We challenge the concept of idiopathic parkinsonism (IP) as inevitably progressive neurodegeneration, proposing a natural history of sequential microbial insults with predisposing host response. Proof-of-principle that infection can contribute to IP was provided by case studies and a placebo-controlled efficacy study of Helicobacter eradication. "Malignant" IP appears converted to "benign", but marked deterioration accompanies failure. Similar benefit on brady/hypokinesia from eradicating "low-density" infection favors autoimmunity. Although a minority of UK probands are urea breath test positive for Helicobacter , the predicted probability of having the parkinsonian label depends on the serum H. pylori antibody profile, with clinically relevant gradients between this "discriminant index" and disease burden and progression. In IP, H. pylori antibodies discriminate for persistently abnormal bowel function, and specific abnormal duodenal enterocyte mitochondrial morphology is described in relation to H. pylori infection. Slow intestinal transit manifests as constipation from the prodrome. Diarrhea may flag secondary small-intestinal bacterial overgrowth. This, coupled with genetically determined intense inflammatory response, might explain evolution from brady/hypokinetic to rigidity-predominant parkinsonism.  相似文献

Summary This paper reviews the biogeography, geographic variation, and habitat characteristics of the Amazilia Hummingbird (Amazilia amazilia). Based on morphological features, habitat preferences, and bioacoustics, I propose that the currently recognized subspeciesA. a. alticola should be regarded as a species distinct fromA. amazilia. Synapomorphic characters found in both taxa indicate membership of the same superspecies, together with the Colombian speciesA. castaneiventris. Considering the distributional focus ofA. alticola in the Andes of southern Ecuador, the English species name Loja Hummingbird is suggested.

Zusammenfassung Die vorliegende Studie stellt die Ergebnisse von Untersuchungen zur Biogeographie, geographischen Variation, Habitatwahl und Taxonomie der Lesson-Amazilie (Amazilia amazilia) vor. Das Taxon umfaßt gegenwärtig fünf Unterarten, die von W-Ecuador bis SW-Peru verbreitet sind und überwiegend submontane Lebensräume besiedeln. Die Ventralfärbung unterliegt klinaler Variation; weiße Gefiederanteile nehmen von nördlichen zu südlichen Populationen ab, rotbraune Partien dagegen zu. Eigenständige morphologisch-morphometrische Kennzeichen sowie Unterschiede in den Habitatpräferenzen und in der Gesangsstruktur rechtfertigen es, die bisherige UnterartA. a. alticola als Paraspezies (A. alticola, Loja-Amazilie) anzuerkennen. Der Vulgärname weist auf den rezenten Verbreitungsschwerpunkt in den südecuadorianischen Anden hin. Aufgrund der engen Verwandtschaft bildenA. amazilia undA. alticola sowie die in Kolumbien endemische ArtA. castaneiventris eine Superspezies.

When synaptic plasma membrane fragments are incubated with ATP in the presence of Mg2+, phosphate is transferred, not only to protein-bound serine, but also to protein-bound histidine. The phosphorylation of protein-bound serine is stimulated by cyclic AMP and has a Km for ATP of about 0.12 mM, both in the presence and absence of cyclic AMP. By contrast, the phosphorylation of protein-bound histidine is unaffected by cyclic AMP and does not follow Michaelis-Menton kinetics since a non-linear double reciprocal plot is given when activity is measured at various ATP concentrations.  相似文献

During an 11-month study of the Black-headed Duck in eastern Argentina, observations were made on the marsh birds of Cape San Antonio, Province of Buenos Aires. These observations supplement the distributional and life history obtained by Gibson (1879–1920), Wetmore (1926) and others. The deep, fresh-water marshes characteristically are dominated by one of three plants: tules, cutgrass or cat-tails. There were surprisingly few marshes with mixtures of these plants and few marshes which showed a gradual transition from marsh to uplands. Marsh birds showed considerable adaptation in chronology of nesting and nest-site selection for the seasonal water cycle of wet springs and dry periods in the late summer and fall. Some marsh birds were involved in nesting over a longer period than is usual in Northern Hemisphere marshes. As seems characteristic of all marsh birds, cover-water edges were usual sites for nests. There were only a few species with restricted nest-site selection, and competition between closely related species was noted only among the three species of coots. A list of the marsh birds of the area is given, and comments are presented on the ecology and behaviour of selected species.  相似文献

Eosinophils, leukocytes involved in allergic, inflammatory and immunoregulatory responses, have a distinct capacity to rapidly secrete preformed granule-stored proteins through piecemeal degranulation (PMD), a secretion process based on vesicular transport of proteins from within granules for extracellular release. Eosinophil-specific granules contain cytokines and cationic proteins, such as major basic protein (MBP). We evaluated structural mechanisms responsible for mobilizing proteins from within eosinophil granules. Human eosinophils stimulated for 30-60 min with eotaxin, regulated on activation, normal, T-cell expressed and secreted (RANTES) or platelet activating factor exhibited ultrastructural features of PMD (e.g. losses of granule contents) and extensive vesiculotubular networks within emptying granules. Brefeldin A inhibited granule emptying and collapsed intragranular vesiculotubular networks. By immunonanogold ultrastructural labelings, CD63, a tetraspanin membrane protein, was localized within granules and on vesicles outside of granules, and mobilization of MBP into vesicles within and extending from granules was demonstrated. Electron tomography with three dimension reconstructions revealed granule internal membranes to constitute an elaborate tubular network able to sequester and relocate granule products upon stimulation. We provide new insights into PMD and identify eosinophil specific granules as organelles whose internal tubulovesicular networks are important for the capacity of eosinophils to secrete, by vesicular transport, their content of preformed and granule-stored cytokines and cationic proteins.  相似文献

The activity of the first enzyme of the shikimate pathway, 3-deoxy-d-arabino-heptulosonate 7-phosphate synthase, varies during the growth cycle of Solanum tuberosum L. cv superior cells in suspension culture. Maximum specific enzyme activity was observed midway through the linear phase of growth. When mid-log phase cells are exposed to glyphosate, the specific activity of the enzyme increases severalfold within 24 hours. The glyphosate-induced increase in enzyme activity is due to an increase in the amount of enzyme as determined by immunoblotting. Glyphosate (up to 2 millimolar) has no effect on the enzyme activity in vitro. Dehydroquinate synthase, the second enzyme of the shikimate pathway, is not induced by glyphosate.  相似文献

A novel series of CCR5 antagonists has been identified, utilizing leads from high-throughput screening which were further modified based on insights from competitor molecules. The evolution of sexual dimorphism depends in part on the additive genetic variance-covariance matrices within females, within males, and across the sexes. We investigated quantitative genetics of floral biomass allocation in females and hermaphrodites of gynodioecious Schiedea adamantis (Caryophyllaceae). The G-matrices within females (G(f)), within hermaphrodites (G(m)), and between sexes (B) were compared to those for the closely related S. salicaria, which exhibits a lower frequency of females and less-pronounced sexual dimorphism. Additive genetic variation was detected in all measured traits in S. adamantis, with narrow-sense heritability from 0.34-1.0. Female allocation and floral size traits covaried more tightly than did those traits with allocation to stamens. Between-sex genetic correlations were all <1, indicating sex-specific expression of genes. Common principal-components analysis detected differences between G(f) and G(m) , suggesting potential for further independent evolution of the sexes. The two species of Schiedea differed in G(m) and especially so in G(f) , with S. adamantis showing greater genetic variation in capsule mass and tighter genetic covariation between female allocation traits and flower size in females. Despite greater sexual dimorphism in S. adamantis, genetic correlations between the two sexes (standardized elements of B) were similar to correlations between sexes in S. salicaria.  相似文献
