Several cell lines of Nicotiana plumbaginifolia resistant tomethotrexate (MTX) were isolated upon gradual elevation of theMTX concentration in the growth medium. One of the MTX-resistantcell lines, NP-19, acquired resistance to 10 mmol m–3MTX, that is, at least 50-fold higher than the lethal dose forthe wild-type cell suspension. Its resistance was stably maintainedupon prolonged withdrawal of the drug. The acquisition of resistancewas accompanied by severe deterioration of regeneration potential.Regenerated shoots still manifested resistance to 50 mmol m–3.MTX-resistant colonies and tiny shoots were recovered followingasymmetric fusion between -irradiated NP-19 protoplasts andN. plumbaginifolia leaf mesophyll-derived protoplasts on a mediumcontaining 10 mmol m–3 MTX. The development of the shootsderived both from NP-19 calli and following somatic fusion wasarrested at the stage of 6–8 leaves. No difference wasfound in uptake of 3H-MTX between cell suspension of NP-19 andthe parental cell line. A 30-fold increase in binding of 3H-MTXto protein extract was found in cell line NP-19, suggestingdifferential capacity of MTX binding as a mechanism involvedin the MTX resistance of this cell line. Since differentiatedorgans seem more sensitive to MTX than undifferentiated tissues,this cell line is a promising source for a gene(s) conferringenhanced MTX-tolerance both in non-differentiated and differentiatedtissues. In Linum pubescens Banks & Solander and L. mucronatum Bertol. two floral morphs occur, differing in style length, stamen length, size of stigmatic papillae and, in L. mucronatum , exine sculpture of pollen grains. In both species artificial self- and intramorph cross-pollinations were incom, patible. Intermorph pollinations resulted in seed production. The inhibition of pollen-tube growth in incompatible pollinations occurs within the stigma. In L. pubescens distyly seems to be governed by a single pair of alleles. In natural populations the two morphs were found to be represented by approximately equal numbers of individuals. The effectiveness of the complementary arrangement of anthers and stigmas in promoting intermorph pollinations and in preventing pollen wastage in incompatible pollinations is discussed.  相似文献

Ethylene at 10 and 100 μl/litre stimulated germ-tube elongation of Botrytis cinerea spores incubated within normal and non-ripening nor tomato fruits, but had little influence on the total percent of germination. Values of germ-tube length within the mature-green normal fruits and the mature-green or mature nor fruits were similar to those recorded within the normal mature fruits when held in air. Exposure of the normal and the mutant fruits to 100 μl/litre ethylene immediately after inoculation with B. cinerea insignificantly increased lesion development, but resulted in increased sporulation. When tomato fruits were exposed to ethylene for 3 days before inoculation a marked stimulatory effect on rot development was exhibited on the mature-green normal fruits but not on the nor mutant fruits. The results indicate that exogenous ethylene may directly stimulate germ tube growth of B. cinerea in both normal and mutant fruit, but that it may affect subsequent fungal growth indirectly, via stimulation of the ripening process, only in preclimacteric normal tomato fruit.  相似文献

In formulating procreative principles, it makes sense to begin by thinking about whose interests ought to matter to us. Obviously, we care about those who exist. Less obviously, but still uncontroversially, we care about those who will exist. Ought we to care about those who might possibly, but will not actually, exist? Recently, unusual positions have been taken regarding merely possible people and the non‐identity problem. David Velleman argues that what might have happened to you – an existent person – often doesn't merit moral consideration since the alternative person one would have been had what might have happened actually happened is a merely possible person about whom one has no reason to care. He argues that his way of thinking can eliminate the non‐identity problem. Caspar Hare argues that merely possible people have interests and are morally relevant. He argues that we can solve the non‐identity problem by rejecting the view that merely possible people are morally irrelevant. Both Hare and Velleman argue that focusing on one's de dicto rather than on one's de re children can help us avoid the non‐identity problem. I analyze the role that merely possible, nonexistent hypothetical entities ought to play in our moral reasoning, especially with regard to procreation. I refute both Velleman's and Hare's views and demonstrate the difficulties we encounter when we try to apply their views to common non‐identity cases. I conclude with the common‐sense view regarding who matters, morally: only those who do, did, or will exist.  相似文献

Resistance of rin and nor tomato mutants to postharvest Rhizopus infection   总被引:1,自引:0,他引:1  
Fruits of the two non-ripening mutants of tomato, rin and especially nor, were markedly more resistant to Rhizopus stolonifer infection than the normal Rutgers fruit. Following artificial inoculations by contact with a diseased normal tomato covered with mycelium and sporangia, no infection of unwounded nor fruit occurred at its mature-green stage. At the mature stage the resistance of nor mutant fruit was manifested by a prolongation of the incubation period of the disease as well as by a markedly reduced incidence of rotted fruits. Chilling injury of fruit, prior to spore inoculation, was found to be a good means for indicating the relative resistance of the mutants as compared with the normal tomato. The relationship between the resistance of the mutant tomatoes to Rhizopus infection and their response to induced peel damage as a result of the contact or the chilling procedure, led to the assumption that fruit resistance is associated with the inability of the fungus to penetrate the periderm, rather than with fungal development within the fruit.  相似文献

Flowers of a normal tomato cultivar and of the two non-ripening mutants rin and nor, were sprayed with a Botrytis cinerea spore suspension. Stem-end infection developed in 84–100% of the harvested fruits of these sprayed flowers when held in high r.h. In both nor and rin fruits, rot development remained restricted to the stem-end area and fruit shoulders, whereas in the normal fruit decay spread rapidly from the stem-end over the entire fruit. Spraying the flowers with iprodione prior to spore inoculation resulted in decreased incidence of decay and suppressed hyphal growth and sporulation in all types of fruits. The results indicate that floral organs serve as a pathway for B. cinerea stem-end initiation in both the normal and the mutant fruit and suggest that this mode of penetration is not related to the marked resistance to infection attributed to the mutant fruits.  相似文献

A marked variation in tree size occurred in grapefruit trees budded in trifoliate orange rootstock and inoculated with a graft-transmissible dwarfing agent (GTD) from grapefruit. Etrog citron (Citrus medico) inoculated with budwood collected from two GTD sub-types displaying very mild (VM) and mild (M) dwarfing effects showed only mild leaf epinasty, whereas the severe (S) subtype induced severe epinasty and stunting typical of infection by the original GTD source 225-T. The characteristic symptoms of the three sub-types were persistent following three serial transfers to 'Etrog' indicators.
Extracts from 'Etrog' citron containing either the 225-T or its derivative S displayed a profile of five viroid bands when analysed by the sequential PAGE system for detection of circular RNAs. The profile obtained included the well characterised 371 nucleotide citrus exocortis viroid (CEV) and four additional viroids of approximately 330, 300, 295 and 275 nucleotides. Sub-types M and VM lacked the CEV band and each contained a complement of only three viroids of 330, 295, 275 and 300, 295, 275 nucleotides, respectively. These results indicate that a segregation of the viroid complex in grapefruit budwood was a major factor in the variation seen among trees inoculated from the GTD source 225-T.  相似文献

Silene vulgaris occurs in Israel as a stolonifcrous tetraploid which forms mostly sexually polymorphic populations. Fully hermaphrodite and fully male-sterile plants are linked by a range of intermediate forms which are gynomonoecious and bear hermaphrodite, male-sterile, and partially male-sterile flowers. Male-sterile flowers differ from hermaphrodite ones by having a narrower calyx, a shorter corolla, a thicker style, and a considerably larger stigmatic area with longer papillae which are more markedly bulbous basally. In individual anthers of hermaphrodite flowers, diameters of stained pollen grains showed a continuous variation from 36 to 80 μm with a modal value of 56. About 30% of pollen was found to be non-stainable. Reduction in male fertility occurs at different levels, from small or inviable pollen grains through abortion of single anthers, abortion of entire androecia, and through different degrees of gynomonoecy to totally male-sterile individuals. The proportion of stainable pollen to ovules in hermaphrodite flowers alone was found to be less than 100. From the data presented, less than a third of all ovules produced in the different kinds of flower develop into seeds. This may reflect an inadequacy of pollen, which the local population of S. vulgaris can tolerate because of its stoloniferous habit.  相似文献
