Cells of the ciliate Tetrahymena pyriformis GL overproduce and accumulate massive quantities of the heme intermediate, protoporphyrin IX. Protoporphyrin is localized intracellularly in discrete membranous compartments. The amount of porphyrin stored in the cell changes dramatically as cells progress through the growth cycle. Porphyrin overproduction is stimulated by δ-aminolevulinic acid, but only during the mid-stationary phase. Overproduction of protoporphyrin IX apparently results from an increase, late in the growth cycle, of activities subsequent to δ-aminolevulinic acid synthetase. Feedback inhibition in the pathway by accumulated protoporphyrin IX does not occur. The presence of Co2+ completely inhibits accumulation of protoporphyrin IX in a manner reversed by δ-aminolevulinic acid. Sn4+ stimulates protoporphyrin IX accumulation in the culture.  相似文献

The capacity of adult newt (Triturus viridescens) spleen cellsto secrete antibody at 4 C allows simultaneous visualizationand quantification of non-secretory (S) and secretory(S+) rosette forming cells (RFC). Visualization of mammalianS+ RFC requires 37 C, a temperature at which S RFC appearto be fragile. RFC can be distinguished as S or S+ dueto whether one or more layers of erythrocytes adhere to thesurface of sensitized spleen cells. Different doses of horseerythrocytes (HRBC) affect newt S RFC and S+ RFC differentially.By varying the time between injections of different concentrationsof chicken erythrocytes (CRBC, the "carrier") and a constantdosage of CRBC-TNP (trinitrophenyl, the hapten) it is possibleto measure "helper" activity that correlates with the populationof S RFC and is both dose and time dependent. By varyingassay time after "helper" activity has been maximized, one candetermine the cytodynamics of anti-TNP antibody producing cell(APC) activity. For the first time these morphologically separableRFC can be related to their suspected physiologic behavior.A shift from S RFC to S+ RFC takes place during the immuneresponse. That similar dose-dependent response curves can beshown in adultRana pipiens suggests that the newt responsesrepresent a fundamental vertebrate pattern.  相似文献

We used a policlonal antiserum against GABA and demonstated GABA-immunoreactivity (GABA-IR) in several populations of amacrine cells in the inner nuclear layer (INL), and other cells in the inner plexiform layer (IPL) of the central and peripheral retina of the chameleon. Horizontal cells do not contain GABA-IR and the chameleon retina is therefore an exception among non-mammals. GABA-IR was not seen in cell bodies in the position of photoreceptor, bipolar and interplexiform cells suggesting that GABA is not involved in synaptic transmission in the outer plexiform layer of chameleon retina.  相似文献

Abstract. A survey was made of the phlebotomine sandfly fauna of La Guaira, a village with coffee plantations near Cali, Colombia, from which cases of American cutaneous leishmaniasis had been reported due to Leishmania (Viannia) panamensis and Le. ( V. ) braziliensis. Among six species of sandfly collected on human bait, Lutzomyia youngi was most important in terms of biting nuisance. Lu. columbiana, Lu. lichyi and Lu. scorzai as well as Lu. youngi adults occurred throughout the year. Sandfly man-biting activity occurred throughout the night and was highest within 2 h of sunset. Despite its abundance in nocturnal samples, Lu. youngi was rarely taken in diurnal resting site collections. In contrast, Lu. lichyi was collected on tree-trunks during the day in large numbers and was the only species biting in daylight. The implications of these and other findings for leishmaniasis control measures in La Guaira are considered.  相似文献

1. In October 2004, plankton samples were collected from six permanent lakes located between 4960 and 5440 m a.s.l. in the Mount Everest region (Nepal) to assess how spatial and local environmental factors affect natural bacterial community composition. Fingerprinting analysis of the bacterial 16S rRNA gene fragment was done by denaturing gradient gel electrophoresis (DGGE).
2. The number of DGGE bands (range: 12–23) was not correlated with lake area or remoteness, but there was a strong negative correlation with the ratio of catchment to lake area ( r  = −0.826, P  <   0.05), suggesting that hydraulic retention time affects the establishment of the bacterial community in these seepage lakes.
3. Most dominant sequences belonged to Betaproteobacteria except in two lakes where members of Bacteroidetes made the largest relative contribution. Up to 81% of the phylotypes had high similarity (>98 to 100%) in partial 16S rRNA gene sequence to those reported from other alpine lakes and glaciers around the world, suggesting the presence of 'cosmopolitan' bacteria.
4. An analysis based on dissimilarity matrices and the Mantel test revealed the existence of dissimilarities in bacterial community composition related to geographical distance over a small spatial scale (<6 km), but determined by local environmental constraints.
5. Our results suggest that several bacterial phylotypes are ubiquitous in the freshwater aquatic realm, but taxon sorting by local environmental constraints is important.  相似文献

Studies of sibling competition within brood hierarchies have rarely assessed simultaneously the effects of sex and rank in the brood hierarchy on traits other than offspring mortality and differential growth. We studied the expression of heat-shock proteins (Hsps) to assess the physiological stress response to different combinations of sex and position within competitive brood hierarchies in the black kite Milvus migrans (Bodd.), a sexually dimorphic raptor showing facultative siblicide. Senior males showed higher stress levels than did senior females and younger siblings of each sex as revealed by Hsp60 values. The analysis of Hsp70 levels indicated that nestlings from broods in which the senior chick was a male showed higher stress levels than did nestlings from broods in which the senior chick was a female. In addition, levels of Hsp60 were related negatively to nutritional condition expressed as levels of plasmatic albumin. This suggests that the sex of senior chicks may be key in determining their stress level and that of their siblings, which is probably associated with sibling competition by fighting within brood hierarchies. The comparatively higher stress levels of senior males (and their siblings) may be a consequence of their ability to exploit their potential advantage from being the head start while avoiding a possible competitive disadvantage from being the smaller sex, independent of environmental conditions determining the probability of brood reduction. Differential stress levels depending on sex and rank in the brood hierarchy may be a consequence of parental control of offspring behaviour through differential resource allocation (e.g. yolk androgens) or it may reflect adaptations of particular chicks (senior males) to enhance their competitive ability within brood hierarchies.  © 2006 The Linnean Society of London, Biological Journal of the Linnean Society , 2006, 88 , 383–390.  相似文献

SYNOPSIS. This report deals with observations which suggestnew insights and speculations with regard to the unique developmentalcycle of lymphoreticular neoplasia in adult Xenopus laevis andthe systemic controls which may be imposed upon it. Evidenceis presented which suggests that: (1) Organismally imposed necrosisof visceral tumor foci was effectively suppressed by subsequentimplantation of allo-tumor foci but not by normal tissue allo-grafts;(2) Recovery of the healthy state of neoplastic foci followingnecrosis appears to depend on the acquisition of new tumor cellsrather than on cloning by survivors of necrosis in primary foci;(3) Diseased organisms eventually develop controls which suppressthe growth of implanted allotumor foci (tumor growth suppressionwas also demonstrated with splenic but not liver fragments fromatoad which survived a prior challenge with a homogenate of neoplastictissue); (4) Animals with this lympho-proliferative disorderappear to have an immunologic deficiency for cell-mediated responses;(5) Normal young adult Xenopus hosts generate a chronic ratherthan acute allograft response. Structural variations between edge and interior areas within forest fragments may bring about differences in food availability that may influence the selective behaviour of predators and prey population dynamics. The purpose of this paper was to assess patterns of artificial prey selection by wild birds (House Sparrow Passer domesticus and Rock Dove Columba livid) between edge and interior areas of woodland, taking into account differences in prey frequency (10% of one food type and 90% of the other) and density (30 baits/m-2 and 50 baits/m-2). Experiments were conducted at 24 plots in 3 forest fragments in the city of Madrid, Spain. Selectivity did not vary among parks or between densities. However, selectivity did vary with the frequency and location of baits, showing an anti-apostatic trend (baits were preferred at low rather than at high frequencies) that was more pronounced at interiors than at edges. Two possible factors that may account for stronger anti-apostatic selection at edges are the higher densities of predators and pedestrians found there. However, there are many other possible explanations, and no specific conclusion can be supported with the current data. The results of this study also point out that site heterogeneity should be taken into account in the experimental design of future studies on frequency-dependent food selection by wild birds, particularly in fragmented landscapes.  相似文献
