest characterised by a large and dense population of the gold coral Savalia savaglia (Bertoloni 1819) on the benthic biodiversity (nematode species richness, and meiofauna community structure and richness of taxa), trophic guilds state (molluscs) and ecosystem functioning in the surrounding sediments. The S. savaglia colonies create elevated and complex tertiary structures. Our results indicate that the presence of these colonies was associated with a significantly increased deposition of bioavailable substrates and enhanced biodiversity, when compared with soft bottoms at the same depth but without gold corals. The higher biodiversity and altered trophic conditions resulted in higher rates of ecosystem functioning (e.g., higher benthic biomasses). These results suggest that S. savaglia should be particularly protected not only for its specific rarity, endemism and vulnerability but also because it has a prominent role in sustaining high levels of biodiversity and ecosystem functioning in the surrounding benthos of the twilight zone.  相似文献

Two symbolic indexes, the percentage of sequences characterized by three heart periods with no significant variations (0V%) and that with two significant unlike variations (2UV%), have been found to reflect changes in sympathetic and vagal modulations, respectively. We tested the hypothesis that symbolic indexes may track the gradual shift of the cardiac autonomic modulation during an incremental head-up tilt test. Symbolic analysis was carried out over heart period variability series (250 cardiac beats) derived from ECG recordings during a graded head-up tilt test (0, 15, 30, 45, 60, 75, and 90 degrees ) in 17 healthy subjects. The percentage of subjects showing a significant linear correlation (Spearman rank-order correlation) with tilt angles was utilized to evaluate the performance of symbolic analysis. Spectral analysis was carried out for comparison over the same series. 0V% progressively increased with tilt angles, whereas 2UV% gradually decreased. The decline of 2UV% was greater than the increase of 0V% at low tilt angles. Linear correlation with tilt angles was exhibited in a greater percentage of subjects for 0V% and 2UV% than for any spectral index. Our findings suggest that symbolic analysis performed better than spectral analysis and, thus, is a suitable methodology for assessment of the subtle changes of cardiac autonomic modulation induced by a graded head-up tilt test. Moreover, symbolic analysis indicates that the changes of cardiac sympathetic and vagal modulations observed during this protocol were reciprocal but characterized by different absolute magnitudes.  相似文献
