The division of chloroplasts in a green alga, Pediastrum duplex,was studied by electron microscopy. Cells were treated for observationwith the freeze-substitution method. Fibrils, or fibrous belts,which we had observed previously at the dividing constrictionsof chloroplasts in Trebouxia potteri were not visible in Pediastrum,even though the method of preparation was the same for bothsets of samples. Microtubules (MTs) and the septum seem notto participate directly in the division of the single chloroplastin Pediastrum cells. Many thin fibrils, 7–20 nm in diameter,attached to, or protruding from, the surface of the dividingconstriction were seen. These fibres were less densely distributedat the constrictions of non-dividing chloroplasts. It is suggestedthat these fibrils are involved in the divison of chloroplastsin Pediastrum duplex. OUR previous studies on the X-linked testicular feminization (Tfm) mutation1 of the mouse2–4 showed that the so-called cytosol and nuclear 5αx-dihydrotestosterone (DHT) receptor protein5–7 might be a regulatory protein specified by the Tfm locus. The dual role of being a translational repressor in the cytoplasma and a mediator of hypertrophy in the nucleus was envisaged8. We found, however, another class of androgen-receptor in the polysome fraction of kidney proximal tubule cells which seems better qualified to be a translational regulator. Since a single gene locus specifies only one kind of polypeptide chain, we re-examined whether the cytosol and nuclear DHT-receptor protein underwent a true mutational change in Tfm/Y individuals.  相似文献

To test the association between temperate forest dynamics and glacial refugia for arboreal small mammals, we studied the phylogeography of the Japanese giant flying squirrel ( Petaurista leucogenys ) using complete mitochondrial cytochrome b gene sequences (1140 bp). This squirrel is endemic to three of Japan's main islands: Honshu, Shikoku, and Kyushu. We examined 58 specimens of P. leucogenys collected from 40 localities in Japan. Additionally, two individuals with unknown sampling localities were included in phylogenetic analyses. There were 54 haplotypes of P. leucogenys. We found five major phylogroups (Northern, Central, South-eastern, South-western, and Southern). These phylogroups may have originated from glacial refugia during the Late Pleistocene. After the last glaciation, the Northern phylogroup, widely distributed in eastern Japan, could have extensively expanded northward from its refugia. By contrast, in western Japan, population expansion was restricted to western Japan. All members of four phylogroups existed in western Japan during glaciations. The complicated phylogeographical pattern of P. leucogenys populations originating from western Japan may have resulted from the long history.  © 2009 The Linnean Society of London, Biological Journal of the Linnean Society , 2009, 98 , 47–60.  相似文献

An aggregateless mutant (HT41), isolated from D. discoideum NP14, was examined for the parameters involved in cell aggregation. HT41 cells were induced to express some of these parameters by cyclic AMP pulses. Spontaneous oscillations in light scattering were not observed in the suspension of the mutant cells. A pulse of cyclic AMP caused a monophasic decrease in light scattering, which was similar to the response of pre-aggregation cells of the wild-type. When cyclic AMP pulses were applied on a cell layer, HT41 cells aggregated without forming the streams. These results suggest that HT41 is a mutant defective in signal emission and relay.  相似文献

The cis‐unsaturated molecular species of phosphatidylglycerol (PG) in chloroplasts have been implicated in the chilling sensitivity of plants. Homozygous lines of transgenic tobacco (Nicotiana tabacum) that overexpressed the cDNA for glycerol‐3‐phosphate acyltransferase, a key enzyme in the determination of the extent of cis‐unsaturation of PG, were established from a chilling‐sensitive squash (Cucurbita moschata). In transgenic plants, the proportion of saturated plus trans‐monounsaturated molecular species of PG increased from 24 to 65%. However, this change did not affect the architecture of the chloroplasts. Chilling stress decreased the growth and biomass production of young seedlings of transgenic plants more severely than those of wild‐type plants, and this observation suggests that the changes in the proportion of cis‐unsaturated PG affected not only leaves but also developing plants. Chilling stress also damaged inflorescences. In particular, the abscission of flower buds and inflorescence meristems from transgenic plants occurred more frequently than that from wild‐type plants. Thus, it is likely that decreases in the proportion of cis‐unsaturated PG enhanced the sensitivity to chilling of reproductive organs.  相似文献

Tropical peatlands, which coexist with swamp forests, have accumulated vast amounts of carbon as soil organic matter. Since the 1970s, however, deforestation and drainage have progressed on an enormous scale. In addition, El Niño and Southern Oscillation (ENSO) drought and large‐scale fires, which grow larger under the drought condition, are accelerating peatland devastation. That devastation enhances decomposition of soil organic matter and increases the carbon release to the atmosphere as CO2. This phenomenon suggests that tropical peatlands have already become a large CO2 source, but related quantitative information is limited. Therefore, we evaluated the CO2 balance of a tropical peat swamp forest in Central Kalimantan, Indonesia, using 3 years of CO2 fluxes measured using the eddy covariance technique from 2002 through 2004. The forest was disturbed by drainage; consequently, groundwater level (GL) was reduced. The net ecosystem CO2 production (NEP) measurements showed seasonal variation, which was slightly positive or almost zero in the early dry season, and most‐negative late in the dry season or early the rainy season. This seasonality is attributable to the seasonal pattern of climate, tree phenology and fires. Slightly positive NEP resulted from smaller ecosystem respiration (RE) and larger gross primary production (GPP) under conditions of high photosynthetic photon flux density (PPFD) and large leaf area index (LAI). The most‐negative NEP resulted from smaller GPP and larger RE. The smaller GPP was related to high vapor pressure deficit (VPD), small LAI and low PPFD because of smoke from fires. The larger RE was related to low GL. Annual NEP values were estimated respectively as −602, −382 and −313 g C m−2 yr−1 for 2002, 2003 and 2004. These negative NEP values show that the tropical peat swamp forest, disturbed by drainage, functioned as a CO2 source. That source intensity was highest in 2002, an ENSO year, mainly because of low PPFD caused by dense smoke emitted from large fires.  相似文献

We investigated temporal and spatial variations in the zooplankton community structure in the Oyashio and Transition region of the subarctic western North Pacific from 1960 to 1999 using principal component analysis (PCA) and zooplankton samples from the historical Odate Collection. In particular, we examined the influence of Kuroshio and Oyashio decadal dynamics on geographical variations in the zooplankton community. The first principal component (PC1) closely represented the interannual variation in cold-water, large copepod species, while the second PC (PC2) represented the variation in warm-water, small copepod species. Using Rodionov's regime-shift method, we detected a significant increase in the PC score after 1976 and 1981 for PC1 and PC2, respectively. After the shift years, (1) warm-water species increased in the Transition zone, (2) the distribution center of the cold-water species shifted southward, and (3) copepod abundance and species diversity increased in the Transition zone as a result of (1) and (2). The timing of these shifts in the zooplankton community roughly coincided with the North Pacific climatic regime shift in 1976/1977. From the mid-1970s to the early 1980s, the southern boundary of the Oyashio shifted southward and increased geostrophic transport was observed in the Kuroshio, indicating spin up of the Kuroshio–Oyashio system. Change in atmospheric circulation during the 1976/1977 regime shift is thought to have caused the spin up of these currents, which subsequently affected the regional zooplankton community through advective processes.  相似文献

Sipwise and flowise presentation of taste stimuli were compared and better taste discrimination was noted for flowise presentation. Using a model system, discrimination between NaCl solutions and purified water, analysis with atomic absorption spectrophotometry indicated that the superiority of flowise presentation was due to reduction in stimulus variance. Flowise presentation was also superior for discrimination between commercially available natural spring waters.  相似文献
