The influence of nonionic surfactants on the activity of a novel thermostable lipase from Bacillus stearothermophilus MC7 was investigated with a view to its potential for synthesis of structured lipids. A large number of modifiers within a broad concentration range were applied. The activity of the enzyme was measured at a relatively high reaction temperature. Highest degree of activation was observed when PEG6000 was applied (up to 2.3-fold increase). Modification essentially changed the performance of the lyophilised preparations—they keep up to 80% of the activity of the native enzyme in the presence of a detergent against 30% in its absence. The effect of sorbitan esters (spans) and polyoxyethylene derivatives of sorbitan esters (tweens) on lipase MC7 was estimated, their HLB value varying within the interval 2.1–16.7. Tweens were strong inhibitors at higher concentrations. For all spans, excepting span 60, an increase of enzyme activity with concentration was observed. All studied additives slow down the process of thermal denaturation. Lipase preparations preserve more than 60% of their activity after 30-min incubation at 75 °C in the presence of tween 60 or PEG4000.  相似文献

Investigations on the microbial life in several coastal solar salterns have revealed the presence of novel organisms and synthesis of unusual molecules active in extreme conditions which might be useful in different biotechnological industries. Biodiversity of heterotrophic aerobic bacteria isolated from two salterns, Pomorie salterns and Burgas salterns located at Burgas Bay, Black Sea coast, Bulgaria, as well as ability of the isolates to synthesize biotechnologically valuable compounds were investigated. The results revealed high taxonomic and metabolic bacterial diversity—we isolated 20 morphologically different moderately halophilic and two halotolerant strains affiliated with 11 species from eight genera referred to the phyla Proteobacteria, Firmicutes, and Actinobacteria. Gram-negative bacteria belonged to the genera Halomonas, Chromohalobacter, Salinivibrio, Cobetia, and Nesiotobacter, and gram-positive strains were representatives of the genera Virgibacillus, Salinicoccus, and Brevibacterium. All isolates were found to be alkalitolerant, and 41% of them were psychrotolerant. The strains degraded nine of the tested 18 substrates; polygalacturonase, catalase, phytase, and lipase producers were predominant. This is the first reported detection of xanthan lyase, gellan lyase, arabinase, and phytase activities in halophilic bacteria. Nine of the strains belonging to five different genera were found to produce exopolysaccharides (EPS). The highest level of EPS was observed in Chromohalobacter canadensis strain 28. More than a half of the strains displayed antimicrobial activity against one to five test bacteria and yeasts. The present study is the first report on halophilic bacteria isolated from salterns at the Black Sea coast indicating that the investigated area is an untapped resource of halophilic bacteria with biotechnological potential.  相似文献

Ccm1p is a nuclear-encoded PPR (pentatricopeptide repeat) protein that localizes into mitochondria of Saccharomyces cerevisiae. It was first defined as an essential factor to remove the bI4 [COB (cytochrome b) fourth intron)] and aI4 [COX1 (cytochrome c oxidase subunit 1) fourth intron] of pre-mRNAs, along with bI4 maturase, a protein encoded by part of bI4 and preceding exons that removes the intronic RNA sequence that codes for it. Later on, Ccm1p was described as key to maintain the steady-state levels of the mitoribosome small subunit RNA (15S rRNA). bI4 maturase is produced inside the mitochondria and therefore its activity depends on the functionality of mitochondrial translation. This report addresses the dilemma of whether Ccm1p supports bI4 maturase activity by keeping steady-state levels of 15S rRNA or separately and directly supports bI4 maturase activity per se. Experiments involving loss of Ccm1p, SMDC (sudden mitochondrial deprivation of Ccm1p) and mutations in one of the PPR (pentatricopeptide repeat) motifs revealed that the failure of bI4 maturase activity in CCM1 deletion mutants was not due to a malfunction of the translational machinery. Both functions were found to be independent, defining Ccm1p as a moonlighting protein. bI4 maturase activity was significantly more dependent on Ccm1p levels than the maintenance of 15S rRNA. The novel strategy of SMDC described here allowed the study of immediate short-term effects, before the mutant phenotype was definitively established. This approach can be also applied for further studies on 15S rRNA stability and mitoribosome assembly.  相似文献

Extracellular bacterial lipases are promising biocatalysts for industry, because they are stable and active enzymes from easily available sources. A lipase from Bacillus stearothermophilus MC7 was immobilized on four polymer carriers by physical adsorption: chitosan, DEAE-cellulose, polypropylene, and polyurethane. The four biocatalysts differ in their hydrolytic activity against long-chain and short-chain triglycerides. Lipase MC7 immobilized on polypropylene (PP-MC7) stands out with its high activity against tributirin. According to the preliminary data, all four preparations were suitable for application in the test reaction of acidolysis of triolein with caprylic acid. The highest degree of conversion of the initial triolein was achieved in the presence of PP-MC7 preparation—50%. But variation of the reaction conditions did not lead to synthesis of the target di-substituted product (dicapryloyl-oleoylglycerol, COC). Reaction proceeds as a selective mono-substitution in the glycerol backbone.  相似文献

Unusual composition of an exopolymer (EP) from an obligate halophilic bacterium Chromohalobacter canadensis 28 has triggered an interest in development of an effective bioreactor process for its production. Its synthesis was investigated in 2‐L bioreactor at agitation speeds at interval 600‐1000 rpm, at a constant air flow rate of 0.5 vvm; aeration rates of 0.5, 1.0, and 1.5 vvm were tested at constant agitation rate of 900 rpm. EP production was affected by both, agitation and aeration. As a result twofold increase of EP yield was observed and additionally increased up to 3.08 mg/mL in a presence of surfactants. For effective scale‐up of bioreactors mass transfer parameters were estimated and lowest values of KLa obtained for the highest productivity fermentation was established. Emulsification activity of EP exceeded that of trade hydrocolloids xanthan, guar gum, and cellulose. A good synergism between EP and commercial cellulose proved its potential exploration as an enhancer of emulsifying properties of trade emulsions. A pronounced lipophilic effect of EP was established toward olive oil and liquid paraffin. Cultivation of human keratinocyte cells (HaCaT) with crude EP and purified γ‐polyglutamic acid (PGA) showed higher viability than control group.  相似文献
